Hybrid quantum mechanical-molecular mechanics (dynamics) were performed on flavin reductase (Fre) and flavodoxin reductase (Fdr), both from Escherichia coli. Each was complexed with riboflavin (Rbf) or flavin mononucleotide (FMN). During 50 ps trajectories, the relative energies of the fluorescing state (S1) of the isoalloxazine ring and the lowest charge transfer state (CT) were assessed to aid prediction of fluorescence lifetimes that are shortened due to quenching by electron transfer from tyrosine. The simulations for the four cases display a wide range in CT–S1 energy gap caused by the presence of phosphate, other charged and polar residues, water, and by intermolecular separation between donor and acceptor. This suggests that the Gibbs energy change (ΔG0) and reorganization energy (λ) for the electron transfer may differ in different flavoproteins. 相似文献

本提出了用于测定色氨酸的土豆汁修饰碳糊电极,该电极选择性好,能有效地消除酪氨酸的干扰,在pH=7.4的NaH2PO4-NaOH生理缓冲液中,电极响应与色氨酸在9.8×10^-6~9.8×10^-5mol/L的浓度范围内呈线性关系,最低检出限是1.5×10^-6mol/L.并详细讨论了电极抗干扰能力与测定条件的关系。用该电极测定了实际样品氨基酸注射液中色氨酸的含量,结果与标准值吻合。 相似文献

研究了色氨酸和5-羟基色氨酸在玻璃碳电极上的电化学行为,研究了不同pH、静置时间、扫描速度以及表面活性剂等的影响,探讨了色氨酸和5-羟基色氨酸在玻璃碳电极上的氧化机理,建立了线扫伏安法同时测定色氨酸和5-羟基色氨酸的方法.实验发现,在含3.33×10-4 mol/L十二烷基磺酸钠的0.1mol/L柠檬酸(pH=4.50)介质中,5-羟基色氨酸和色氨酸分别在 0.675V和 1.070V产生一灵敏的氧化峰.对5-羟基色氨酸,相应的氧化峰的峰高与浓度在2.40×10-5 mol/L~8.00×10-4 mol/L范围内呈良好的线性关系,线性相关系数为0.9994;对色氨酸,其氧化峰的峰高与浓度在2.40×10-5 mol/L~6.40×10-4 mol/L浓度范围内呈良好的线性关系,线性相关系数为0.9929.该方法测量2.00×10-4 mol/L色氨酸和5-羟基色氨酸的相对标准偏差分别为3.2%和3.8%(n=10). 相似文献

Electrochemical behavior of cerium hexacyanoferrate (CeHCF) incorporated on multi-walled carbon nanotubes (MWNTs) modified GC electrode is investigated by scanning electron microscopy (SEM) and electrochemical techniques. The CeHCF/MWNT/GC electrode showed potent electrocatalytic activity toward the electrochemical oxidation of tryptophan in phosphate buffer solution (pH 7.0) with a diminution of the overpotential of 240 mV. The anodic peak currents increased linearly with the concentration of tryptophan in the range of 2.0 × 10−7 to 1.0 × 10−4 M with a detection limit of 2.0 × 10−8 M (at a S/N = 3). And the determination of tryptophan in pharmaceutical samples was satisfactory. 相似文献

In this study,Au nanoparticles/poly 5-[(2-hydroxynaphthalen-l-yl)diazenyl]isophthalic acid film modified glassy carbon electrode(AuNPs/poly(NDI)/GCE) has shown excellent electrocatalytic activity toward the oxidation of adrenaline(ADR),paracetamol(PAC),and tryptophan(Trp).The bare glassy carbon electrode(GCE) fails to separate the oxidation peak potentials of these molecules,while the poly(NDI) film modified electrode can resolve them.Electrochemical impedance spectroscopy(EIS)indicates that the charge transfer resistance of the bare electrode decreases as 5-[(2-hydroxynaphthalen-l-yl)diazenyl]isophthalic acid is electropolymerized on the bare electrode.Furthermore,EIS exhibits enhancement of electron transfer kinetics between analytes and the electrode after electrodeposition of Au nanoparticles.Differential pulse voltammetry results show that the electrocatalytic current increases linearly in the ranges of 0.01-680.0 μmol L~1 for ADR,0.05-498.0 μmol L~1 for PAC,and 3.0-632.0 μmol L~1 for Trp;with detection limits(S/N = 3) of 0.009 μmol L~1,0.005 μmol L~1,and 0.09 μmol L~1 for ADR,PAC,and Trp,respectively.The proposed method has been successfully applied for simultaneous determination of ADR,PAC,and Trp in biological samples. 相似文献

