蚕蛹水解液的分子量测定

摘    要:凝胶过滤法是测定蛋白质分子量的常用方法之一,但标准蛋白质不易得到,且保存较困难。本工作用已知分子量的聚乙二醇作标样,采用凝胶过滤色谱外插法定量测定蚕蛹酶解液的分子量。实验色谱柱填料用Sephadex—50,聚乙二醇分子量为20000~1000,洗脱液用紫外光谱检测。根据洗脱体积与蛋白质分子量成直线关系的原理,作分子量与洗脱体积的关系曲线,并用已知分子量的细胞色素C验证。实验表明,该法所得结果与聚丙烯酰胺凝胶电泳和GPC法的测定结果一致。


DETERMINATION OF MOLECULAR WEIGHT ON SILKPUPA HYDROLYSIS PRODUCTS
Abstract:It is one the general methods of detemining molecular weight of protein by gel filtration chromatalogy (G. F. C.). but the reference agents (standard protein) are not easy to get and keep well. In this paper, the molecular weights of silkpupa hydrolysis products have been quantitatively determined with extenal reference by G.F.C.polyethlene glycol is used as reference agent. The fillers of column are sephadex G-50, the molecular weight of polyethlene glycol which is used in this paper is 20000-1000, and the fractions of elution are identificated by UV spectrophotometer. Determination of molecular weight of silkpupa hydrolyuis products was carried out according to principle that elution volume to molecular weight relationship was linear and was evidanced by detemination of Cytochromum C (Mw 12400)。Our studies is shown in Fig 1, the analysis result are agree withthe data of polyacrylamide-gel electrophores is and G. P. C. analysis.
