Abstract: | Summary A sensitive, convenient and reliable free-zone capillary electrophoretic method has been developed for the determination of
adenylate nucleotides in cell lysates without pre-analytical sample clean up. Simultaneous quantification of AMP, ADP, and
ATP can be performed with precision (within run) of 1.7–2.7% and reproducibility (day to day) of 1.3–3.5%. Detection limits
for AMP, ADP, and ATP were 10 μm each and the linear range was 10–200 μm for each. The sample volume required was less than 10 nL. The proposed method greatly simplifies and accelerates determination
of cellular ‘adenylate energy charge’, the most reliable measure of cellular energy status. |
