Dilatometrical indication can be employed to the neutralizing reactions of polybasic acids and is demonstrated by the following acid-base couples: H2SO4/NaOH, H3PO4/NaOH and H5JO6/NaOH. With amounts between 0.5 and 4 mMol several, frequently all, equivalence points can be determined with a deviation of less than 0.2%. The determination of the changes of molar volume of the reactions depending on the concentrations allow conclusions as to the course of the reactions.  相似文献

A sensitive and selective bioanalytical liquid chromatographic method for diclofenac is described. The drug was detected as a flourescent derivative, which was demonstrated by 1H NMR and mass spectrometric studies to be carbazole acetic acid. Diclofenac was derivatized by UV irradiation of the substance performed as a post-column photoreaction. The reactor was a PTFE capillary wound around a 254-nm UV lamp. Diclofenac was isolated from the plasma samples by precipitation of the proteins with acetonitrile. A 50-microliters volume of the supernatant was injected onto a Nucleosil C18 column. The mobile phase was 32% acetonitrile in pH 6.6 buffer. Carbazole acetic acid was detected by a fluorescence detector using an excitation wavelength of 288 nm and an emission wavelength of 360 nm. The recovery was 92%, the standard curve was linear in the range 10-5500 ng diclofenac per ml plasma, and the relative standard deviation at 10 and 5000 ng of diclofenac per ml plasma was 9.0% and 3.3%, respectively. The limit of detection was 6 ng/ml at an injection volume of 50 microliters. Chromatograms of human and rat plasma containing diclofenac are shown.  相似文献

With the help of a dilatometer which permits the continuous determination of volume changes occuring in chemical reactions in aqueous solutions, the volume change during the precipitation of BaSO4 was followed and applied to the determination of the endpoint of the precipitation. In the case of precipitations of 1 mM BaSO4 the end-point determination can be reproduced with a standard deviation of less than 0.2%. A strict linearity between the amount of Na2SO4 applied and the position of the end-point does not exist. However, a correction factor could be established, so that the absolute determination of sulphate is possible (standard deviation 0.2%). The time required to carry out a single measurement is about 1.5 min. The acid concentration does not influence the composition. On the other hand, the composition was shown to be considerably dependent on Na+ and NH4 + ions. The presence of the cation N(CH3)4 + does not alter the composition of the precipitate. It is obviously too big to substitute the Ba2+ ion.  相似文献

The fine structures of the threshold of subsidiary absorption and parallel pumping, observed on highly polished YIG spheres in 1975 by W. Jantz et al., are discussed with a model which uses instead of plane spin-waves the true modes regarding the shape of the sphere. These spherical modes are magnetostatic modes taking additionally the exchange interaction into account. At the closely spaced resonances of the fine structure of parallel pumping single spherical modes with angular-momentum quantum numberm=0 are excited, whereas at the resonances of subsidiary absorption pairs of spherical modes with big angular-momentum quantum numbers (|m|1000) satisfying the selection rulem+m=1 are excited. The analysis of the fine structure of subsidiary absorption shows that in regimes where the plane wave approximation predicts the excitation of spin-waves with formalk0, modes withk1.5·105 cm–1 have the lowest threshold. This discrepancy is discussed with the effect of surface pit scattering, which increases the threshold of modes withk < 2/pit, where pit is a typical size of the surface pits. Consequently modes withk22/pit instead ofk0 have the lowest threshold.  相似文献

Six new ( 2 , 4 – 8 ) and two known polyketides with a basic structure of an anthraquinone‐xanthone were isolated from mycelia and culture broth of the fungus Engyodontium album strain LF069. The structures and relative configurations of these compounds were established by spectroscopic means, and their absolute configurations were defined mainly by comparison of quantum chemical TDDFT calculated and experimental ECD spectra. Compounds 2 and 4 – 8 were given the trivial names engyodontochone A ( 2 ) and B–F ( 4 – 8 ). Compounds 5 – 8 represent the first example of a 23,28 seco‐beticolin carbon skeleton. The relative and absolute configurations of two known substances JBIR‐97/98 ( 1 ) and JBIR‐99 ( 3 ) were determined for the first time. All isolated compounds were subjected to bioactivity assays. Compounds 1 – 4 exhibited inhibitory activity against methicillin‐resistant Staphylococcus aureus (MRSA) that was 10‐fold stronger than chloramphenicol.  相似文献

An efficient method for analyzing illegal and medicinal drugs in whole blood using fully automated sample preparation and short ultra-high-performance liquid chromatography–tandem mass spectrometry (MS/MS) run time is presented. A selection of 31 drugs, including amphetamines, cocaine, opioids, and benzodiazepines, was used. In order to increase the efficiency of routine analysis, a robotic system based on automated liquid handling and capable of handling all unit operation for sample preparation was built on a Freedom Evo 200 platform with several add-ons from Tecan and third-party vendors. Solid-phase extraction was performed using Strata X-C plates. Extraction time for 96 samples was less than 3 h. Chromatography was performed using an ACQUITY UPLC system (Waters Corporation, Milford, USA). Analytes were separated on a 100 mm?×?2.1 mm, 1.7 μm Acquity UPLC CSH C18 column using a 6.5 min 0.1 % ammonia (25 %) in water/0.1 % ammonia (25 %) in methanol gradient and quantified by MS/MS (Waters Quattro Premier XE) in multiple-reaction monitoring mode. Full validation, including linearity, precision and trueness, matrix effect, ion suppression/enhancement of co-eluting analytes, recovery, and specificity, was performed. The method was employed successfully in the laboratory and used for routine analysis of forensic material. In combination with tetrahydrocannabinol analysis, the method covered 96 % of cases involving driving under the influence of drugs. The manual labor involved in preparing blood samples, solvents, etc., was reduced to a half an hour per batch. The automated sample preparation setup also minimized human exposure to hazardous materials, provided highly improved ergonomics, and eliminated manual pipetting.

Textbook procedures require the use of individual aptamers enriched in SELEX libraries which are subsequently chemically synthesized after their biochemical characterization. Here we show that this reduction of the available sequence space of large libraries and thus the diversity of binding molecules reduces the labelling efficiency and fidelity of selected single aptamers towards different strains of the human pathogen Pseudomonas aeruginosa compared to a polyclonal aptamer library enriched by a whole-cell-SELEX involving fluorescent aptamers. The library outperformed single aptamers in reliable and specific targeting of different clinically relevant strains, allowed to inhibit virulence associated cellular functions and identification of bound cell surface targets by aptamer based affinity purification and mass spectrometry. The stunning ease of this FluCell-SELEX and the convincing performance of the P. aeruginosa specific library may pave the way towards generally new and efficient diagnostic techniques based on polyclonal aptamer libraries not only in clinical microbiology.  相似文献
