摘    要:制备了人胶质瘤细胞SWO-38条件培养基(SWO-38CM),经超滤浓缩,截留分子量大于10kD的SWO-38CM,并运用快速自动比色微量分析法检测其对体外培养的SD大鼠小脑皮质神经元的作用,结果表明,分子量大于10kD的SWO-38CM对神经元的作用,与对照组比较有明显的神经营养活性,分子量大于10kD的SWO-38CM经SephacrylS-200-HR凝胶层析并结合生物活性鉴定,获得了具有神

Abstract:Human glioma cell line (SWO-38) conditioned medium (SWO-38 CM)was prepared,ultratiltrated and concentrated, so the fraction of molecular weight (MW)>10 kD from SWO-38CM was obtained. The effect of SWO-38 CM fractions (MW>10 kD and MW<10kD)on theactivity of cultured cerebellar cortical neurons of Spraque-Dawley rat was measured by a rapidautomated colorimetric microassay, respectively. The data indicated that SWO-38 CM fraction(MW>10 kD) possessed significant neurotrophic activity to the neurons, as compared to thecontrol (P<0.05). Additionally, the active fraction of SWO-38 CM was purified further bySephacryl S-200-HR gel chromatography and peak Ⅱ was only identified to possess neurotrophicactivity.
