Exosomes are nanovesicles secreted by most cellular types that carry important biochemical compounds throughout the body with different purposes, playing a preponderant role in cellular communication. Because of their structure, physicochemical properties and stability, recent studies are focusing in their use as nanocarriers for different therapeutic compounds for the treatment of different diseases ranging from cancer to Parkinson's disease. However, current bioseparation protocols and methodologies are selected based on the final exosome application or intended use and present both advantages and disadvantages when compared among them. In this context, this review aims to present the most important technologies available for exosome isolation while discussing their advantages and disadvantages and the possibilities of being combined with other strategies. This is critical since the development of novel exosome‐based therapeutic strategies will be constrained to the effectiveness and yield of the selected downstream purification methodologies for which a thorough understanding of the available technological resources is needed.  相似文献

In this article, we present a higher‐order finite volume method with a ‘Modified Implicit Pressure Explicit Saturation’ (MIMPES) formulation to model the 2D incompressible and immiscible two‐phase flow of oil and water in heterogeneous and anisotropic porous media. We used a median‐dual vertex‐centered finite volume method with an edge‐based data structure to discretize both, the elliptic pressure and the hyperbolic saturation equations. In the classical IMPES approach, first, the pressure equation is solved implicitly from an initial saturation distribution; then, the velocity field is computed explicitly from the pressure field, and finally, the saturation equation is solved explicitly. This saturation field is then used to re‐compute the pressure field, and the process follows until the end of the simulation is reached. Because of the explicit solution of the saturation equation, severe time restrictions are imposed on the simulation. In order to circumvent this problem, an edge‐based implementation of the MIMPES method of Hurtado and co‐workers was developed. In the MIMPES approach, the pressure equation is solved, and the velocity field is computed less frequently than the saturation field, using the fact that, usually, the velocity field varies slowly throughout the simulation. The solution of the pressure equation is performed using a modification of Crumpton's two‐step approach, which was designed to handle material discontinuity properly. The saturation equation is solved explicitly using an edge‐based implementation of a modified second‐order monotonic upstream scheme for conservation laws type method. The combination of 2D materials opens a wide range of possibilities to create new-generation structures with multiple applications. Covalently cross-linked approaches are a ground-breaking strategy for the formation of homo or heterostructures made by design. However, the covalent assembly of transition metal dichalcogenides flakes is relatively underexplored. Here, a simple covalent cross-linking method to build 2H-MoS2–MoS2 homostructures is described, using commercially available bismaleimides. These assemblies are mainly connected vertically, basal plane to basal plane, creating specific molecular sized spaces between MoS2 sheets. Therefore, this straightforward approach gives access to the controlled connection of sulfide-based 2D materials.  相似文献

Atom transfer radical polymerization (ATRP) has been utilized to synthesize tri‐ and star‐block copolymers of poly(di(ethylene glycol)methyl ether methacrylate) (PDEGMA) and quaternized poly(2‐(dimethylamino)ethyl methacrylate) (qPDMAEMA). The block copolymers, all with a minimum of two cationically charged blocks, were sequentially used for electrostatic macrocrosslinking of a dilute dispersion of anionic TEMPO‐oxidized cellulose nanofibrils (CNF, 0.3 wt%), forming free‐standing hydrogels. The cationic block copolymers adsorbed irreversibly to the CNF, enabling the formation of ionically crosslinked hydrogels, with a storage modulus of up to 2.9 kPa. The ability of the block copolymers to adsorb to CNF was confirmed by quartz crystal microbalance with dissipation monitoring (QCM‐D) and infrared spectroscopy (FT‐IR), and the thermoresponsive properties of the hydrogels were investigated by rheological stress and frequency sweep, and gravimetric measurements. Bioconversion of l-phenylalanine to 2-phenylethanol using Saccharomyces cerevisiae is connected with the growth of biomass strongly limited by product inhibition. Therefore, fermentation can proceed only at low conversions of l-phenylalanine with very low yield of the desired product, which allows reaching the maximum concentration of 2-phenylethanol, 4 g L?1, in an ordinary batch, fed-batch, or chemostat bioreactor. To minimize capital and operating costs in the bioproduction of chemical specialties where the product inhibits the bioreaction, using a hybrid system based on the application of membrane extraction integrated in the bioreactor to remove the product is a suitable solution. Integration can be done by an external module for membrane extraction or, as a more efficient solution, by an extraction membrane module immersed directly in the bioreactor. Such a hybrid system can be used to remove 2-phenylethanol from the fermentation media and thus to overcome the product inhibition of the biotransformation process. In this paper, a hybrid system consisting of a stirred tank bioreactor (3.5 L) and an immersed extraction hollow fiber membrane module was studied. In the proposed system, the kinetics of 2-phenylethanol extraction from a water solution with and without biomass in the bioreactor to alkanes at different operational conditions was measured. Extraction kinetics was compared with the predictions obtained by a mathematical model. In the hybrid system, two extractive biotransformation experiments were performed and compared with that without product removal. Experimental data were also mathematically predicted with good accuracy between the simulation and the experiment.  相似文献

We confront a practical cutting stock problem from a production plant of plastic rolls. The problem is a variant of the well-known one dimensional cutting stock, with particular constraints and optimization criteria defined by the experts of the company. We start by giving a problem formulation in which optimization criteria have been considered in linear hierarchy according to expert preferences, and then propose a heuristic solution based on a GRASP algorithm. The generation phase of this algorithm solves a simplified version which is rather similar to the conventional one dimensional cutting stock. To do that, we propose a Sequential Heuristic Randomized Procedure (SHRP). Then in the repairing phase, the solution of the simplified problem is transformed into a solution to the real problem. For experimental study we have chosen a set of problem instances of com-mon use to compare SHRP with another recent approach. Also, we show by means of examples, how our approach works over instances taken from the real production process. The work considers an optimal design problem in the context of nonlinear elastica. More specifically, we deal with finding the best way of mixing fixed amounts of two different elastic materials, so as to minimize the tip deflection of a cantilever beam loaded on its free extreme under the assumption of large deflections. Applying an optimality criteria method to the relaxed problem, simulations give us numerical evidence that the original design problem admits classical solutions (i.e. there is no microstructure) and those are the same as the respective ones for the case of small deflections.  相似文献
