建立了HPLC法测定多西他赛中间体(1)及其光学异构体(2)。色谱柱采用Lux Cellulose-2手性柱(250 mm×4.6 mm, 5μm),以0.02%三氟乙酸溶液/甲醇(15∶85)为流动相,等度洗脱。检测波长为210 nm,柱温为25℃,流速为0.4 mL/min,进样量为5μL。结果显示,1和2分别在0.1~0.8 mg/mL和0.35~8.73μg/mL内线性关系良好,检测限分别为0.12、0.14μg/mL。低、中、高浓度的平均回收率(n=3)分别为100.8%、100.6%、101.3%,RSD分别为0.7%、0.5%、0.7%;精密度RSD(n=6)为0.6%。建立的方法灵敏度高、操作简单,可用于1中2的测定,也为多西他赛合成过程中的质量控制提供了参考。  相似文献

采用仿生合成法制备麦角硫因。以L-组氨酸(2)为起始原料,经还原胺化、甲基化得直接生物前体L-组氨酸甜菜碱(4); 4与L-半胱氨酸、3-巯基丙酸经“一锅法”合成麦角硫因(1),总收率46.95%,其结构经1H NMR, 13C NMR和MS(ESI)谱确证,纯度为99.56%(HPLC法)。该路线步骤简短、原料价廉易得、反应条件温和、操作简便、收率较高,适用于百克级麦角硫因的制备。  相似文献

Photochromic spiropyran with a long chain alkyl substitute can form axial complexes with α-, β-, and γ-cyclodextrin, respectively. The complexes show normal photochromism. The novel property of the colored forms of the inclusion complexes is that they can assemble into dimers at relatively low concentration or J-aggregates at relatively high concentration. For α-, β-, and γ-cyclodextrin, λ_(max) of the J-aggregates appear at 700 650, and 630 nm, respectively. The sizes of the cavities of cyclodextrins have very little effect on the spectra and decoloration kinetics of the dimers, but have great effects on the spectra of the J-aggregates. Unlike the charge transfer complex of Krongauz, the decoloration process of the dimers or J-aggregates cannot be described by an exponential or a two-exponential kinetics, but obey half-order kinetics very well. The verification and calculation of the negative refractive index of a meta-material is carried out by the finitedifference time-domain method. A slab and a prism of the meta-material are simulated. A genuine plane wave is generated by a two-direction periodic boundary condition (PBC) in the slab model. Based on an advanced phase extraction technique, the negative refractive index of the meta-material is verified by phase velocity measurement in the slab and prism measurement. From our results, not only the phenomenon of backward phase propagation but also the negative refraction is clearly observed. The index is also calculated precisely. The results from the two models are consistent.  相似文献

Wedge-shaped left-handed materials (LHMs) with split ring resonators and wires structures are fabricated by photolithography and lift-off techniques, and the variation of left-handed frequency induced by substrates with different dielectric parameters is investigated. The Snell refraction experiments of the LHM samples are carried out on an angular resoled microwave spectrometer, and the results indicate that the left-handed frequencies of the LHMs shifted downward from 10.57 GHz to 9.74 GHz when the dielectric parameters of the LHM substrates increase from 3.7 to 4.8. Moreover, the finite difference time domain method is used to simulate the microwave transmission properties of the left-handed materials with different substrates, and the experimental results are in agreement with the numerical simulation results. In addition, the reason for the shifting of the left-handed frequency of the LHMs is discussed theoretically.  相似文献

以2-(3-氰基-4-羟基苯基)-4-甲基-1,3-噻唑-5-羧酸乙酯为原料,经水解得到中间体10;以二甲基膦乙酸苄酯为原料,经Witting反应、氢化还原双键、环合及钯/碳催化氢化脱苄基四步反应,得到中间体BODIPY-FL;10与BODIPY-FL缩合得到非布司他荧光药物探针分子,其结构经1H NMR,MS(ESI) 表征。体外生物活性测试结果显示:该荧光探针的IC50为8.3nM,说明接入荧光基团后探针分子与黄嘌呤氧化酶(XOD)的亲和力略有下降。   相似文献
