Coordination within and between organisms is one of the most complex abilities of living systems, requiring the concerted regulation of many physiological constituents, and this complexity can be particularly difficult to explain by appealing to physics. A valuable framework for understanding biological coordination is the coordinative structure, a self-organized assembly of physiological elements that collectively performs a specific function. Coordinative structures are characterized by three properties: (1) multiple coupled components, (2) soft-assembly, and (3) functional organization. Coordinative structures have been hypothesized to be specific instantiations of dissipative structures, non-equilibrium, self-organized, physical systems exhibiting complex pattern formation in structure and behaviors. We pursued this hypothesis by testing for these three properties of coordinative structures in an electrically-driven dissipative structure. Our system demonstrates dynamic reorganization in response to functional perturbation, a behavior of coordinative structures called reciprocal compensation. Reciprocal compensation is corroborated by a dynamical systems model of the underlying physics. This coordinated activity of the system appears to derive from the system’s intrinsic end-directed behavior to maximize the rate of entropy production. The paper includes three primary components: (1) empirical data on emergent coordinated phenomena in a physical system, (2) computational simulations of this physical system, and (3) theoretical evaluation of the empirical and simulated results in the context of physics and the life sciences. This study reveals similarities between an electrically-driven dissipative structure that exhibits end-directed behavior and the goal-oriented behaviors of more complex living systems.  相似文献

Serrated jet nozzles are considered to be an efficient and practical passive control approach for jet noise. However, some fundamental mechanisms of serration effects on jet noise are not fully understood, especially in terms of the sound source. In this paper, a high-fidelity simulation framework using large-eddy simulation (LES) is demonstrated to predict near-field turbulence and far-field acoustics from an ultra-high-bypass-ratio engine with round and serrated nozzles. Far-field sound is predicted using Ffowcs Willams–Hawkings (FWH) integration. The results show that the serrated nozzle increases mixing near the nozzle and hence the turbulence decay rate, reducing the turbulence level downstream. The serrations shift the energy from the low frequencies to the high frequencies and decrease overall sound pressure levels by about 3 dB over the low-frequency range. Sound sources are analysed based on fourth-order space–time correlations. There are six major source components (R1111, R2222, R3333, R1313, R1212, and R2323) inside the jet shear layers. The serrations are able to reduce the amplitude of these source terms, causing them to decay rapidly to a level below the round nozzle jet within 2D downstream of the nozzle.  相似文献

The use of nanoparticle (NP) systems to control cellular physiology, including membrane potential, can facilitate furthered understanding of many disparate cellular processes ranging from cellular proliferation to tissue regeneration. A gold NP (AuNP) bioconjugate system based on the honeybee venom peptide, tertiapin‐Q (AuNP‐TPN‐Q), that depolarizes membrane potential by targeting inward rectifier potassium channels (Kir), is developed. The conjugate elicits, in a peptide concentration–dependent manner, a greater and more rapid depolarization response compared to the free peptide alone. The specificity of the interaction of the AuNP‐TPN‐Q conjugate with the Kir channel using immunocytochemistry and competition binding assays is confirmed. It is further shown that membrane depolarization is photothermally reversible via the laser‐induced plasmonic heating of the AuNP, providing a level of control over Kir channels not afforded by currently available drugs. The functional nanobioconjugate described herein provides a new research tool for understanding the intricacies of ion channel activity and the modulation of cellular membrane potential.  相似文献

A novel PCR technology was developed to detect short DNA fragments using species-specific primers for rapid and non-sequencing authentication of Bombyx batryticatus based on differences in the mitochondrial genome. Three specifically designed primer reactions were established to target species for the reliable identification of their commercial products. They were confirmed to have a high inter-species specificity and intra-species stability. The limit of detection was estimated as 1 ng of genomes for Beauveria bassiana and 100 pg for Bombyx mori and Metarhizium anisopliae. Furthermore, validation results demonstrated that raw materials and their processed products can be conveniently authenticated with good sensitivity and precision using this newly proposed approach. In particular, when counterfeits were assayed, these primer sets performed well, whereas COI barcoding technology did not. These could also assist in the discrimination and identification of adulterates of other animal-derived medicines in their pulverized and processed forms and even in complexes.  相似文献

Ketones 2 (MSDC-0160) and 3 (MSDC-0602) had been selected for clinical development, however their initial syntheses were considered suboptimal for application deep into clinical trials. Difficulties ranging from the nature of the starting material, alcohol oxidation problems, epoxide opening regioisomeric issues, and endgame ketone redox problems had been encountered. Direct ketone introduction/maintenance was desired for maximum efficiency and convergence was found to be critically dependent upon the acidity of the nucleophilic species (13, 18) and the use of pre- or post-alkylative oximino-ether/oxime protection (vide infra). Improvements in overall yield for the syntheses of 2 (MSDC-0160) and 3 (MSDC-0602) from 20% (2) and 31% (3) respectively, to 44% (2) and 59% (3) were realized.  相似文献

The total synthesis of representative members of the schizozygine alkaloids, (+)‐vallesamidine and (+)‐14,15‐dehydrostrempeliopine, were completed from a late‐stage divergent intermediate. The synthesis took advantage of efficient nitro‐group reactions with the A/B/C ring skeleton constructed concisely on a gram scale through an asymmetric Michael addition, nitro‐Mannich/lactamisation, Tsuji–Trost allylation, and intramolecular C?N coupling reaction. Other key features of the synthesis are a novel [1,4] hydride transfer/Mannich‐type cyclisation to build ring E and a diastereoselective ring‐closing metathesis reaction to construct ring D. This approach gave access to a late‐stage C14,C15 alkene divergent intermediate that could be simply transformed into (+)‐vallesamidine, (+)‐14,15‐dehydrostrempeliopine, and potentially other schizozygine alkaloids and unnatural derivatives.  相似文献
