The first total synthesis of lajollamycin B, a structurally novel nitro-tetraene spiro-β-lactone/γ-lactone antibiotic, is described. The convergent synthesis involves the construction of the C8′–C11′ nitrodienylstannane and its coupling with the segment prepared from the C1′–C7′ ω-iodoheptadienoic acid and the right-hand heterocyclic fragment, which has been utilized for our previous syntheses of oxazolomycin A. The revision of the geometry of the terminal Δ10′, 11′-double bond from E to Z is also described for the structure of natural lajollamycin B.  相似文献

The nondestructive testing technique is applied to evaluate the temper embrittlement of normalized and tempered steel; it is based on the anodic polarization behavior in calcium nitrate solution. The steel specimen is aged at a stress of 210 MPa at 450°C for up to 8800 h in order to examine the effect of applied stress on the enhancement of intergranular segregation of phosphorus. The secondary peak observed in the polarization curve for the material aged under stress was compared with that for the non-stressed. It can be quantified with the degree of intergranular segregation of phosphorus. The data suggest that the enhancement of phosphorus segregation due to the applied stress is only recognized after 3000 h of aging. This effect diminishes with increasing aging time.  相似文献

Highly heterotactic poly(4‐vinyl pyridine)s (P4VPs) with the fraction of mr content (fmr) > 0.81 were synthesized by free radical polymerization of 4‐vinyl pyridine (4VP) with randomly methylated β‐cyclodextrin (β‐RMCD) in acidic aqueous media of HNO3 and CF3COOH at 40 °C. The heterotacticity of P4VP strongly depended on the neutralization of 4VP. The complete neutralization of 4VP with HNO3 or CF3COOH increased the heterotacticity of P4VP, whereas atactic P4VP was obtained in water. The partial decomposition of β‐RMCD by HCl reduced the heterotacticity of P4VP (fmr ≈ 0.74). The structures of inclusion complexed monomers were determined by Job's plot, 2D NMR with nuclear Overhauser enhancement spectroscopy analyses, and simulation by MM2. The 1:2 complex with [β‐RMCD]:[4VP] with meso placement of 4VPs in β‐RMCD was formed when 4VP was completely neutralized with acid, whereas the 1:1 complex was formed in water. Bacterial trans-acyltransferase polyketide synthases (trans-AT PKSs) are multimodular megaenzymes that biosynthesize many bioactive natural products. They contain a remarkable range of domains and module types that introduce different substituents into growing polyketide chains. As one such modification, we recently reported Baeyer–Villiger-type oxygen insertion into nascent polyketide backbones, thereby generating malonyl thioester intermediates. In this work, genome mining focusing on architecturally diverse oxidation modules in trans-AT PKSs led us to the culturable plant symbiont Gynuella sunshinyii, which harbors two distinct modules in one orphan PKS. The PKS product was revealed to be lobatamide A, a potent cytotoxin previously only known from a marine tunicate. Biochemical studies show that one module generates glycolyl thioester intermediates, while the other is proposed to be involved in oxime formation. The data suggest varied roles of oxygenation modules in the biosynthesis of polyketide scaffolds and support the importance of trans-AT PKSs in the specialized metabolism of symbiotic bacteria.  相似文献

Transformation of plant-origin 7,14-dihydroxy-ent-kaurenes to ent-abietanes having a cis-fused α-methylene γ-lactones was accomplished efficiently under the Mitsunobu reaction conditions. The yields of the desired products were apparently influenced by the steric hindrance at C-1. The cytotoxic activity on P388 murine leukemia cells of the ent-abietanes having cis-fused α-methylene γ-lactones produced were assayed.  相似文献

Site-directed spin labeling electron spin resonance (ESR) was applied to investigate the local environment of the cyanobacterial circadian clock protein KaiB. We prepared five cysteine residue-substituted mutants of KaiB labeled with maleimide spin label (MSL). By comparing the ESR spectra of KaiBs carrying MSL at different positions (Thr64, Lys67, Tyr94, Gly98, and Ala101), local conformational changes were identified. The ESR spectra of MSL-T64C and MSL-K67C showed the relatively slow motion of MSL characterized by τ?=?79 and 59?ns at 4°C, respectively. The spectra of MSL-Y94C, MSL-G98C and MSL-A101C showed relatively fast motion characterized by τ?=?8.0, 4.1 and 3.1?ns at 4°C, respectively. These differences were explained by the local environments of the position in KaiB. On incubation at 40°C for 24?h, all ESR spectra of the labeled KaiBs changed, which can be explained by the structural relaxation of KaiB.  相似文献