Polystyrene (PS) nanoparticles coated by BSA, hereafter denoted as PS/BSA, were prepared and chemically immobilized for the first time onto a capillary inner wall for open‐tubular CEC (OTCEC). EOF and scanning electron micrography were used to characterize the prepared nanoparticle‐coated capillaries. To investigate the performance of the prepared columns in OTCEC, chiral separation of d ,l ‐tryptophan (dl ‐Trp) was performed in monolayer BSA‐modified capillary and PS/BSA nanoparticle‐coated columns. The results indicated that the nanoparticle‐modified column afforded a higher resolution compared with the monolayer type. Rapid enantioseparation of dl ‐Trp (within 3 min) was achieved with the PS/BSA‐immobilized column using an electroosmotic pump‐assisted CEC. Enantiomer separations of other compounds like dl ‐tyrosine and warfarin were also achieved with the column. Besides, run‐to‐run and column‐to‐column repeatabilities of the PS/BSA‐coated column in the chiral separation were systematically introduced. 相似文献

Tryptophan is an essential amino acid, and understanding the conformational preferences of monomer and dimer is a subject of outstanding relevance in biological systems. An exhaustive first principles investigation of tryptophan ( W ) and its ionized counterparts cations (WC) , anions (WA) , and zwitterions (WZ) has been carried out. A comprehensive and systematic study of tryptophan dimer (WD) conformations resulted in about 62 distinct minima on the potential energy surface. The hydrogen bonds and a variety of noncovalent interactions such as OH‐π, NH‐π, CH‐π, CH‐O, and π‐π interactions stabilized different forms of tryptophan and its dimers. Over all in monomeric conformers which have NH‐O, hydrogen bonds showed higher stability than other conformers. A cursory analysis reveal that the most stable dimers stabilized by hydrogen bonding interactions while the less stable dimers showed aromatic side chain interactions. Protein Data Bank analysis of tryptophan dimers reveals that at a larger distance greater than 5 Å, T‐shaped orientations (CH‐π interactions) are more prevalent, while stacked orientations (π‐π interactions) are predominant at a smaller distance. © 2013 Wiley Periodicals, Inc. 相似文献

The effect of tryptophan on the membrane stability was studied by using three artificial biological membranes including liposome, Langmuir monolayer and solid supported bilayer lipid membrane (s-BLM) as models. All the results indicate that the penetration of tryptophan can destabilize different artificial biological membranes. The diameter of liposome and the leakage of calcein from liposome increased with the increase of tryptophan concentration because the penetration of tryptophan was beneficial for dehydrating the polar head groups of lipids and the formation of fusion intermediates. π-A isotherms of lecithin on the subphase of tryptophan solution further confirm that tryptophan can penetrate into lipid monolayer and reduce the stability of lipid monolayer. When the concentration of tryptophan increased from 0 to 2 × 10−3 mol L−1, the limiting molecular area of lecithin increased from 110.5 to 138.5 Å2, but the collapse pressure of the monolayer decreased from 47.6 to 42.3 mN m−1, indicating the destabilization of lipid monolayer caused by the penetration of tryptophan. The resistance spectra of s-BLM demonstrate that the existence of tryptophan leads to the formation of some defects in s-BLM and the destabilization of s-BLM. The values of electron-transfer resistance and double layer capacitance respectively decreased from 5.765 × 106 Ω and 3.573 × 10−8 F to 1.391 × 106 Ω and 3.340 × 10−8 F when the concentration of tryptophan increased from 0 to 2 × 10−3 mol L−1. 采用溶剂热法合成了发光性能和水溶性良好的YVO4∶Eu纳米探针(YVO4∶Eu NPs).由于YVO4∶Eu NPs的激发光谱与色氨酸的吸收光谱有很大程度的重叠,二者可发生荧光内滤效应,其中色氨酸为吸光体、YVO4∶Eu NPs为荧光体,YVO4∶Eu NPs的发光被猝灭.基于此,建立了基于YVO4∶Eu NPs内滤效应测定色氨酸含量的方法.对YVO4∶Eu NPs的加入量、反应溶液pH值和反应时间进行了优化,在最佳反应条件下,本方法测定色氨酸的线性范围为4.0×10-6~4.0×10-4 mol/L,检出限为1.0 ×10-6 mol/L(3σ).采用本方法测定了酱油中色氨酸的含量,回收率为95.2%和97.3%.本方法具有简便快速、灵敏准确的特点. 相似文献
