Summary An indirect polarographic method for the determination of phosmet was elaborated. It is based on the fact that N-hydroxymethylphthalimide formed by the reaction of phosmet with sulphuric acid is polarographically active and exhibits a behaviour during electrolysis at a Hg dropping-electrode similar to that of phthalimide. The method has been applied to the determination of phosmet in apples. The sensitivity is 0.1 ppm.

Zusammenfassung Das Verfahren beruht darauf, daß N-Hydroxymethylphthalimid, das bei der Reaktion von Phosmet mit Schwefelsäure gebildet wird, polarographisch aktiv ist und sich bei der Elektrolyse mit der Quecksilber-Tropfelektrode ähnlich wie Phthalimid verhält. Die Methode wurde zur Bestimmung von Phosmet in Äpfeln angewandt. Die Empfindlichkeit beträgt 0,1 ppm.

Raman spectroscopy combined with surface enhanced technology was adopted for analysis of phosmet pesticide. Continuous wavelet transforms (CWT) and successive projections algorithm (SPA) were used for Raman spectral preprocess and characteristic Raman shifts selection, respectively. Multi-linear regression (MLR) was used for spectral modeling. It is shown that enhanced chips can achieve enhanced Raman spectral signal for low concentration of pesticides. CWT can improve spectral resolution and smoothness, and remove translation error. Characteristic Raman shifts selection method of SPA can improve analytical precision, and simplify modeling variables of MLR CWT-SPA-MLR model can improve correlation coefficient (r) of prediction from 0. 823 to 0. 903, and reduce root mean square error of prediction (RMSEP) from 1. 640 to 1. 122. CWT-SPA-MLR method can be used for constructing analytical models for Raman spectra and has good interpretability and repeatability.  相似文献

In this study, a panel of haptens was synthesized for immunoconjugate preparation, and several haptens for heterologous tracer conjugates were also prepared. A highly sensitive polyclonal antibody against the organophosphorus insecticide phosmet was obtained and competitive direct enzyme-linked immunosorbent assays (cd-ELISA) for this pesticide were developed. In the cd-ELISA, the limit of detection (IC15) was 0.6 μg kg−1 and the sensitivity (IC50) was 20 μg kg−1. The suitability of the ELISA for pesticide quantification in peach, apple, orange juice, and apple juice was also studied. Good accuracy and precision were obtained with mean recoveries between 78% and 102.3% and mean coefficients of variation below 13.63%. Validation of the ELISA was conducted by high-performance liquid chromatography. The correlation between the data obtained using the microwell assay and the high-performance liquid chromatography was good (R 2 = 0.9849). The developed immunoassay methods were suitable for the rapid quantitative or qualitative determination of phosmet in food samples.  相似文献

拉曼光谱分析方法结合表面增强技术用于亚胺硫磷农药的检测。连续小波变换(continuous wavelet transforms, CWT)和连续投影算法(successive projections algorithm, SPA)分别用于拉曼光谱的预处理和特征拉曼位移的选择,多元线性回归(multi-linear regression, MLR)用于特征拉曼吸收的回归分析。结果表明,芯片增强可以实现低浓度农药溶液拉曼响应的增强;CWT可以提高拉曼光谱的分辨率和平滑度,消除光谱的平移误差;基于SPA方法的特征位移选择方法,既可以提高模型的分析精度,又可以简化MLR模型的回归变量;与特征增强波段的MLR模型比较,CWT-SPA-MLR模型可将验证集的预测相关系数(correlation coefficient,r)和预测均方根误差(root mean square error of prediction,RMSEP)由0.823和1.640改善为0.903和1.122。CWT-SPA-MLR可用于拉曼光谱定量解析模型的建立,该方法简单易用具有良好的重复性。  相似文献

An accurate method based on matrix solid-phase dispersion (MSPD), and gas chromatography-mass spectrometry (GC/MS) was developed for determination of phosmet residues and its metabolites (phosmet-oxon, phthalimide, N-hydroxymethylphthalimide, and phthalic acid) in olive fruits. After testing different sorbents and eluents for MSPD extraction, C18 and acetonitrile were found to be the most appropriate for clean-up of the samples, in terms of yields and efficient removal of interfering compounds. All analytes were determined in selective ion monitoring (SIM) mode following a derivatization step with N,O-bis-trimethylsilyltrifluoroacetamide (BSTFA) containing 1% of trimethylchlorosilane (TMCS), except for phosmet and phosmet-oxon which were analyzed directly. The method showed suitable linearity (correlation coefficients higher than 0.8919 for all the compounds) and suitable sensitivity (limit of detection lower than 0.06 mg/kg). It was successfully applied in the analysis of olive fruits collected during the preharvest interval and olive oil. Phosmet residues found in all samples were lower than the maximum residue limits established by legislation (2 mg/kg).  相似文献

A statistical approach for the analysis of complex samples by immunoassay is proposed in this article. Two enzyme-linked immunosorbent assays (ELISAs), one of them in the conjugate-coated format and the other in the antibody-coated format, were evaluated for their suitability to the analysis of mixtures of three organodithiophosphorus pesticides: azinphos-methyl, azinphos-ethyl and phosmet. It was found that the apparent affinity of the antibody to each analyte changed in the presence of a cross-reacting compound in the antibody-coated ELISA format, but not when the conjugate-coated ELISA format was used. The assays were thereafter applied to the analysis of mixtures of the three recognized pesticides. With the conjugate-coated ELISA format, accurate and precise determinations of mixtures could be performed if an azinphos-methyl standard curve was employed, with recoveries between 71% and 130% and with coefficients of variation lower than 12.7%. Neither accurate nor precise measurements could be accomplished with the enzyme immunoassay using the antibody-coated ELISA format, independently of the standard curve used. It is thought that the study presented here will have applicability in a variety of cases where the analytical goal is semiquantitative screening based on the total quantity of an unknown mixture of related compounds.  相似文献
