A method is formulated for the identification of an unknown physical parameter of a fluid-filled pipe using the measurement of sound speed in the pipe. The method uses a simple formula which provides the relationship between the sound speed and a few physical parameters of the pipe: thickness, diameter, wall material constants and fluid constants. Once the sound speed in the pipe is measured, the simple formula can be used to extract one of the unknown parameters providing the remaining ones are known.The sound speed in the pipe is measured using a 3-transducer array. In order to demonstrate the potential of the technique the results of several measurements obtained in a water-filled steel pipe are presented.The required accuracy of the measurement of sound speed and of the specification of known parameters is analysed. The accuracy depends on the unknown parameter which is to be identified. For example, if the pipe thickness is the unknown parameter, the other parameters have to be known within a very narrow margin of error. On the contrary, if the fluid properties have to be identified the needed accuracy of known parameters gets much lower.  相似文献

Immunoassays are one of the most convenient methods for environmental monitoring, but have been limited so far by low yield and low affinity antibodies (Abs). With the advent of recombinant Ab (rAb) technology and the expression of these Abs in organisms such as yeast, bacteria, insects and plants, widespread monitoring of our food and environment for organic contaminants using immunoassays has become feasible. A multitude of immunoassays have been developed to detect pesticides in soils, ground and river water, foods and biological samples, such as urine and semen. In this paper, we describe advances in Ab production, the move away from using animals, phage-display technologies and the advent of plant-derived Ab expression systems. Finally, we describe future possibilities in Ab technology for environmental monitoring.  相似文献

A new alternative method for bioprocess monitoring based on bienzymatic analytical microreactors integrated in a flow injection analysis (FIA) system is described. Glucose-, alcohol-, lactate-, galactose- and l-amino acid oxidases (GO, AO, LacO, GalO and LAAO) and horseradish peroxidase (HRP) are immobilized on controlled pore glass (CPG) and used for the development of glucose, ethanol, lactate, galactose and amino acid sensors. The analytical methodology is based on HRP catalysed reaction of hydrogen peroxide produced by oxidases with phenol-4-sulfonic acid and 4-aminoantipyrine. The immobilized enzymes are characterized and used for preparation of the packed bed analytical microreactors. Shelf life and operational stability of the microeactors are determined. GO/HRP, AO/HRP and LAAO/HRP microreactors showed excellent shelf life, they could be stored and reused for more than 6 months with no or very little activity loss, while GalO/HRP and LacO/HRP could be stored for shorter periods of time (10-20 days). Operational stability of GO and LacO microreactors was very good: an equivalent to 16,900 FIA injections of 25 μl to a LacO microreactor resulted in loss of half of its activity, immobilized GO was so stable that it was impossible to evaluate enzyme halflife. Immobilized GalO and LAAO lose their operational activity much faster: approximately 1400 and 8000 FIA injections of the respective substrate solution in a FIA set-up resulted in 50% activity loss. The methods with all the described microreactors were successfully validated using off-line samples from S. cerevisiae, E. coli and mesenchymal stem cell cultures with HPLC as the reference method.  相似文献

A sequential injection analysis (SIA) system was developed to monitor the concentration of l-cysteine in biological processes on-line. It is based on the redox reaction of l-cysteine with iron(III) in the presence of 1,10-phenanthroline (phen) and the detection of the red-iron(II)-phen complex with a spectrophotometry. The system was fully automated using software written in the LabVIEW™ development environment. A number of system variables such as the flow rate of the carrier buffer solution, the volume ratio of the sample to the reagents, and the reaction coil length, etc., were evaluated to increase the sensitivity and performance of the SIA system. Under partially optimized operating conditions the performance of the SIA system was linear up to a concentration of l-cysteine of 1 mM (R2 = 0.998) with a detection limit of 0.005 mM and a sample frequency of 15 hr−1. The SIA system was employed to monitor the concentration of l-cysteine on-line in a continuously stirred reactor and a fermentation process of Saccharomyces cerevisiae. The on-line monitored data were in good agreement with the off-line data measured by a HPLC with a fluorescence detector (n = 15, R2 = 09899).  相似文献

The acid-catalysed esterification of myristic acid with isopropanol was studied by using near-infrared spectroscopy (NIR) in combination with soft-modeling curve resolution (MCR) methodology with a view to establishing the effect of experimental variables on the reaction kinetics. The reaction was conducted at temperatures above the boiling point of the alcohol, with continuous addition of an isopropanol/water mixture to the reactor. Spectral and concentration profiles were determined by applying soft-modeling curve resolution methodology to a column-wise augmented data matrix containing the spectra for the pure components. MCR profiles were compared with reference values and found to depart from then by less than 3% as %RSE for concentrations and to exhibit correlation above 0.999 for spectra.The reaction kinetics as estimated from the concentration profiles was found to be pseudo-first-order. Also, the pseudo-first-order rate constant was found to depend on the flow-rate of the isopropanol/water mixture and its water content; although the constant decreased with increase in the proportion of water, a content of ca. 15% could be used without important retarding effects on the kinetics. The proposed NIR-MCR method allows the rate constant and the influence of the initial water content to be determined with a view to minimizing consumption of the raw materials and optimizing the experimental conditions.  相似文献

