摘    要:建立了竹叶中总多酚含量的Folin-Ciocalteu比色测定方法.以没食子酸为对照品,对竹叶中总多酚含量测定的比色条件,包括检测波长、碳酸钠溶液用量、Folin-Ciocalteu试剂用量、反应温度及时间对吸光度的影响进行了研究.结果表明:在0.944-7.552μg/mL范围内,对照品校准曲线为y=0.1237x+0.0469,R2=0.9991,线性关系良好.优化的测定方法为样品提取液0.5mL,Folin-Ciocalteu试剂1.0mL、16%碳酸钠溶液4.0mL、30℃避光反应1.5h,此条件下平均回收率为105.2%(RSD=4.225%).采用建立的检测方法,测定了12种竹叶中总多酚含量,结果表明竹叶中总多酚平均含量为3.265mg/g;含量最高为龙竹6.059mg/g,最低为佛肚竹0.968mg/g.该方法简便快速、结果准确、重现性和稳定性好,可用于竹叶中总多酚含量的测定.

Abstract:An assay method for the determination of total polyphenol in bamboo leaves was set up. The determination conditions of total polyphenol content in bamboo leaves including the detection wavelength, the amount of sodium carbonate solution, Folin-Ciocalteu reagent, reaction temperature and time were studied and gallic acid was used as standard. The standard curve was linear in the range of 0. 944-7. 552μg /mL(y=0. 1237x+0. 0469,R2= 0. 9991) and the optimum conditions for determining total polyphenol in bamboo leaves were as followed:0. 5mL of the bamboo leaves extract polyphenol, 1.0mL of Folin-Ciocalteu reagent dosage and 4.0mL of 16% sodium carbonate solution, treatment at 30℃ for 1. 5h,the average recovery was 105. 2%(RSD=4. 225%). Under these conditions,the average polyphenol content among 12 kinds of bamboo leaves was 3. 625mg/g, the highest content was 6. 059mg/g in Dendrocalamus giganteus Munro leaves,and the lowest content was 0. 968mg/g in Bambusa ventricosa McClure leaves. This method was simple and quick,the result was accurate, repeatable and stable. It can be used for the determination of total polyphenol content in bamboo leaves.
