Although macromolecules on cell surfaces are predominantly targeted and drugged with antibodies, they harbor pockets that are only accessible to small molecules and constitutes a rich subset of binding sites with immense potential diagnostic and therapeutic utility. Compared to antibodies, however, small molecules are disadvantaged by a less confined biodistribution, shorter circulatory half-life, and inability to communicate with the immune system. Presented herein is a method that endows small molecules with the ability to recruit and activate chimeric antigen receptor T cells (CAR-Ts). It is based on a CAR-T platform that uses a chemically programmed antibody fragment (cp-Fab) as on/off switch. In proof-of-concept studies, this cp-Fab/CAR-T system targeting folate binding proteins on the cell surface mediated potent and specific eradication of folate-receptor-expressing cancer cells in vitro and in vivo.  相似文献

We describe a simple method for tracking the course of microbial degradation of polysaccharide-rich feedstocks. The method involves determining total polysaccharides present in the feedstock, measured in glucose equivalents, relative to the fractional component of polysaccharides exhibiting 2,3-dinitrosalycylic acid aldehyde activity. The ratio of total polysaccharide to aldehyde activity, defined as the end-point fragmentation (EPF) index, is then calculated and tracked as it shifts as microbial degradation of polysaccharide-rich feedstock progresses. While degradation occurs, the EPF index falls. It bottoms out at an asymptotic limit marking the point in time where further degradation of the polysaccharide-rich feedstock has ceased. The EPF index can be used to follow the progressive breakdown of composting polysaccharide-rich waste. It may also have applicability as a means of tracking the turnover of polysaccharides in other complex environments including soil, sediments, wetlands, and peat bogs.  相似文献

A mild and improved method for the synthesis of thioethers has been developed. In the presence of cesium carbonate, tetrabutylammonium iodide, and DMF, various alkyl and aryl thiols underwent S-alkylation to afford structurally diverse sulfides in high yield. Unprotected mercaptoalcohols and thioamines reacted chemoselectively at the sulfur moiety exclusively. An example of a one-pot, solid-phase synthesis of a thioether is also described.  相似文献

Oligomers of arginine, such as octa-d-arginine amide, are excellent transporters for active drugs through cell membranes and tissue. The synthesis of octa-d-arginine amide, as the nonahydrochloride salt, was approached via a solution phase synthetic route involving the preparation of an octa-d-ornithine intermediate, which was then converted into the desired octa-d-arginine compound through a guanidinylation step. The multi-step synthesis was carried out at pilot scale, resulting in the preparation of 700 g of the target molecule. No chromatographic purification was needed at any step of the process.  相似文献

The surfaces of poly(methyl methacrylate) (PMMA) films modified by O2H2O and H2O radio-frequency glow discharge plasmas were studied using electron spectroscopy for chemical analysis (ESCA or XPS), low energy ion scattering (LEISS or ISS), Fourier transform IR spectroscopy (FTIR) with attenuated total reflectance (ATR) sampling, and critical surface energy from contact angle measurements. The extent and nature of modification with respect to promotion of a hydrophilic surface compared to the hydrophobic surface of the unmodified PMMA has been probed. Results show drastic decreases in C/O ratio at the near surface, which increases to that of the unmodified PMMA as deeper cross sections are analyzed. In addition peak fitting of ESCA data correlated with FTIR functional group information allows for the qualitative and quantitative analysis of the resulting bonding and structure of the modified layer. From these results combined with the polarity and surface energy differences obtained from contact angle measurements, the structural changes are discussed with respect to plasma reaction mechanisms and differences in the structure of the modified polymer films.  相似文献

The geometries, energies and vibrational frequencies of various polyborates in both gaseous and aqueous phase were calculated at the B3LYP/aug-cc-pVDZ level. The calculated total symmetrical stretching Raman shifts of B(OH)(3), B(OH)(4)(-), B(2)O(OH)(4), B(2)O(OH)(5)(-), B(2)O(OH)(6)(2-), B(3)O(3)(OH)(3), B(3)O(3)(OH)(4)(-), B(3)O(3)(OH)(5)(2-), B(3)O(3)(OH)(6)(3-), B(4)O(5)(OH)(4)(2-) and B(5)O(6)(OH)(4)(-) were assigned to 877.40, 735.33, 785.22, 792.90, 696.79, 587.72, 599.06, 740.16, 705.01, 551.67 and 521.04cm(-1), respectively. The results can be used as the characteristic frequency for polyborates in aqueous phase at room temperature. At least six types of polyborates B(OH)(3), B(OH)(4)(-), B(3)O(3)(OH)(4)(-), B(3)O(3)(OH)(5)(2-), B(4)O(5)(OH)(4)(2-) and B(5)O(6)(OH)(4)(-), occur in aqueous solutions at ambient temperature. The chemical species distribution and the relevant interaction mechanisms among polyborates in the solutions were also suggested.  相似文献

Results from infrared photodissociation (IRPD) spectroscopy and kinetics of singly hydrated, protonated proline indicate that the water molecule hydrogen bonds preferentially to the formally neutral carboxylic acid at low temperatures and at higher temperatures to the protonated N-terminus, which bears the formal charge. Hydration isomer populations obtained from IRPD kinetic data as a function of temperature are used to generate a van't Hoff plot that reveals that C-terminal binding is enthalpically favored by 4.2-6.4 kJ/mol, whereas N-terminal binding is entropically favored by 31-43 J/(mol K), consistent with a higher calculated barrier for water molecule rotation at the C-terminus.  相似文献

Recent work in our laboratories has fully characterized the surface region of a segmented poly(ether-urethane) (PEU) extending from the air/polymer interfacial region through bulk depths in the micron range. This characterization utilized energy and angle dependent Electron Spectroscopy for Chemical Analysis (ESCA), Attenuated Total Reflectance–Fourier Transform Infrared Spectroscopy (ATR–FTIR), and Comprehensive Wettability Profiling (contact angle using a homologous series of liquids) as defined by Zisman. In this study this same multi-analytical-technique approach is used to elucidate changes in these PEU surfaces induced through an H2O Radio Frequency Glow Discharge (RFGD) plasma. This investigation reports both qualitative and quantitative changes due to the modification treatments as well as the permanency of the changes effected on these surfaces through the plasma treatment. From our analyses, the amount of surface residing polyurethane (hard segment) is observed to increase due to a proposed plasma etching mechanism. Further, the addition of oxygen containing functionality is detected at the modified surfaces unique with respect to the unmodified PEU. These surface modifications which show large increases in wettability, are finally observed to be semi-permanent over a time period of 6 months.  相似文献
