摘    要:以无水葡萄糖为标准品,对用荧光光谱法测定灵芝中多糖的含量进行了探讨,确定最佳实验条件为12.00mL浓硫酸,o.17mL 5%的苯酚溶液,室温下静置50min,荧光参数λex=478nm,λem=510nm.在此条件下对6种不同种灵芝的多糖含量进行了测定,多糖浓度与生成物的荧光强度呈良好的线性关系,线性方程为y=486...

Abstract:The content of polysaccharides in ganoderma lucidum was determined by fluorescence spectrometry with anhydrous glucose as standard.The optimal experimental conditions were as follows:concentrated sulfuric acid of 12.00mL,5% phenol solution of 0.17mL,reaction time of 50min at room temperature and fluorescence parameters at λex=478nm and λem=510nm.Polysaccharides of six different kinds of ganoderma lucidum were analyzed under the optimal experimental conditions.The concentration of polysaccharide had a good linear relationship with its fluorescence intensity.The linear equation was y=4868.6x-3.9048,r2 was 0.9990,and the detection limit was 0.008mg/mL.The results was compared with the method of determining polysaccharides of ganoderma lucidum in "The People's Republic of China Pharmacopoeia(2005 Edition)".It was showed that the results of fluorescence spectrometry were in agreement with the "Pharmacopoeia" method.The RSD of repeatability experiment was 0.41%,and average recovery was 98.81%.
