3-Chloroquinoline-2,4-diones react with cyanide ions in dimethyl formamide to give 3-cyanoquinoline-2,4-diones in small yields due to the strong hindrance of the substituent at the C-3 atom. Good yields can be achieved if the substituent at this position is the methyl group. In the methanol solution, the reaction proceeds by an addition mechanism to form 2-oxo-1a,2,3,7b-tetrahydrooxireno[2,3-c]quinoline-7b-carbonitriles, from which 4-hydroxy-3-methoxy-2-oxo-1,2,3,4-tetrahydroquinoline-4-carbonitriles are subsequently formed by opening of the epoxide ring with methanol. Some minor products of these reactions have also been isolated. The 1H, 13C and 15N NMR spectra of the prepared compounds were measured, and all resonances were assigned using appropriate two-dimensional spectra.  相似文献

3‐Hydroxyquinoline‐2,4‐diones react with KSCN in the presence of the NH$\rm{{_{4}^{+}}}$ ions to generate 2,3‐dihydro‐3‐thioxoimidazo[1,5‐c]quinazolin‐5(6H)‐ones, 2,3‐dihydro‐2‐thioxo‐1H‐imidazo[4,5‐c]quinolin‐4(5H)‐ones, and products of molecular rearrangement of the 3‐aminoquinolinedione intermediates. Starting compounds with a benzyl (Bn) group at C(3) afford 3‐aminoquinolinediones, even when only AcONH4 is used. The results of the reaction between 3‐hydroxyquinoline‐2,4‐diones and KSCN in the presence of BuNH2 show that replacing a OH group with a secondary NH2 group is also possible.  相似文献

The reaction of tertiary α-chloroketones with ethanolamine has not been hitherto described in the literature. Herein, we describe the reaction of tertiary 3-chloroquinoline-2,4-diones with ethanolamine to give novel 3-(2-hydroxyethylamino)quinoline-2,4-diones. These compounds provide 3-(2-oxooxazolidin-3-yl)quinoline-2,4(1H,3H)-diones and new compounds with dimeric character after reaction with triphosgene. Molecular rearrangement proceeds during the reaction of 3-(2-hydroxyethylamino)quinoline-2,4-diones with isocyanic acid. Three types of reaction products arise: 2-(2-hydroxyethyl)imidazo[1,5-c]quinazoline-3,5-diones, 3-(2-hydroxyethyl)-3,3a-dihydro-2H-imidazo[4,5-]quinoline-4(5H)diones and primarily 5-hydroxy-1-(hydroxyethyl)-1′H-spiro[imidazolidine-5,3′-indole]-2,2′-diones. The reaction mechanism and product stereochemistry are discussed. The 1H, 13C and 15N NMR spectra of the prepared compounds were measured, and all resonances were assigned from appropriate two-dimensional experiments.  相似文献

3‐Alkyl/aryl‐3‐hydroxyquinoline‐2,4‐diones were reduced with NaBH4 to give cis‐3‐alkyl/aryl‐3,4‐dihydro‐3,4‐dihydroxyquinolin‐2(1H)‐ones. These compounds were subjected to pinacol rearrangement by treatment with concentrated H2SO4, resulting in 4‐alkyl/aryl‐3‐hydroxyquinolin‐2(1H)‐ones. When a benzyl (Bn) group was present in position 3 of the starting compound, its elimination occurred during the rearrangement, and the corresponding 3‐hydroxyquinolin‐2(1H)‐one was formed. The reaction mechanisms are discussed for all transformations. All compounds were characterized by IR, 1H‐ and 13C‐NMR spectroscopy, as well as mass spectrometry.  相似文献

3‐Chloroquinoline‐2,4‐diones do not react with phosphoryl chloride, however, 2,4‐dichloroquinolines and/or 4‐chloroquinolin‐2‐ones are formed in the presence of N,N‐dimethylaniline. Along with these compounds, small quantities of novel dihydrogen phosphates of 4‐hydroxyquinolin‐2‐ones were isolated. We outline a simple procedure that allows for the preparation of these compounds in moderate to good yields. All compounds were characterized by 1H and 13C NMR, IR, EI‐MS, and ESI‐MS spectroscopy, and in select cases by 31P NMR spectroscopy.  相似文献

Perimidine-based chelators 1 and 2 were prepared, and their structures were confirmed by 1H and 13C NMR, MS spectroscopy and elemental analysis. These compounds were studied as specific synthetic receptors for the recognition of transition metal ions. They exhibited high affinity and selectivity towards Cu(II) ions. The conditional binding constants, linear dynamic range and detection limit were determined by UV–vis spectroscopy. These parameters demonstrated high potential of the prepared synthetic receptors for the recognition and determination of Cu(II) ions. The minimum detectable concentrations of Cu(II) ions for the synthetic receptors 1 and 2 were 270 and 75 nM (R 2 = 0.9915 and 0.9964) in aqueous medium (water/DMSO; 99:1 (v/v)), respectively.  相似文献

Highly functionalized fluorinated porphyrins were synthesized by a convergent strategy. Nucleophilic substitution using fluorinated branched unit and 5,10,15,20-tetrakis(pentafluorophenyl)porphyrin afforded highly functionalized fluorinated porphyrin 8 containing 24 fluorinated chains in the peripheral region.  相似文献

Summary The aim of our study was to monitor Cyclosporine A (CA) during a long-term period and to determine whether the whole-blood levels, estimated by HPLC and radio-immunoassay (RIA), provide a relevant indicator of the toxic effect of CA, of the interference of endogenous substances with CA and of interactions of erythromycin and ketoconazole with CA metabolims. The ratios of CA concentrations, estimated by RIA and by HPLC were calculated in order to analyse changes in CA metabolism. A decrease in this ratio was observed in these patients during periods of erythromycin and ketoconazole therapy deteriorating hepatic function. We observed unusual higher CA concentrations measured by HPLC compared with RIA in heart transplant patient with marked renal insiffuciency after one-year follow-up period of treatment of CA. The comparison of CA whole-blood levels estimated using HPLC and RIA makes it possible to analyse the individual biotransformation of CA to its metabolites, and interference of endogenous substance in special clinical situations.  相似文献
