A quartz crystal microbalance (QCM) immunosensor was developed for the determination of the insecticide carbaryl and 3,5,6-trichloro-2-pyridinol (TCP), the main metabolite of the insecticide chlorpyrifos and of the herbicide triclopyr. The detection was based on a competitive conjugate-immobilized immunoassay format using monoclonal antibodies (MAbs). Hapten conjugates were covalently immobilized, via thioctic acid self-assembled monolayer (SAM), onto the gold electrode sensitive surface of the quartz crystal. This covalent immobilization allowed the reusability of the modified electrode surface for at least one hundred and fifty assays without significant loss of sensitivity. The piezoimmunosensor showed detection limits (analyte concentrations producing 10% inhibition of the maximum signal) of 11 and 7 μg l−1 for carbaryl and TCP, respectively. The sensitivity attained (I50 value) was around 30 μg l−1 for both compounds. Linear working ranges were 15-53 μg l−1 for carbaryl and 13-83 μg l−1 for TCP. Each complete assay cycle took 20 min. The good sensitivity, specificity, and reusability achieved, together with the short response time, allowed the application of this immunosensor to the determination of carbaryl and TCP in fruits and vegetables at European regulatory levels, with high precision and accuracy.  相似文献

A flow-injection procedure is described for the determination of carbaryl based on its inhibition effect on luminol-cobalt(II) chemiluminescence reaction in alkaline medium in the presence of hydrogen peroxide. The calibration data over the range 5.0?×?10?7 to 20?×?10?6?M give a correlation coefficient (r 2) of 0.9972 with relative standard deviations (RSD; n?=?4) in the range of 1.0–2.1% with a limit of detection (3?×?blank noise) of 2.37?×?10?7?M for carbaryl. The sample throughput was 120?h?1. The effects of some carbamates, anions, and cations were studied on luminol CL system for carbaryl determination. The proposed method has been applied to determine carbaryl in natural waters.  相似文献

A simple method for the rapid and simultaneous analysis of dichlorvos (DDVP), malathion, carbaryl, and 2,4-dichlorophenoxy acetic acid (2,4-D) in citrus fruit, which uses flow-injection ion spray ionization tandem mass spectrometry, has been developed for the first time. The method involves the combined use of stable isotopically labeled internal standards (DDVP-d6, malathion-d10, carbaryl-d7, and 2,4-D-d5) and a multiple reaction monitoring technique. The average recoveries for the pesticides at the same concentrations as their tolerance levels (DDVP: 0.1-0.2 μg g−1; malathion: 0.5-4.0 μg g−1; carbaryl: 1.0 μg g−1; 2,4-D: 1.0-2.0 μg g−1) ranged from 90 to 119% with the relative standard deviation (R.S.D.) ranging from 1.0 to 13.1% (n = 5). Analysis time, including sample preparation and determination, was only 15 min. The present method is effective for screening DDVP, malathion, carbaryl, and 2,4-D in citrus fruit.  相似文献

The absorption and fluorescence spectra of carbaryl (CB), carbofuran (CF) and carbendazim (MBC) have been studied. Fluorescence lifetime and fluorescence quantum yields are also reported as well as the influence of pH, solvent and presence of humic acids on fluorescence. The limit of detection (LD) of the three compounds has been measured by direct analysis by laser-induced fluorescence (LIF) using a pulsed YAG laser with an Optical Parametric Oscillator (OPO) as excitation source and an Intensified Charged Coupled Device (ICCD) camera for the fluorescence detection. Instrumental LD found for CB, for MBC and for CF are respectively 4, 50 and 1000 ng L−1. In tap water, the LD obtained is 800 ng L−1 for MBC and 20,000 ng L−1 for CF. For CB, the use of a time shift between excitation and emission allows to reach a LD of 20 ng L−1 in tap water.  相似文献

