Change of α-helical structure of heme protein (Hb) to a β-sheet and random coil conformation because of the interaction of glycine capped gold nanoparticles (20–60 nm) as observed from attenuation total reflectance, absorption, Fourier transform infra red, and Circular Dichroism spectroscopy has been reported in this article. Upon interaction, protein takes a cylindrical shape of length 12 μm and diameter 0.35 μm as revealed from scanning electron microscopy and transmission electron microscopy. The Selected-Area Electron beam Diffraction pattern shows change of crystalline structure in GNP to amorphous nature with the interaction of Hb.  相似文献

pH (low) insertion peptide (pHLIP) is a 36-residue peptide derived from the third transmembrane helix of the membrane protein bacteriorhodopsin. The hydrophobicity of this peptide makes it prone to aggregation even at low concentrations, but this has not been studied in detail. In this work, we characterized monomeric and aggregated forms of pHLIP in aqueous solution (pH 8) at low concentrations (~μM) using fluorescence-based approaches, complemented by circular dichroism (CD) spectroscopy. We show here that monomeric and aggregated pHLIP display differential red edge excitation shift (REES) and CD spectra. These spectroscopic features allowed us to show that pHLIP aggregates even at low concentrations. A detailed knowledge of the aggregation behavior of pHLIP under these conditions will be useful for monitoring and quantifying its interaction with membranes.  相似文献

We have examined the formation of a local moment by considering various defects in ZnO. The localization of the defect induced state is found to determine the presence/absence of a local moment. A lot of attention on the probable origin of magnetism in wide band gap oxides has focused on cation vacancies. Here we show that oxygen interstitial atoms give rise to a large magnetic moment which results in a spin polarization of both the conduction and valence bands, in addition to spin polarized gap states. A Stoner mechanism is invoked and the relevant Stoner parameters are determined to be 0.7 eV for an oxygen atom in the presence of an oxygen interstitial but reduced to 0.2 eV on oxygen in the presence of a Zn vacancy.  相似文献

A highly efficient and mild protocol for the synthesis of imidazo-fused polyheterocycles via Groebke–Blackburn–Bienaymè reaction under the influence of catalytic amount of lanthanum chloride heptahydrate has been described. A real time multimodal data acquisition imaging setup is developed for the electrophoretic movement of plasmonic nanoparticles. Movement of the nanoparticles is recorded by time-lapse digital imaging at 8-megapixel resolutions. The analysis of the moving nanoparticle band is performed using threshold image and color extraction at respective time frames. The migration dynamics is sensitive to size, nature and the extent of the surface conjugation to the nanoparticle. The dynamics of color intensity of the nanoparticles is shown to be dependent on the extent of stabilization of nanoparticle (by a given agent). The stability of nanoparticle is determined by stationary nature and also the relative proportions of pixel intensities in respective color planes (R, G and B). Detergents stabilize nanoparticles in a concentration dependent fashion. In case of neutral polymers the extent of stabilization depends on the relative proportion of the polymer and also on the nature of the same, e.g., PEG (polyethylene glycol) at low concentration imparts higher stability as compared to PVP (polyvinylpyrilidone). The ascending or declining temporal dynamics of color profiles observed in case of citrate stabilized or amino acid conjugated nanoparticles, represent enrichment of plasmonic particles, or their diffusion resulting from loss of charge during migration. The higher dimensional imaging technique thus can be exploited for discriminating the nanoparticles on the basis of their migration behavior and their stability as reflected from color dynamics. The technique is applicable to other nano-sized colored objects, e.g. proteins like hemoglobin where the protein color has important clinical value.  相似文献

Herein, we explored the photophysical properties of the antimalarial, anticancer drug cryptolepine (CRYP) in the presence of the macrocyclic host cucurbit[7]uril (CB7) and DNA with the help of steady‐state and time‐resolved fluorescence techniques. Ground‐state and excited‐state calculations based on density functional theory were also performed to obtain insight into the shape, electron density distribution, and energetics of the molecular orbitals of CRYP. CRYP exists in two forms depending on the pH of the medium, namely, a cationic (charge transfer) form and a neutral form, which emit at λ=540 and 420 nm, respectively. In a buffer solution of pH 7, the drug exists in the cationic form, and upon encapsulation with CB7, it exhibits a huge enhancement in fluorescence intensity due to a decrement in nonradiative decay pathways of the emitting cryptolepine species. Furthermore, docking and quantum chemical calculations were employed to decipher the molecular orientation of the drug in the inclusion complex. Studies with natural DNA indicate that CRYP molecules intercalate into DNA, which leads to a huge quenching of the fluorescence of CRYP. Keeping this in mind, we studied the DNA‐assisted release of CRYP molecules from the nanocavity of CB7. Strikingly, DNA alone could not remove the drug from the nanocavity of CB7. However, an external stimulus such as acetylcholine chloride was able to displace CRYP from the nanocavity, and subsequently, the displaced drug could bind to DNA.  相似文献

Oxicam group of non steroidal anti-inflammatory drugs has been chosen as a prototype molecular group that shows diverse biological functions and dynamic structural features. Photophysical studies of three drugs from this group viz., piroxicam, meloxicam and tenoxicam have been carried out in different solvents with varying polarity, H-bond character and viscosity. The spectral responses of different prototropic forms of these drugs towards varying solvent parameters have been studied, with the aim to characterize their interaction in biomimetic environment non-invasively. The nature of the lowest transition has been identified. The extinction coefficient, quantum yield and viscosity dependence on the nature of the solvents, all indicate the extreme sensitivity of these drugs to their microenvironment.  相似文献

Electronic shearography (ES), a laser interferometry technique, has the potential for large-scale structural inspection and for identifying cracks and strain anomalies. A system based on this technology could possibly be used for noninvasive inspection of structures with high insensitivity and robustness. One major problem is the existence of largerigid body motions in typical engineering structures such as bridges and high-rise buildings. These rigid-body motions are large enough to cause a complete decorrelation of the characteristic speckle pattern that is obtained by illuminating the object surface. This nullifies any possibility of interference between speckled images obtained at different stages of loading, as is necessary for interferometry applications. A systematic study was conducted to characterize the speckle pattern obtained from typical civil structures as a function of the illumination and imaging system. Experiments were carried out to quantify speckle decorrelation as a function of object motion and instrumentation characteristics. A fracture mechanics based finite element (FEM) analysis was carried out on an existing fractured bridge to determine the strains and displacements. The results of these studies were subsequently used to define the parameters of a structural inspection system based on ES for field applications.  相似文献

An all-optical pseudo random binary sequence (PRBS) generator is designed using the hard-limiters and serially interconnected D flip-flops based on two coupled polarization switches (PSWs). The performance of the circuit is evaluated through numerical simulation to confirm its feasibility in terms of the choice of the critical parameters. The proposed scheme has been theoretically demonstrated for a 3-bit degree PRBS.  相似文献

A technique based on self-imaging at finite conjugates has been experimentally demonstrated for visualization of laser-induced photorefractive gratings in BSO. A simple model theory has been used to provide the correct rules of geometrical optics of self-imaging. A novel technique has been used for photoinducing the periodic structure. An application has been made to determine the harmonic components of the nonsinusoidal refractive index modulation in the BSO for a grating period of 10 m and for external DC voltages of 6.0 and 3.0 kV. The technique is directly applicable to the visualization and study of laser-induced gratings (LIG) in thin nonlinear optical media.  相似文献
