摘    要:以蛤蜊为研究对象,建立了水产品中胂蛋白的提取方法.采用超声波细胞破碎法以Tris-HCl作为缓冲溶液提取蛤蜊中的胂蛋白;对提取的胂蛋白进行电泳分离纯化,采用氢化物发生-原子荧光光谱法对蛋白条带中As含量进行测定.根据实验结果分析,胂蛋白的最佳提取条件为:超声功率190 W,超声波每次辐射时间/间歇时间为5 s/5 s,...

Abstract:A rapid and intuitive extraction method was established by using clam as the research object for the analysis of arsenic binding protein in aquatic products.By using ultrasonic cell crushing method and Tris-HCl as buffer solution to extract the arsenic binding protein in clam,choosing polyacrylamide gel electrophoresis to purify the arsenic binding protein,the arsenic content in protein bands was determined by using hydride generation-atomic fluorescence spectrometry.According to the experimental results,the optimal extraction conditions of arsenic binding protein were as follows:ultrasonic power of 190 W,ultrasonic irradiation time/intermittent time of 5 s/5 s,total ultrasonic working time of 11 min,and solid-liquid ratio of 1:25.The molecular weight range of purified arsenic binding protein was 40-50 KD and 60-120 KD,which provided data support for the identification and sequencing of arsenic binding protein.