Multi-analyte SPR immunoassays for environmental biosensing of pesticides   总被引:1,自引:0,他引:1  
Multi-analyte detection of environmentally relevant pesticides is performed by using a two-channelled surface plasmon resonance (SPR) biosensor. The special design of the SPR instrument allows the determination of several analytes (DDT, chlorpyrifos and carbaryl) via different immobilization formats. First, simultaneous pesticide monitoring is possible by flowing chlorpyrifos, carbaryl or DDT samples separately over each channel of the SPR system, wherein their corresponding recognition element was previously immobilized. The second approach is based on the multiple and combined immobilization of several analyte recognition elements on the sensing surface of one individual flow cell. In this format, the analysis time for all three pesticides varied from 40 to 60 min depending on the number of regeneration cycles. In most cases, similar detection limits were attained for the target analyte irrespective of the assay format, with sensitivity values at the nanogram per litre level (18–50 ng L−1). The assay reproducibility was proved through the repeated use of the same sensor surface for over more than 200 assay cycles, whereas the absence of biosensor response to non-related analytes showed the specificity and reliability of the analysis. 利用直接竞争酶联免疫吸附分析技术研发了一种快速、灵敏的膜载体酶标记dip-stick农残西维因检测方法。该方法以带正电尼龙膜(Millipore)为固相载体,将西维因抗体包被于膜上,包被有抗体的膜插入含有西维因和西维因酶标抗原的混合液中竞争反应10 min后直接目视判断分析结果。该方法的整个检测过程只需15 min,对西维因的检出限可达到10μg.L-1,且稳定性较好。通过与高效液相色谱法(HPLC)比较,验证了该检测方法的有效性,可作为品质控制和快速定性检测的一种有效工具。  相似文献

甲萘威在0.01mo1/L KOH溶液中可降解为强荧光产物。十六烷基三甲基溴化铵对其增敏后,荧光强度达到其自身荧光强度的6倍。基于此,建立了对残留甲萘威进行荧光测定的新方法。线性动态范围为2.00~1.70×103μg/L;检出限为0.95μg/L;样品的加标回收率在90.2%~99.9%。本法成功地用于叶菜中残留甲萘威的测定。  相似文献

Amperometric acetylcholinesterase biosensors have been developed for quantification of the pesticides carbofuran, carbaryl, methylparaoxon, and dichlorvos in phosphate buffer containing 5% acetonitrile. Three different biosensors were built using three different acetylcholinesterase (AChE) enzymes—AChE from electric eel, and genetically engineered (B394) and wild-type (B1) AChE from Drosophila melanogaster. Enzymes were immobilized on cobalt(II) phthalocyanine-modified electrodes by entrapment in a photocrosslinkable polymer (PVA-AWP). Each biosensor was tested against the four pesticides. Good operational stability, immobilisation reproducibility, and storage stability were obtained for each biosensor. The best detection limits were obtained with the B394 enzyme for dichlorvos and methylparaoxon (9.6 × 10−11 and 2.7 × 10−9 mol L−1, respectively), the B1 enzyme for carbofuran (4.5 × 10−9 mol L−1), and both the B1 enzyme and the AChE from electric eel for carbaryl (1.6 × 10−7 mol L−1). Finally, the biosensors were used for the direct detection of the pesticides in spiked apple samples.  相似文献

The anodic voltammetric behavior of carbaryl on a boron‐doped diamond electrode in aqueous solution is reported. The results, obtained by square‐wave voltammetry at 0.1 mol L?1 Na2SO4 and pH 6.0, allow the development of a method to determine carbaryl, without any previous step of extraction, clean‐up, preconcentration or derivatization, in the range 2.5–30.0×10?6 mol L?1, with a detection limit of 8.2±0.2 μg L?1 in pure water. The analytical sensitivity of this electrochemical method diminished slightly, from 3.07 mA mmol?1 L to 2.90 mA mmol?1L, when the electrolyte was prepared with water samples collected from two polluted points in an urban creek. In these conditions, the recovery efficiencies obtained were around 104%. The effect of other pesticides (fenthion and 4‐nitrophenol) was evaluated and found to exert a negligible influence on carbaryl determination. The square‐wave voltammetric data obtained for carbaryl were typical of an irreversible electrode process with mass transport control. The combination of square‐wave voltammetry and diamond electrodes is an interesting and desirable alternative for analytical determinations.  相似文献

在NaOH介质中,铁氰化钾氧化鲁米诺产生化学发光,甲萘威对该发光反应有较强的抑制作用且抑制发光强度与甲萘威的浓度呈较好的线性关系,基于此性质建立了测定甲萘威的新方法。在实验的优化条件下,测定甲萘威的检出限为5.5×10-11mol.L-1,线性范围为1.5×10-10—4.0×10-8mol.L-1,对5.0×10-9mol.L-1甲萘威进行了11次平行测定,其RSD为1.74%。该法用于合成样品中甲萘威的测定,结果令人满意。  相似文献
