Small GTPases are a family of GDP‐/GTP‐binding proteins that serve as biomolecular switches inside cells to control a variety of essential cellular processes. Aberrant function and regulation of small GTPases is associated with a variety of human diseases, thus rendering these proteins highly interesting targets in drug discovery. However, this class of proteins has been considered “undruggable”, as intensive decade‐long efforts did not yield clinically relevant direct modulators of small GTPases. Recently, the targeting of small GTPases has gained fresh impetus through the discovery of novel transient cavities on the protein surfaces and the application of new targeting strategies. Besides Ras proteins, other small GTPases have attracted increased attention since improved biological insight in combination with novel targeting strategies identified them as promising targets in drug discovery. This Review gives an overview of relevant aspects of the superfamily of small GTPases and summarizes recent progress and perspectives for the direct modulation of these challenging targets. 相似文献

The interplay between the self-assembly and surface chemistry of 2,3,6,7,10,11-hexaaminotriphenylene (HATP) on Cu(111) was complementarily studied by high-resolution scanning tunneling microscopy (STM) and X-ray photoelectron spectroscopy (XPS) under ultra-high vacuum conditions. To shed light on the competitive metal coordination, comparative experiments were carried out on pristine and nickel-covered Cu(111). Directly after room-temperature deposition of HATP onto pristine Cu(111), self-assembled aggregates were observed by STM, and XPS results indicated still protonated amino groups. Annealing up to 200 °C activated the progressive single deprotonation of all amino groups as indicated by chemical shifts of both the N 1s and C 1s core levels in the XP spectra. This enabled the formation of topologically diverse π–d conjugated coordination networks with intrinsic copper adatoms. The basic motif of these networks was a metal–organic trimer, in which three HATP molecules were coordinated by Cu3 clusters, as corroborated by the accompanying density functional theory (DFT) simulations. Additional deposition of more reactive nickel atoms resulted in both chemical and structural changes with deprotonation and formation of bis(diimino)–Ni bonded networks already at room temperature. Even though fused hexagonal metal-coordinated pores were observed, extended honeycomb networks remained elusive, as tentatively explained by the restricted reversibility of these metal–organic bonds. 相似文献

Abstract The temperature dependence of the three elastic constants k ii (i = 1, 2, 3) and the twist viscosity γ1 of two nematic side chain polyacrylates and one comparable low molecular weight compound have been measured by means of the Freedericksz effect. The change from the low to the high molecular liquid crystal causes a change of the ratio k 33/k 11 from greater to less than unity, but the order of magnitude of the elastic constants remains the same. In contrast, the twist viscosity of the polymeric liquid crystal is about 1000 times greater in magnitude than that of a comparable low molecular weight liquid crystal. The activation energy for the viscosity of the polymer differs by a factor 3–4 from that of the low molecular weight liquid crystal. The elastic constants as well as the twist viscosity show a quadratic dependence on the order parameter S over a wide temperature range. 相似文献

The flow structure of strongly turbulent Taylor-Couette flow with Reynolds numbers up to Re(i)=2×10(6) of the inner cylinder is experimentally examined with high-speed particle image velocimetry (PIV). The wind Reynolds numbers Re(w) of the turbulent Taylor-vortex flow is found to scale as Re(w)∝Ta(1/2), exactly as predicted by Grossmann and Lohse [Phys. Fluids 23, 045108 (2011).] for the ultimate turbulence regime, in which the boundary layers are turbulent. The dimensionless angular velocity flux has an effective scaling of Nu(ω)∝Ta(0.38), also in correspondence with turbulence in the ultimate regime. The scaling of Nu(ω) is confirmed by local angular velocity flux measurements extracted from high-speed PIV measurements: though the flux shows huge fluctuations, its spatial and temporal average nicely agrees with the result from the global torque measurements. 相似文献