A monitoring study was carried out in 1998-1999 on a medium-sized (c. 4580 km2) watershed of the Dyle (or Dijle) river in central Belgium, composed of both rural and urbanized areas. This watershed may be considered as representative of a large part of the country. Samples were taken each month from seven sites along the course of the river, plus one from its major affluent the Demer. The eight molecules monitored were the main herbicides used on the major crops in the area, plus some used in nonagricultural sectors. The concentrations found were mostly in the 0-3 μg/L range, with some peaks between 3 and 14 μg/L. The averages over all spring and summer samples analyzed ranged from 0.14 (bentazone) to 1.54 μg/L (diuron). Time and space specific patterns could be observed.  相似文献

The interaction between antibody and antigen is characterised by relatively high affinity and specificity, making this type of reaction a prime candidate for use as an analytical tool. The interaction may be combined with biosensors in the production of immunosensors for environmental monitoring. Polyclonal and monoclonal antibodies have had a significant impact in analytical detection systems over the past few decades with antibody fragments becoming important in recent years. Production of antibodies to small haptens requires the initial conjugation of hapten to a larger carrier molecule. Once hapten-carrier conjugates have been produced, polyclonal, monoclonal and various antibody fragments may be produced by differing protocols. A critical step in the production of antibody fragments is the development of efficient screening procedures to identify suitable antibody-producing clones and this has been reviewed in this article. Various antibody types may then be used in the generation of immunosensors for the monitoring of environmental pollutants. The selection of the appropriate sensor technology applicable for the determination of an antibody-antigen interaction is of prime importance for immunosensor development. One example of such an application is surface plasmon resonance-based biosensors, as they provide real-time analysis of interactions between the antibody and antigen of interest.  相似文献

Scour monitoring is an important concern for subsea pipeline systems. The active-thermometry-based scour monitoring is based on the difference of heat transfer properties between sediment and sand, recognizes the surrounding media though temperature changing patterns during heating and cooling processes, and hence detects the free spans. Based on the scour monitoring system, a two-layer BP neural network was employed to process the monitoring data and achieved media recognition. The network's inputs were normalized temperature time histories. The network's outputs denoted different media: sediment and water. To validate the method, three experiments were conducted; one was used for training the network and the other two for testing. Also, the effect of noise on the network's performance was studied through simulation. The results demonstrated the feasibility and robustness of the neural network.  相似文献

The wide use of pesticides can lead to environmental and human adverse effects. Diazinon, as an organophosphorous pesticide, is used in agriculture because of its low cost and high efficiency on insects. Due to the increasing application of pesticides, accurate analytical methods are necessary. The aim of this work was modification of carbon paste electrode composition and applying it as a sensor for determination of diazinon in biological and environmental samples. Multi‐walls carbon nanotubes and a molecularly imprinted polymer were used as modifiers in the sensor composition. A molecularly imprinted polymer and a non‐imprinted polymer were synthesized for applying in the electrode. After optimization of electrode composition, it was used to determine the analyte concentration. Instrumental parameters affecting the square wave voltammetric response were adjusted to obtain the highest current intensity. The modified electrode with MIP showed very high recognition ability compared to the electrode containing NIP. The obtained linear range was 5×10−10 to 1×10−6 mol L−1. The detection limit of the sensor was 1.3×10−10 mol L−1 and the relative standard deviation for analysis of target molecule by the proposed sensor was 2.87 %. This sensor was used to determine the diazinon in real samples (human urine, tap, and river water samples) without special sample preparation before analysis. The optimization of electrode composition containing mentioned modifiers improved its response considerably.  相似文献

Abstract

A new automated method for measuring cyclosporine in plasma has been introduced by Abbott Laboratories, involving the TDx instrumentation and fluorescence polarization immunoassay. We have used currently the Sandoz Ciclosporin RIA for cyclosporine therapeutic monitoring for a long time. In order to find a method that will decrease the labor requirements associated with RIA procedure, we used specimens from 64

transplant patients to compare, both from analytical and clinical points of view, the Abbott assay with the Sandoz Ciclosporin assay. We believe that the Abbott assay offers some analytical advantages over the ciclosporin RIA procedure and can be used with confidence for cyclosporine therapeutic monitoring for kidney, heart and allogeneic bone marrow transplantation.  相似文献
