在体积排阻色谱(SEC)法测定胸腺肽分子量校准曲线过程中,流动相中乙腈的比例对核糖核酸酶A、人胰岛素、胸腺肽α1和生长激素释放抑制因子4种蛋白的保留时间有重要影响,进而影响校准曲线的线性关系。当乙腈比例为75%时,胸腺肽分子量校准曲线线性最好,此时分子量校准方程为y=-3.138 6x+21.724,线性相关系数r2=0.988 5。4种蛋白的理论塔板数在45 783~63 345之间,拖尾因子在0.96~1.18之间,分离度在3.52~8.82之间。SEC法测定胸腺肽分子量校准曲线的液相色谱条件对4种蛋白的分离效果优异,分子量校准曲线线性良好,可用于胸腺肽制剂中高分子量物质的检测。  相似文献

Thymosin‐β4 is a peptide found at high concentrations in a wide range of mammalian cells. It has been shown to be elevated in metastatic cells. In this study, the surface‐enhanced Raman spectrum of thymosinβ4 was obtained using a silver nanorod array surface‐enhanced Raman scattering substrate. Significant Raman peaks were found, and the corresponding vibrational modes were assigned based on previously published literature, with amide A–B, amide I, and amide III modes all identified. The majority of the remaining peaks were assigned modes based on the spectra of lysine and glutamic acid, the two most abundant amino acids in the thymosinβ4 sequence. A standard Raman spectrum of thymosinβ4 was also obtained and analyzed for comparison. Because thymosinβ4 plays a significant role in regulating the formation of cellular cytoskeleton and other several biological processes, its Raman spectrum will be of use to researchers. PEG修饰是改善蛋白质及肽类药物药代动力学特性的有效途径。然而与蛋白质相比,肽类化合物的分子较小,PEG的分子体积较大,其长链很可能会遮蔽肽的活性位点。因此,肽类化合物PEG修饰的位置和数量对于保持肽的生物活性至关重要。为阐明PEG修饰的位置与肽生物活性之间的关系,对肽类药物日达仙(胸腺素α1,Tα1)进行了定点修饰。Tα1具有α-螺旋、β-转角和无规卷曲的结构区域。分别在这些区域选择不同的位点进行PEG修饰。PEG的定点修饰是通过引入Cys,利用其-SH与mPEG-MAL的特异性反应而实现的。Con A刺激下的脾细胞产生IFN-γ试验的初步结果表明,PEG修饰对活性的影响与修饰的位置有一定的关系,大多数情况下,PEG修饰能保持Tα1的免疫活性。PEG修饰的位点对于保持肽的生物活性是很重要的。  相似文献

本文以外周血白细胞总数、淋巴细胞与粒细胞比率、ANAE~+淋巴细胞百分率、SP与DP淋巴细胞比率、骨髓有核细胞数和~3H-TdR掺入到骨髓有核细胞量的变化,探讨了胸腺素对经450仑辐射鼠体的抗辐射效应。结果表明:小牛胸腺素对白细胞,特别对淋巴细胞的回升,对T淋巴细胞,特别对T_μ淋巴细胞损伤防护和回升,对骨髓有核细胞数和~3H-TdR掺入量都有明显的促进作用。结果提示,小牛胸腺素具有调整因辐射所引起的鼠休淋巴细胞低降,从而恢复免疫功能的效应。  相似文献

N-Terminally acetylated thymosin beta4, a species implicated for use as a cancer biomarker, was identified in a human lung cancer cell line using ion trap tandem mass spectrometry at the whole protein level. Ion-ion proton transfer reactions were used for parent ion concentration/manipulation and to simplify interpretation of product ion spectra. Dissociation data for the +6 to +3 charge states are reported. As is usually the case, structural information available from the ion trap collisional activation of the protein is sensitive to parent ion charge state. Each parent ion charge state selected, however, provided sufficient information to make a confident identification. Furthermore, each charge state provided relatively rich fragmentation. Therefore, any of the charge states can be used to detect with high specificity thymosin beta(4) in a complex protein mixture. There are advantages associated with the rapid detection of protein biomarkers at the whole protein level, as opposed to the peptide level following protein digestion, particularly for relatively small protein and polypeptide biomarkers. Having identified and characterized the protein, product ion spectra obtained directly, without recourse to ion-ion proton transfer reactions, can be used for library matching. However, ion-ion proton transfer reactions for parent ion concentration and charge state purification are advantageous in addressing relatively complex mixtures.  相似文献
