当前,在医学核磁共振成象领域内,临床上广泛采用的是近似的自旋密度象、T1-和T2-加权密度象。但是,由于人体的正常组织和肿瘤之间的密度差别不大,从质子密度象很难区分人体的正常组织与肿瘤,而肿瘤与人体正常组织之间的在弛豫时间T1、T2的数值上差别较大。另外,自七十年代以来,大量关于离体(in vitro)核磁共振弛豫时间测量的文献表明,肿瘤的核磁共振弛豫时间T1,T2值具有明显的规律性,从而利用核磁共振弛豫时间成象的方法,及人体正常组织、良性及恶性肿瘤的活体(in vivo)T1,T2数值的测量,将有助于实现肿瘤识别的定量方法。 相似文献

The relative contributions of intravascular and intracellular compartments to the proton transverse relaxation of the isolated and excised rat liver were determined during the phagocytosis of superparamagnetic particles. The evolution of the proton transverse magnetization of the organ perfused with increasing doses of starch-coated magnetic microspheres was followed up using a Carr–Purcell–Meiboom–Gill sequence with various echo times. From the multiexponential fit of the echo train, the amplitudes and the relaxation ratesR2of the liver tissue were obtained. The results clearly indicate that shortly after contrast medium administration, an internalization takes place which can be followed by the rapid and biphasic evolution of the transverse relaxation rate of the water protons. A very fast decaying component looking like an initial loss of the magnetization is observed together with an increase of the relaxation rate of the remaining water tissue. This regime is strongly dependent on both the echo time and the iron concentration, a behavior characteristic of the agglomeration of magnetic particles. The examination of the liver tissues by electron microscopy shows that this clustering arises in cytoplasmic vacuoles. 相似文献

Modern drug development requires technologies that allow rapid translation from the preclinical to the clinical stage. It is obvious that non-invasive imaging modalities such as magnetic resonance imaging (MRI) will play a central role in this regard. This article reviews the use of structural and functional MRI readouts for characterization of central nervous system (CNS) disorders and evaluation of the efficacy of potential CNS drugs. Examples comprise dementia of Alzheimer's type, cerebral ischemia, and neuroinflammation covering both clinical and preclinical aspects. In these examples MRI has been used to obtain relevant structural information on brain atrophy, on the location and extent of ischemic brain areas, and on the number and distribution of demyelinated plaques. These structural data are complemented by readouts assessing the functional consequences associated with the pathomorphological changes. In the last decade, MRI has evolved into a standard tool for the development of CNS drugs. With regard to target-specific/molecular imaging applications MRI is limited by its inherently low sensitivity; complementary imaging modalities utilizing optical and radionuclear reporter systems will thus be required. 相似文献

Ro 28-2653 is a new synthetic inhibitor of matrix metalloproteinases. The ability of these enzymes to degrade various components
of the extracellular matrix seems to play a major role in tumors progression and is potentially effective against bronchial
remodeling in asthma and BPCO. Ro 28-2653 is very poorly soluble in water. This low solubility estimated at about 0.56 μg/ml
in water at 25 °C gives rise to difficulties in pharmaceutical formulation of oral, injectable or nebulizable solutions. The
purpose of our study is to prepare and to characterize inclusion complexes between Ro 28-2653 and cyclodextrins and to investigate
the biopharmaceutical repercussion of the inclusion of the active substance.
The complex formation was investigated by phase solubility studies. 1H-NMR spectroscopy and molecular modeling studies were carried out to elucidate the structure of the inclusion complex between
Ro 28-2653 and cyclodextrin. Oral, intravenous and nebulizable solutions of Ro 28-2653 were developed with cyclodextrin. The
in vivo studies were performed on healthy sheep for the pharmacokinetic evaluation of the oral and intravenous formulations

A composite of iron oxide nanoparticles and mineral matrix has been studied by XRD, Mössbauer spectroscopy, and TEM. Magnetite and superparamagnetic magnetite have been identified by Mössbauer spectroscopy in the nanocomposite. A relationship between the hyperfine parameters and iron oxide particle size has been confirmed by TEM. The optimal concentration of “magnetite—bentonite” composite, when the MRI signal is fully reduced, was found for using this composite as a negative contrast agent. 相似文献

Although magnetic resonance imaging (MRI) is one of the most important imaging modalities of the central nervous system (CNS),
one of the main drawbacks of MRI is its limited specificity. This can potentially be partially alleviated by target-specific
contrast agents. In the present paper we describe a simple high yield synthesis of two such gadolinium-based spiperone targeted
MRI contrast agents, 1a and 1b. The R1 relaxivities of 1a and 1b were evaluated and found to be 5.94 and 8.31 mM−1 s−1, respectively at 9.4T, while their R2 relaxivities at the same magnetic field were found to be 18.05 and 22.60 mM−1 s−1, respectively. In addition and very importantly compound 1a, which is a gadolinium-based, spiperone-targeted MRI contrast agent, was found to preserve some of the spiperone affinity
toward the dopamine D2 receptor. Compounds 1a and 1b thus represent potential agents for in vitro dopamine receptor imaging using MRI in experimental models.

目的研究衢州地区痰涂片阳性肺结核患者对常用抗结核药物的耐药情况,为结核病控制和临床用药提供对策.方法对衢州地区的743例痰涂片阳性肺结核患者痰标本进行酸性改良罗氏培养基痰培养,将培养阳性菌株以硝基苯甲酸(PNB)培养基和噻吩-2-2羧酸肼(TCH)培养基进行菌种鉴定,采用绝对浓度间接法对异烟肼(INH)、利福平(RFP)、链霉素(SM)、氧氟沙星(OFLX)、乙胺丁醇(EMB)、卡那霉素(KM)6种抗结核药物进行敏感性试验.结果743例获得菌种鉴定和药物敏感试验结果者中有结核分枝杆菌复合群728例(97.98%),非结核分枝杆菌15例(2.02%).728例结核分枝杆菌复合群中总耐药率为26.63%,初治耐药率为19.35%,复治耐药率为57.32%;总耐多药率为5.36%,初治耐多药率1.39%,复治耐多药率36.59%.6种抗结核药物的耐药顺序依次为INH 8.20%、SM 7.74%、OFLX 4.80%、RFP 1.86%、KM 1.24%、EMB 1.08%.结论衢州地区肺结核患者中耐药情况较为严重,应开展分枝杆菌培养和药敏试验并根据药敏结果组成个体化化疗方案,同时进一步落实和提高非住院肺结核患者全面监督化学治疗(DOTS)质量,提高各类患者的治愈率,减少各种耐药的发生概率. 相似文献
