Abstract: | Abstract This paper reports the results of Raman spectroscopy of botulin type C. The results show that natural protein and renatured protein have dif- ferent space structure. The natured protein (BC_1)has predominantly a-helical and β-sheet structure whereas the renatured protein contains predominantly andom-coil content. The natured protein has also a gauche-gauche-gauche configuration of the C-C-S-S-C-C linkage while the Raman lines indicate thatthe geometric difference existing among the disulphide linkages of renatured protein are not large enough to be detected by Raman spectroscopy. The S- CH_2 and the CH_2-CH_2 bond of the former is trans-gauche configuration whereas the latter is gauche- trans configration. about 30%tyrosine residues are buried inside the molecules and 70%tyrosine residues are located On molecular sur-face in natured protein. |
