The current study attempted, for the first time, to qualitatively and quantitatively determine the phytochemical components of Elatostema papillosum methanol extract and their biological activities. The present study represents an effort to correlate our previously reported biological activities with a computational study, including molecular docking, and ADME/T (absorption, distribution, metabolism, and excretion/toxicity) analyses, to identify the phytochemicals that are potentially responsible for the antioxidant, antidepressant, anxiolytic, analgesic, and anti-inflammatory activities of this plant. In the gas chromatography-mass spectroscopy analysis, a total of 24 compounds were identified, seven of which were documented as being bioactive based on their binding affinities. These seven were subjected to molecular docking studies that were correlated with the pharmacological outcomes. Additionally, the ADME/T properties of these compounds were evaluated to determine their drug-like properties and toxicity levels. The seven selected, isolated compounds displayed favorable binding affinities to potassium channels, human serotonin receptor, cyclooxygenase-1 (COX-1), COX-2, nuclear factor (NF)-κB, and human peroxiredoxin 5 receptor proteins. Phytol acetate, and terpene compounds identified in E. papillosum displayed strong predictive binding affinities towards the human serotonin receptor. Furthermore, 3-trifluoroacetoxypentadecane showed a significant binding affinity for the KcsA potassium channel. Eicosanal showed the highest predicted binding affinity towards the human peroxiredoxin 5 receptor. All of these findings support the observed in vivo antidepressant and anxiolytic effects and the in vitro antioxidant effects observed for this extract. The identified compounds from E. papillosum showed the lowest binding affinities towards COX-1, COX-2, and NF-κB receptors, which indicated the inconsequential impacts of this extract against the activities of these three proteins. Overall, E. papillosum appears to be bioactive and could represent a potential source for the development of alternative medicines; however, further analytical experiments remain necessary. 相似文献
A new vacancy ordered, anion deficient perovskite modification with composition of BaCoO2.67 (Ba3Co3O8□1) has been prepared via a two-step heating process. Combined Rietveld analysis of neutron and X-ray powder diffraction data shows a novel ordering of oxygen vacancies not known before for barium cobaltates. A combination of neutron powder diffraction, magnetic measurements, and density functional theory (DFT) studies confirms G-type antiferromagnetic ordering. From impedance measurements, the electronic conductivity of the order of 10−4 S cm−1 is determined. Remarkably, the bifunctional catalytic activity for oxygen evolution reaction (OER) and oxygen reduction reaction (ORR) is found to be comparable to that of Ba0.5Sr0.5Co0.8Fe0.2O3–y, confirming that charge-ordered anion deficient non-cubic perovskites can be highly efficient catalysts. 相似文献
Mirabegron is a novel β3-adrenoceptor agonist containing an amide group. It was subjected to stress conditions of acidic and alkaline hydrolyses. The hydrolytic degradation product was isolated and its structure was confirmed using mass and IR spectrometry. Two stability-indicating chromatographic methods have been proposed for the determination of mirabegron. TLC method was applied using silica gel as stationary phase and chloroform–methanol–ammonia (9.0:1.0:0.1 by volume) as the mobile phase, and chromatograms were scanned at 250 nm. Accurate determination of the drug was achieved over the concentration range of 2–12 μg per band. In addition, an isocratic HPLC method was developed on Agilent C18 column (150 mm × 4.5 mm I.D., particle size 5 µm) using ethanol-phosphate buffer pH 2.5 (30:70, by volume) as a mobile phase with flow rate of 1 mL min?1.The intact drug was detected at 250 nm with running time less than 5 min. Mirabegron was determined accurately in a concentration range of 1–25 µg mL?1. The proposed chromatographic methods were applied successfully for the assay of mirabegron in pharmaceutical dosage form and both methods were validated as per the International Conference on Harmonization guidelines and statistically compared with a reported gradient HPLC method. 相似文献
