耐热酵母菌株HU-TY-1的耐热机理初探

摘    要:以酿酒酵母耐热菌株HU-TY-1和亲株LK为材料,通过SDS-聚丙烯酰胺凝胶电泳等技术,对它们在热休克应答前后的耐热性及蛋白图谱进行了分析,发现两个菌株经温和高温(37 ℃)诱导后可大幅提高对致死温度(53 ℃)的耐受力,同时伴随着分子量为72 kD和84 kD的热击蛋白(HSP)的合成,其中耐热株HU-TY-1在常态时就有这两种热击蛋白的少量表达.由此表明:耐热酵母菌株HU-TY-1的高耐热性与HSP72和HSP84的组成性合成密切相关.

Abstract:The differences of protein synthesis and the survivors in high temperature between the thermotolerant S.cerevisiae stain HU-TY-1 and its primary stain LK have been compared by SDS-PAGE(sodium dodecanoyl sulphate-polyarcy lamide gel electrophoresis)and counting colonies. The results show that both the stains can synthesize HSP72 and HSP84 in heat shock response, and HU-TY-1 can synthesize these HSPs weakly at normal temperature. This suggests that the high thermotolerance of strain HU-TY-1 is relative tightly to the constitutive synthesis of HSP70 and HSP84.
