Based on the investigation of a non-ionic surfactant-salt-H2O liquid-liquid extraction system, general rules concerning salt selection are concluded and the mechanism of phase separation is explained. The extraction behavior of chlorpromazine hydrochloride (CPZ) and procaine hydrochloride (PCN) in such a system is studied. Research shows that the extraction efficiency of CPZ can amount to 96% by twice extraction, which means that quantitative extraction is realized, while that of PCN is 77%. This system produces distribution coefficients (KD) of 12.3 and 2.6, respectively, for CPZ and PCN. Extraction mechanism is deduced according to ultraviolet absorbance; and molecular fluorescence spectra change of the drugs in the system studied.  相似文献

Summary The side-chain conformations of psychoactive phenothiazine drugs in crystals are different from those of biologically inactive ring sulfoxide metabolites. This study examines the potential energies, molecular conformations and electrostatic potentials in chlorpromazine, levomepromazine (methotrimeprazine), their sulfoxide metabolites and methoxypromazine. The purpose of the study was to examine the significance of the different crystal conformations of active and inactive phenothiazine derivatives, and to determine why phenothiazine drugs lose most of their biological activity by sulfoxidation. Quantum mechanics and molecular mechanics calculations demonstrated that conformations with the side chain folded over the ring structure had lowest potential energy in vacuo, both in the drugs and in the sulfoxide metabolites. In the sulfoxides, side chain conformations corresponding to the crystal structure of chlorpromazine sulfoxide were characterized by stronger negative electrostatic potentials around the ring system than in the parent drugs. This may weaken the electrostatic interaction of sulfoxide metabolites with negatively charged domains in dopamine receptors, and cause the sulfoxides to be virtually inactive in dopamine receptor binding and related pharmacological tests.  相似文献

在pH 1.8~3.3的酸性介质中,金纳米微粒本身有一定的共振瑞利散射(RRS)强度,但盐酸氯丙嗪本身的RRS强度十分微弱,当二者共存时,溶液的RRS强度显著增强并出现新的RRS光谱,在280~368 nm之间产生强烈的散射带,其最大散射波长位于368 nm,并在284、440、498 nm处有明显的散射峰。在一定条件下,盐酸氯丙嗪在0~0.08 mg/L范围内与ΔIRRS强度成正比,方法具有较高的灵敏度,对盐酸氯丙嗪的检出限(3σ)达到1.75μg/L。本文考察了反应体系的RRS光谱特征,研究了适宜的反应条件、影响因素及分析化学性质,研究了共存物质的影响,表明方法具有较好的选择性,据此发展了一种用金纳米微粒作RRS探针测定盐酸氯丙嗪的新方法。  相似文献

Herein, we reported the fabrication of porous iron oxide/carbon black (P–Fe2O3/CB) composite through a two-step engineering method. At first, Prussian blue microcubes were used as a precursor and further calcined to form P–Fe2O3 microcubes. The intercalation of CB nanoparticles with P–Fe2O3 nanocubes was processed through the ultrasonication method. The obtained P–Fe2O3/CB were successfully scrutinized through various physiochemical characterization methods. The proposed P–Fe2O3/CB-modified glassy carbon electrode sensor was successfully implemented in the electrochemical sensing of chlorpromazine hydrochloride due to its very low charge transfer resistance (Rct) compared to the other electrode modifiers. The sensitive detection of CPMH through differential pulse voltammetry exemplifies an excellent electroanalytical performance such as a wide linear range of 0.5–1472 μM, a lower detection limit (0.001 μM), and an appraisable sensitivity of 1.99 μA/μM cm?2 due to its availability of a high number of active sites and its large surface area, respectively. It also expresses excellent selectivity, repeatability, reproducibility, and stability results. Moreover, the practical feasibility of the as-fabricated P–Fe2O3/CB/glassy carbon electrode sensor shows exquisite recovery (98.1–100.8%) results with an appraisable current response in various biological, pharmaceutical, and environmental samples.  相似文献

Summary.  The electrochemical behaviour of chlorpromazine has been examined in phosphate buffers in aqueous as well as micellar media at a pyrolytic graphite electrode surface. Two oxidation peaks were obtained in linear sweep voltammetry of chlorpromazine. The first peak corresponds to the formation of the cationic free radical, which on further 1e-oxidation gives a dication. The spectroscopic changes and kinetics of the cationic free radical and dication species generated during electrooxidation of chlorpromazine were investigated in both media. The decay of the dication was studied chronoamperometrically and was found to follow first-order kinetics with a half-life of ∼25 ms. Surfactants affect both E p and i p values. The anionic surfactant SDS has been found to catalyze the reaction of the free radical cation and the dication.  相似文献

制备了多壁碳纳米管修饰玻碳电极,采用循环伏安法(CV)研究了盐酸氯丙嗪在修饰电极上的电化学特性,发展了一种新的检测盐酸氯丙嗪的电化学分析方法。在最佳实验条件下,用循环伏安法检测盐酸氯丙嗪,其响应电流与盐酸氯丙嗪的浓度在8.0×10-5~1.0×10-3mol/L范围内有很好的线性关系,线性方程为Ip(A)=0.0106c(mol/L)-8×10-8(R2=0.999,n=6),检出限为6.2×10-6mol/L(S/N=3)。方法可用于盐酸氯丙嗪片的测定。  相似文献

A new post-chemiluminescence (PCL) phenomenon was observed when phenothiazine medications were injected into the reaction mixture after the chemiluminescence (CL) reaction of luminol and potassium ferricyanide had finished. A possible reaction mechanism was proposed based on studies of the kinetic characteristics of the CL, CL spectra, fluorescence spectra, and on other experiments. The feasibility of determining various phenothiazine medications by utilizing these PCL reactions was examined. A molecular imprinting–post-chemiluminescence (MI-PCL) method was established for the determination of chlorpromazine hydrochloride using a chlorpromazine hydrochloride-imprinted polymer (MIP) as the recognition material. The method displayed high selectivity and high sensitivity. The linear range of the method was 1.0×10−8∼1.0×10−6, with a linear correlation coefficient of 0.9985. The detection limit was 3×10−9 g/ml chlorpromazine hydrochloride, and the relative standard deviation for a 1.0×10−7 g/ml chlorpromazine hydrochloride solution was 4.0% (n=11). The method has been applied to the determination of chlorpromazine hydrochloride in urine and animal drinking water with satisfactory results.   相似文献

The effect of electrolytes on the micellar behavior of an amphiphilic drug,chlorpromazine(CPZ)hydroehloride,was studied using cloud point(CP)and dye solubilization techniques.In the presence of KBr,increase because of deprotonation of drug molecules at high pH. The visible absorbance increased(due to dye solubilization)with the increase in pH from 6.5 to 6.9,which indicated micellar growth.At fixed pH(6.7),addition of inorganic salts(KF,KC1,effecfiveness being in the order:F-Na >K ,which Was explained by considering cognizance of their hydrated radii.Compared with anions,their effect was small.Increase in[CPZ]caused micellar growth and hence the CP as well as the visible absorbance increased.The overall behavior Was discussed in terms of electrostatic interactions and micellar growth.  相似文献

在pH 4.04的HAc-NaAc缓冲液中, 刚果红与盐酸氯丙嗪在室温下迅速结合生成缔合物, 且缔合物在480 nm处有最大吸收. 基于此建立了流动注射光度法测定药物中盐酸氯丙嗪的含量. 方法线性范围为0.25~50.0 μg/mL, 检出限为0.082 μg/mL, 测定频率达80次/h.  相似文献
