Abstract:To optimize the micellar electrokinetic chromatography (MEKC) method, the buffer composition, buffer pH, the content of organic additive and phosphate concentration are investigated and optimized by a statistic uniform design. The optimal MEKC buffer con- sists of 70 mmol/L of CTMAC, 20% acetonitrile and 70 mmol/L of phosphate at pH 2. 5. By the optimal MEKC method, ten crude peptides are analyzed and the results are compared with that obtained by the optimal CZE and HPLC. In view of the purity, migration time, column efficiency and the total number of peaks in each crude peptide, we might conclude that the MEKC method is superior to HPLC and HPLC superior to CZE.
