摘    要:利用自组装单分子膜技术,将巯己基修饰的具有乙肝病毒(HBV)DNA序列特异性的单链DNA探针固定在金电极表面,制得DNA电化学传感器;以电活性的Hoechst 33258为指示剂,考察了该传感器对血清样品中乙肝病毒DNA的响应;探讨DNA电化学传感器在临床检测中的应用;将传感器法与荧光聚合酶链反应(PCR)法进行对比,两者的分析结果具有一致性。

Detection of Hepatitis B Virus in Serum Sample by DNA Electrochemical Sensor
Abstract:A DNA electrochemical sensor was prepared by fixing thiolhexanyl _ modified ssDNA probe with parti _ cular DNA sequence of hepatitis B virus(HBV) on the surface of an Au electrode using self _ assembled monomolecular film technology. The response of the sensor to DNA of HBV in serum samples was examined using Hoechst 33258 as an electroactive indicator. The clinical applicability of the sensor was discussed. The determination results of practical samples obtained by the DNA electrochemical sensor method were consistent with those obtained by fluorescence polymerase chain reaction method.
