胶束增敏催化荧光法测定血红蛋白

摘    要:基于血红蛋白(Hb)的过氧化物酶活性,催化过氧化氢氧化对甲基酚的反应体系,发现表面活性剂SDS对该体系反应速率有明显的增强效应;从而建立了新的高灵敏测定Hb的催化荧光分析法。在1.5×10-3mol/L SDS存在下,测定Hb的线性范围1×10-9~8×10-8mol/L,检出限为3.4×10-10mol/L,用于尿中Hb的测定,结果满意。

Abstract:A highly sensitive catalytic-fluorimetric method based on Hemoglobin (Hb) enzymatic activityto catalyze the oxidation of p-Cresol with hydrogen peroxide for the determination of hemoglobin in the presence of sodium dedecyl sulphonate (SDS) has been developed, indicated that the initial rate of this reaction was accelerated with the addition of SDS. The linear relationship between the initial reaction rate and the hemoglobin concentration was in the range of 1×10-9~8×10-8mol/L. The detection limit was 3.4×10-10mol/L. The method was used to determine hemoglobin in the urine samples with satisfactory result.
