Polyphenolic and Terpenoids are potent natural antiparasitic compounds. This study aimed to identify new drug against Leishmania parasites, leishmaniasis’s causal agent. A new in silico analysis was accomplished using molecular docking, with the Autodock vina program, to find the binding affinity of two important phytochemical compounds, Masticadienonic acid and the 3-Methoxycarpachromene, towards the trypanothione reductase as target drugs, responsible for the defense mechanism against oxidative stress and virulence of these parasites. There were exciting and new positive results: the molecular docking results show as elective binding profile for ligands inside the active site of this crucial enzyme. The ADMET study suggests that the 3-Methoxycarpachromene has the highest probability of human intestinal absorption. Through this work, 3-Methoxycarpachromene and Masticadienonic acid are shown to be potentially significant in drug discovery, especially in treating leishmaniasis. Hence, drug development should be completed with promising results.  相似文献

The vertical profile of Saharan dust in the atmosphere is generally characterized by a large aerosol concentration in the mid troposphere, differently from the climatological distribution of other types of particles, that show a peak at the surface and a rapid decrease with height. Saharan dust is also characterized by particles of relatively large size of irregular shape, and variable values of the single scattering albedo (the ratio between radiation scattering and extinction). The dust's peculiar vertical distribution is expected to produce an effect on the calculation of the direct aerosol radiative forcing at the surface and at the top of the atmosphere. This effect is investigated by comparing estimates of aerosol direct visible radiative forcing at the surface and at the top of the atmosphere for dust vertical profiles measured in the Mediterranean, and for the climatological profile. The radiative forcing is estimated by means of an accurate radiative transfer model, and for the ocean surface. The sensitivity of the results on the solar zenith angle, aerosol optical depth, and aerosol absorption is also investigated. The aerosol radiative forcing at the surface shows a very small dependency on the aerosol vertical profile. At the top of the atmosphere, the radiative forcing is weakly dependent on the vertical profile (up to 10% variation on the daily average forcing) for low absorbing particles; conversely, it shows a strong dependency (the daily radiative forcing may vary up to 100%) for absorbing particles. The top of the atmosphere visible radiative forcing efficiency produced by dust having single scattering albedo <0.7 is higher by 4 W m−2 when the observed vertical profile instead of the standard profile is used in the calculations (i.e. it produces a lower cooling). For values of the single scattering albedo around 0.67, the sign of the forcing depends on the vertical profile. The influence of the vertical distribution on the radiative forcing is largest at small values of the solar zenith angle, and at short wavelengths.  相似文献

This article presents the development of a sensor made from a gold electrode and a receiving polymeric membrane based on tannin extracted from the skin of Punica granatum. L (pomegranate) for real-time detection of heavy metals in a hydrous environment. The basic principle of this device is the complexing (chelating) of metal ions through the adjacent hydroxyl groups contained in the chemical structures of the tannins. The electrochemical characterisation was performed by using electrochemical impedance spectroscopy and square wave voltammetry. Other morphological and structural analyses were performed by using Fourier transform infrared spectroscopy and atomic-force microscopy. The results obtained showed the high sensitivity of the developed device (detection limits of 6.35 × 10?9 g L?1 for Cu2+, 1.1 × 10?8 g L?1 for Cd2+ and 2 × 10?7 g L?1 for Pb2+) and the possibility of simultaneously detecting several heavy metals, each one in a highly selective manner with highly acceptable response time (48s).  相似文献

Digital total variation filtering is analyzed as a fast, robust, post-processing method for accelerating the convergence of pseudospectral approximations that have been contaminated by Gibbs oscillations. The method, which originated in image processing, can be combined with spectral filters to quickly post-process large data sets with sharp resolution of discontinuities and with exponential accuracy away from the discontinuities.  相似文献

Studying neurite guidance by diffusible or substrate bound gradients is challenging with current techniques. In this study, we present the design, fabrication and utility of a microfluidic device to study neurite guidance under chemogradients. Experimental and computational studies demonstrated the establishment of a steep gradient of guidance cue within 30 min and stable for up to 48 h. The gradient was found to be insensitive to external perturbations such as media change and movement of device. The effects of netrin-1 (0.1-10 μg mL(-1)) and brain pulp (0.1 μL mL(-1)) were evaluated for their chemoattractive potential on neurite turning, while slit-2 (62.5 or 250 ng mL(-1)) was studied for its chemorepellant properties. Hippocampal or dorsal root ganglion (DRG) neurons were seeded into a micro-channel and packed onto the surface of a 3D collagen gel. Neurites grew into the matrix in three dimensions, and a gradient of guidance cue was created orthogonal to the direction of neurite growth to impact guidance. The average turning angle of each neurite was measured and averaged across multiple devices cultured under similar conditions to quantify the effect of guidance cue gradient. Significant positive turning towards gradient was measured in the presence of brain pulp and netrin-1 (1 μg mL(-1)), relative to control cultures which received no external guidance cue (p < 0.001). Netrin-1 released from transfected fibroblasts had the most positive turning effect of all the chemoattractive cues tested (p < 0.001). Slit-2 exhibited strong chemorepellant characteristics on both hippocampal and DRG neurite guidance at 250 ng mL(-1) concentration. Slit-2 also showed similar behavior on DRG neuron invasion into 3D collagen gel (p < 0.01 relative to control cultures). Taken together, the results suggest the utility of this microfluidic device to generate stable chemogradients for studying neurobiology, cell migration and proliferation, matrix remodeling and co-cultures with other cell lines, with potential applications in cancer biology, tissue engineering and regenerative medicine.  相似文献

Homopolymer grafts from α-tert-butoxy-ω-vinylbenzyl-polyglycidol (PGL) were prepared on gold and stainless steel (SS) substrates modified by 4-benzoyl-phenyl (BP) moieties derived from the electroreduction of the parent salt 4-benzoyl benzene diazonium tetrafluoroborate. The grafted BP aryl groups efficiently served to surface-initiate photopolymerization (SIPP) of PGL. In similar conditions, SIPP of hydroxyethyl methacrylate (HEMA) permitted the production of PHEMA grafts as model surfaces. Water contact angles were found to be 66°, 15°, and 0° for SS-BP, SS-PHEMA, and SS-PPGL, respectively. The spontaneous spreading of water drops on SS-PPGL was invariably observed with 1.5 μL water drops. PPGL thus appears as a superhydrophilic polymer. Resistance to nonspecific adsorption of proteins of PPGL and PHEMA grafts on gold was evaluated by surface plasmon resonance (SPR) using antibovine serum albumin (anti-BSA). The results conclusively show that PPGL-grafts exhibit enhanced resistance to anti-BSA adsorption compared to the well-known hydrophilic PHEMA. PPGL grafts were further modified with BSA through the carbonyldiimidazole activation of the OH groups providing immunosensing surfaces. The so-prepared PPGL-grafted BSA hybrids specifically interacted with anti-BSA in PBS as compared to antimyoglobin. It is clear that the superhydrophilic character of PPGL grafts opens new avenues for biomedical applications where surfaces with dual functionality, namely, specific protein grafting together with resistance to biofouling, are required.  相似文献

Differentiation matrices associated with radial basis function (RBF) collocation methods often have eigenvalues with positive real parts of significant magnitude. This prevents the use of the methods for time‐dependent problems, particulary if explicit time integration schemes are employed.
