Drug-induced renal failure (DIRF) poses a serious medical complication with high mortality risk. However, early diagnosis or prognosis of DIRF remain challenging, as current methods rely on detecting late-stage biomarkers. Herein we present a library of zwitterionic unimolecular hemicyanines (ZCs) available for constructing activatable reporters to detect DIRF since its initial stage. Zwitterionic properties of these probes are achieved through interspersedly integrating alkyl sulfonates and quaternary ammonium cations onto hemicyanine skeleton, which result in record low plasma protein binding (<5 %) and remarkable renal clearance efficiencies (≈96 %). An activatable reporter ZCRR is further developed by masking the optimal candidate ZC6 with a tetrapeptide specifically cleavable by caspase-8, an initiating indicator of apoptosis. In living mice with cisplatin-induced DIRF, systematically administered ZCRR efficiently accumulates in kidneys and responds to elevated caspase-8 for near-infrared fluorescence signals ‘turn-on’, enabling sensitive detection of intrarenal apoptosis 60 h earlier than clinical methods, and precise evaluation of apoptosis remediation effects by different medications on DIRF mice. As it's urinary excretable, ZCRR also allows for remote detection of DIRF and predicting renoprotective efficacy through in vitro optical urinalysis. 用降解法制得了Dawson结构铬、铁取代的磷钼钨酸钾,并将其与溴化(E)-N-丁基-4-(2-(4-二甲氨基苯基)乙烯基)吡啶反应制备了有机复合材料.通过元素分析和TG-DTA确定了配合物的组成分别为K7[P2Mo4W13M(H2O)O61](M=Cr(1),Fe(2))和(C19H25N2)6K3[P2Mo4W13MO62](M=Cr(3),Fe(4)).利用红外光谱、紫外-可见光谱、固体漫反射紫外-可见-近红外光谱、X射线光电子能谱以及荧光光谱对上述化合物进行了表征,并研究了复合材料中无机与有机组分间的相互作用及其荧光性质. 相似文献

The design of solvatochromic fluorescent nucleosides with visible excitation wavelengths is a goal towards the generation of modified oligonucleotides for fluorimetry-based molecular imaging. Herein, two hemicyanine-linked C5-2′-deoxyuridine nucleosides (PyI-dU and APPy-dU) have been synthesized by first generating hemicyanine-alkyne precursors that were attached via the alkyne moiety to 5-iodo-2′-deoxyuridine (5-I-dU) by Sonogashira coupling. The photophysical properties of the hemicyanine-linked dU probes have been characterized and compared to the corresponding properties of the hemicyanine-alkyne precursors. The nucleoside probe PyI-dU exhibits optical features that mimic the properties of the free hemicyanine-alkyne precursor, while the APPy-dU probe displays more favorable optical properties (longer excitation wavelength, brighter emission in water) than its precursor that is ascribed to π-stacking interactions between the hemicyanine dye with the dU nucleobase. Overall, probe APPy-dU is a superior solvatochromic fluorophore than PyI-dU suggesting its greater utility for fluorescent imaging applications. 相似文献

The near-infrared luminescence properties of three (E)-N-hexadecyl-N′,N′-dimethylamino-stilbazolium tetrakis(1-phenyl-3-methyl-4-benzoyl-5-pyrazolonato) lanthanide(III) complexes are described. These three complexes, containing trivalent neodymium, erbium and ytterbium, respectively, show near-infrared luminescence in acetonitrile solution upon UV irradiation. Luminescence decay times have been measured. The complexes consist of a positively charged hemicyanine chromophore with a long alkyl chain and a tetrakis(pyrazolonato) lanthanide(III) anion. Because of the absence of an -hydrogen atom in the pyrazolonato ligands, and because of the saturation of the coordination sphere by four bidentate ligands, the luminescence properties are enhanced when compared to, e.g. quinolinate complexes. 相似文献

通过紫外-可见吸收光谱、二次谐波振荡和稳态荧光的测量,研究了半花菁(DAEP)Langmuir-Blodgett(LB)膜在不同制膜条件(压膜速度、半花菁和花生酸混合、亚相中加入碘离子)下,LB膜样品中半花菁分子的聚集体性质的改变和对非线性光学性能的影响.实验发现,纯半花菁LB膜中分子形成H-聚集体,从而导致吸收峰、荧光峰的蓝移和分子二阶非线性极化率β的减少.分子聚集程度的减少和分子二阶非线性系数β的增加可以通过增大压膜速度、半花菁和花生酸混合、亚相中加入碘离子等方法实现. 相似文献

A new highly sensitive and selective colorimetric method for fluoride determination in water is described. The novel reagent used is a hemicyanine dye (HC), which forms a stable complex with Zr-EDTA, the fluoride-binding site, through the hydroxyl groups. The hemicyanine-chelating Zr-EDTA complex (HC-Zr-EDTA) is ready to react with fluoride ion to release HC. This result in remarkable color change of the sensing solutions from red (λmax = 513 nm) to yellow (λmax = 427 nm) at pH 4.40. When applied to the colorimetric determination of fluoride ions, a linear range from 3.0 × 10−6 to 5.0 × 10−5 mol/L and a detection limit of 2.8 × 10−6 mol/L with a correlation coefficient of 0.9993 can be achieved under the optimized conditions. Because of the specific affinity of fluorides for the [Zr-EDTA], there is little interference by other ions. This method has been successfully applied to the determination of fluorides in toothpaste samples. 相似文献