The anticancer activity of terretonin N (1) and butyrolactone I (2), obtained from the thermophilic fungus Aspergillus terreus TM8, was intensively studied against prostate adenocarcinoma (PC-3) and ovary adenocarcinoma (SKOV3) human cell lines. According to this study, both compounds showed potent cytotoxicity towards ovarian adenocarcinoma cells (SKOV3) with IC50 1.2 and 0.6 μg/mL, respectively. With respect to metastatic prostate cells (PC-3), the two compounds 1 and 2 showed a significantly promising cytotoxicity effect with IC50 of 7.4 and 4.5 μg/mL, respectively. The tested fungal metabolites showed higher rates of early and late apoptosis with little or no necrotic apoptotic pathway in all treated prostate adenocarcinoma (PC-3) and ovary adenocarcinoma (SKOV3) human cell lines, respectively. The results reported in this study confirmed the promising biological properties of terretonin N (1) and butyrolactone I (2) as anticancer agents via the induction of cellular apoptosis. However, further studies are needed to elucidate the molecular mechanism by which cellular apoptosis is induced in cancer cells. 相似文献
One-pot three-component synthesis of twelve different chromeno[2,3-b]indole derivatives were achieved by the condensation of β-naphthol, oxindole and various substituted aldehydes. Two more chromeno[2,3-b]indole derivatives were also synthesized through one-pot two-component condensation of salicylaldehyde with oxindole/chlorooxindole. Both the condensations were achieved by using Keggin-type heteropoly-11-molybdo-1-vanadophosphoric acid, H4[PVMo11O40] supported on montmorillonite K-10 clay for about 10% as catalyst under environmentally benign solvent-free reaction condition. Shorter reaction time, excellent yield of product, sustainability of catalytic material and simple workup procedure under green experimental conditions are the advantages of this protocol.
The environmental degradation, combined with the continuous depletion of the world's fossil fuel reserves, has forced the search for alternative fuels. This study was performed to investigate the performance of novel biodiesels in the CI engine. The experiments were performed at three different compressions ratios (16:1, 17:1, 18:1) and four loading conditions (25%, 50%, 75%, 100%). Different types of fuels such as jatropha biodiesel (JB), roselle biodiesel (RB), and ternary biodiesel (TB) were prepared and analyzed. The thermal performance of different fuels was analyzed in terms of brake thermal efficiency (BTE), brake specific fuel consumption (BSFC), and exhaust gas temperature (EGT). The emission characteristics such as CO2 emission, NOx emission, and smoke emission were analyzed for all types of fuels. The results of these fuels in the engine were compared with mineral diesel (MD). The BTE was increased with increasing compression ratios and loads for all types of fuels. The BSFC was increased with increasing compression ratios but decreased with increasing loads. The increase in emission of NOx was observed at higher compression ratios and loads. However, the CO2 emission was decreased at higher loads and lower compression ratio. The performance curves achieved with a 20% jatropha biodiesel blend showed results that were approximate to those obtained with pure MD. The comparative analysis between different fuels showed that JB exhibit higher thermal performance as compared to other biodiesels. The steady laminar flow and thermal characteristics of a continuously moving vertical sheet of extruded material are studied close to and far downstream from the extrusion slot. The velocity and temperature variations, obtained by a finite volume method, are used to map out the entire forced, mixed and natural convection regimes. The effects of the Prandtl number (Pr) and the buoyancy force parameter (B) on the friction and heat transfer coefficients are investigated. Comparisons with experimental measurements and solutions by others in the pure forced and pure natural convection regions are made. In the mixed convection region, the results are compared with available finite-difference solutions of the boundary layer equations showing excellent agreement. The region close to the extrusion slot is characterized as a non-similar forced-convection dominated region in which NuxRex−1/2 drops sharply with increasing Richardson number (Rix). This is followed by a self-similar forced-convection dominated region in which NuxRex−1/2 levels off with increasing Rix until the buoyancy effect sets in. The existence and extent of the latter region depend upon the value of B. A non-similar mixed convection region where increasing buoyancy effect enhances the heat transfer rate follows. Finally, this region is followed downstream by a self-similar natural-convection dominated region in which NuxRex−1/2 approaches the pure natural convection asymptote at large Rix. Critical values of Rix to distinguish the various convection regimes are determined for different Pr and B. 相似文献
