摘    要:我们成功地用微区拉曼光谱探针技术获得了培养单个活体 Hela 细胞核中染色质的拉曼光谱。从光谱中我们看到正常 Hele 细胞核蛋白的二级结构为 a 螺旋,DNA 为 A 型构象。经10~(-3)moI/L 过氧化氢处理20分钟及8Gy x 射线照射后的 Hela 细胞核蛋白的二级结构出现反平行β折叠,DNA 出现 B 型构象。这种结果与我们的假设是相符合的。

Abstract:Well resolved Raman spectra of the in vivo chromosomes were obtained for single HeLa cells and used as the sensitive measure of structural changes to study the damage induced by X-ray irradiation and/or H_2O_2.based on the analysis and comparison of the scatter-ing lines of the chromosomal DNA and proteins,it was found that:(1)In untreated cells,the chromosomal proteins and DNA exhibit a α-helical and A-form conformation respectively; (2)Either treatment with 10~(-3) mol/L H_2O_2 or 8Gy 200KV X ray irradiation resulted in the appearance of β-Sheet in conformation of proteins and of B-form in the conformation of DNA.
