摘    要:以PEG为乳剂,建立了一种基于PEG凝集的液相压电免疫分析法,并用于人血清白蛋白(Alb)的测定。详细考察了PEG浓度,抗体稀释度、PEG分子量及反应温度等因素对频移的影响,采用终点法和初始速率法测得Alb线性范围分别为47.83-103.1mg/L和40.62-101.2mg/L。对7个血清样本的分析,其结果与双缩脲法基本一致。用一简单的方法合晶振再生后可获得重视的结果。

Abstract:A liquid piezoelectric immunoassay, which is based on the agglutination of antigen and anti- body polyethylene glycol (PEG) complex by an imunoreaction, has been developed for the determination of human serum albumin (Alb). The effect of experimental conditions such as PEG concentration, the antibody dilution ratio, the molecular weight of PEG and the reaction temperature on the frequency response were investigated. The linear ranges for Alb concentration determined by the end-point method and the initial rate method were 47. 83 - 103. 1 mg/L and 40. 62 - 101. 2 mg/l, respectively. Analytical results of seven clinical specimens obtained using the developed method were in satisfactory agreement with those given by the biuret method. With a simple regeneration method devised, the crystal can be used repeat- edly with acceptable reproducibility.
