An efficient biocatalytic process has been developed to obtain optically pure (1S,4R)-N-(benzylcarbamoyl)- 4-aminocyclopent-2-en-1-ol which can be used as the key intermediate of ticagrelor. In this research, several N-(benzylcarbamoyl)-4-aminocyclopent-2-en-1-ol derivatives have been investigated in which Candida antarctica lipase B (CALB) was used to catalyze the asymmetric hydrolysis reaction. As expected, some of these substrates successfully gave (1S,4R)-N-(benzylcarbamoyl)-4-aminocyclopent- 2-en-1-ol in >98% enantiomeric excess (ee) with conversion yields up to 45%.  相似文献

Starting from readily available oleic and erucic acid, macrocyclic nonadecalactone (C19) and tricosalactone (C23) can be synthesized in polymerization grade purity in a four‐step reaction sequence. Ring‐opening polymerization (ROP) of these strainless macrolactones can be performed utilizing an enzyme as a catalyst. Despite the missing ring‐strain as key driving force for smaller (strained) lactones, high molar masses (M n ≈ 105 g mol−1) can be accessed in an entropically driven ROP. Polyester‐19 and polyester‐23 prepared feature melting temperatures well above 100 °C. Further analysis of the mechanical properties of these materials displays the resemblance to polyethylene. For example, Young's moduli on the order of 600 MPa are observed as a result of the high crystallinity of the polymer.

Abstract

The aliphatic polyesters are normally synthesized by ester interchange reactions or direct esterification of hydroxyacids or diacid/diol combinations. Biotransformation, utilizing the enzymes as catalysts, was accepted as an alternative route for the synthesis of aliphatic polyesters and offers various advantages compared with the conventional, metal-catalyzed polymerization reactions. Previous studies indicated that lipase-catalyzed polycondensation reactions between diols and diacids occurred preferentially at primary hydroxyl groups of diols, when diols contained both primary and secondary hydroxyl groups. In this work, we investigated lipase-catalyzed polycondensation of diacids and secondary hydroxyl group–containing diols, and successfully synthesized polyesters by polycondensation with secondary hydroxyl groups as well as primary hydroxyl groups. Various diols, glycerol, 1,2-propanediol, 1,3-butanediol, 2,3-butanediol, and 2,4-pentanediol were tested for the polycondensation. The polymerization was achieved by heating a mixture of lipase B, sebacic acid, and the diols in anhydrous toluene at 100 °C for 72 h. The resulting polymers were characterized by 1H and 13C NMR spectroscopy, Fourier transform–infrared spectroscopy, thermogravimetric analysis, and gel permeation chromatography.  相似文献

In this study, we have investigated the antifungal activity of ZnO nanorods prepared by the chemical solution method against Candida albicans. In the study, Zinc oxide nanorods have been deposited on glass substrates using the chemical solution method. The as-grown samples are characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD). X-ray diffraction (XRD) showed zinc oxide nanorods grown in (0 0 2) orientation. The antifungal results indicated that ZnO nanorod arrays exhibit stable properties after two months and play an important role in the growth inhibitory of Candida albicans.  相似文献

Pentacyclic triterpenoids α- and β-amyrin possess a wide range of biological and pharmacological activities. High structural similarity between these two structural isomers makes their chromatographic separation an ineffective and tedious choice. In this study, Candida rugosa lipase catalyzed separation protocol for the isolation of individual isomers has been developed. In the presence of vinyl acetate as the acyl donor, Candida rugosa lipase carried out acetylation of β-amyrin more efficiently as compared to α-amyrin leading to a kinetic separation. The conditions of transesterification reaction were optimized systematically, which was utilized to separate α- and β-amyrin from a mixture obtained from the latex of Plumeria obtusa.  相似文献

Potentiometric biosensors based on Candida rugosa lipase was described for the detection of organophosphorus pesticide; methyl-parathion and tributyrin. Lipase was immobilized on the glass electrode by means of a gelatin membrane, which is then cross-linked with glutaraldehyde. The principle of the biosensor is based on the measurement of pH variation which was recorded in millivolts due to the enzymatic hydrolysis of tributyrin to butyric acid. For the inhibitor detection, biosensor responses were measured after pesticide treatment, which caused a drop in enzyme activity because of the irreversible inhibition. Reactivation conditions of the reused enzyme electrodes were also investigated by pyridine-2-aldoxime methiodide (2-PAM). The limit of detection for tributyrin was estimated as 93?µM for lipase sensor within the linear range of 65–455?µM.  相似文献

以布洛芬乙酯为反应底物,探索了酶促水解法拆分布洛芬的工艺。以曲拉通X-100为表面活性剂,利用南极假丝酵母脂肪酶B(CAL-B)催化拆分外消旋布洛芬乙酯,并对表面活性剂含量、有机溶剂种类和添加量、酶量、缓冲溶液pH、温度及时间等反应条件进行了优化;得到的最佳反应条件为:在pH为9.0的缓冲溶液中,添加80 mg曲拉通X-100、50 μL二氯甲烷和15 mg CAL-B,于30℃下反应48 h;在该条件下,S-布洛芬乙酯的剩余率(C)为65%,布洛芬乙酯的对映体过量值(ees)为94%。  相似文献
