Experiments demonstrate that Mg(2+) is crucial for structure and function of RNA systems, yet the detailed molecular mechanism of Mg(2+) action on RNA is not well understood. We investigate the interplay between RNA and Mg(2+) at atomic resolution through ten 2-μs explicit solvent molecular dynamics simulations of the SAM-I riboswitch with varying ion concentrations. The structure, including three stemloops, is very stable on this time scale. Simulations reveal that outer-sphere coordinated Mg(2+) ions fluctuate on the same time scale as the RNA, and that their dynamics couple. Locally, Mg(2+) association affects RNA conformation through tertiary bridging interactions; globally, increasing Mg(2+) concentration slows RNA fluctuations. Outer-sphere Mg(2+) ions responsible for these effects account for 80% of Mg(2+) in our simulations. These ions are transiently bound to the RNA, maintaining interactions, but shuttled from site to site. Outer-sphere Mg(2+) are separated from the RNA by a single hydration shell, occupying a thin layer 3-5 ? from the RNA. Distribution functions reveal that outer-sphere Mg(2+) are positioned by electronegative atoms, hydration layers, and a preference for the major groove. Diffusion analysis suggests transient outer-sphere Mg(2+) dynamics are glassy. Since outer-sphere Mg(2+) ions account for most of the Mg(2+) in our simulations, these ions may change the paradigm of Mg(2+)-RNA interactions. Rather than a few inner-sphere ions anchoring the RNA structure surrounded by a continuum of diffuse ions, we observe a layer of outer-sphere coordinated Mg(2+) that is transiently bound but strongly coupled to the RNA. 相似文献

A dual-wavelength image-plane digital holography, combined with a windowed Fourier analysis, is presented for dynamic measurement of a vibrating object. In order to expand the range of the velocity measurement, the object is simultaneously illuminated by two lasers with different wavelengths. A sequence of digital holograms of a vibrating object is captured by a CCD camera and two wrapped phase maps are retrieved from each digital hologram. At each instant, a new phase distribution with a synthetic wavelength is obtained by subtracting these two wrapped phase maps. A windowed Fourier analysis is then applied spatially and temporally to retrieve the high-precision displacement and velocity of the tested object. Experimental results show the requirement on the camera capture frequency is reduced by the proposed method. 相似文献

In speckle-based metrology systems, a finite range of possible motion or deformation can be measured. When coherent imaging systems with a single limiting aperture are used in speckle metrology, the observed decorrelation effects that ultimately define this range are described by the well-known Yamaguchi correlation factor. We extend this result to all coherent quadratic phase paraxial optical systems with a single aperture and provide experimental results to support our theoretical conclusions. 相似文献

It is shown that both surface tilting and translational motion can be independently estimated by use of the speckle photographic technique by capturing consecutive images in two different fractional Fourier domains. A geometric interpretation, based on use of the Wigner distribution function, is presented to describe this application of the optical fractional Fourier transform when little prior information is known about the motion. 相似文献

This paper investigates the application of Fresnel based numerical algorithms for the reconstruction of Gabor in-line holograms. We focus on the two most widely used Fresnel approximation algorithms, the direct method and the angular spectrum method. Both algorithms involve calculating a Fresnel integral, but they accomplish it in fundamentally different ways. The algorithms perform differently for different physical parameters such as distance, CCD pixel size, and so on. We investigate the constraints for the algorithms when applied to in-line Gabor digital holographic microscopy. We show why the algorithms fail in some instances and how to alter them in order to obtain useful images of the microscopic specimen. We verify the altered algorithms using an optically captured digital hologram. 相似文献

The relationship between optical fractional Fourier transforms (OFRTs) obtained at different wavelengths is derived by use of the ABCD matrix formalism. It is shown that varying the wavelength while retaining the same optical system can be used to control the order of the OFRT. The advantage of this method of varying OFRT order is that no variation in the characteristics of the bulk optics is required. A general experimental verification of the theory is provided by showing the exact equivalence of two OFRT systems of different order when they are replayed using the same input function at different wavelengths. 相似文献

A novel technique for multiplexing complex images is proposed in which each image may be demultiplexed only if a set of random encryption keys is known. The technique utilizes the ability of the double random phase encoding method to spread a signals' energy in both the space and the spatial frequency domains in a controlled manner. To multiplex, images are independently encrypted with different phase keys and then superimposed by recording sequentially on the same material. Each image is extracted by using the particular key associated with it. During decryption the energy from the other images is further spread, making it possible to minimize its effects by using suitable filters. Wigner analysis is applied to the technique, and numerical results are presented. 相似文献

A number of methods have recently been proposed in the literature for the encryption of two-dimensional information by use of optical systems based on the fractional Fourier transform. Typically, these methods require random phase screen keys for decrypting the data, which must be stored at the receiver and must be carefully aligned with the received encrypted data. A new technique based on a random shifting, or jigsaw, algorithm is proposed. This method does not require the use of phase keys. The image is encrypted by juxtaposition of sections of the image in fractional Fourier domains. The new method has been compared with existing methods and shows comparable or superior robustness to blind decryption. Optical implementation is discussed, and the sensitivity of the various encryption keys to blind decryption is examined. 相似文献
