摘    要:研究了蛋白质在超细粒子固体表面吸附过程中构象的变化;选择的吸附剂粒径很小,可以直接应用圆二色性光谱测定蛋白质吸附层的二级结构变化;研究了pH、固体吸附表面在蛋白质吸附过程中对蛋白质构象的影响,得到了较好的实验数据,为进一步进行蛋白质吸附机理的理论研究提供了基础。

Abstract:In order to study the conformational changes during protein adsorption on particulate surfaces, it is necessary to use ultrafine particles. The average diameters of the silica oxide and titanium oxide particles employed in our case are 15 and 20 nm, respectively. Since the intensity of light scattered by the ultrafine particles is negligible, direct estimation of conformational changes of adsorbed protein molecules from circular dichroism(CD) spectra is feasible. The effects of adsorption pH and the solid surfaces on protein conformational changes were studied. Data on protein conformational changes thus obtained can serve as a basis for further theoretical studies on the protein adsorption mechanism.
