To assess the bioequivalence of two zolpidem hemitartrate formulations in 30 healthy volunteers. Plasma samples were obtained over a 24 h period. Plasma concentrations of zolpidem were analyzed by liquid chromatography coupled to tandem mass spectrometry with positive ion electrospray ionization using multiple reaction monitoring. Values of peak concentration (Cmax), area under curve (AUC), half-life, elimination constant, volume of distribution and clearance showed statistically significant differences when comparing women (604.34 ng h/ml, 127.36 ng/ml, 4.4 h, 0.18 1/h, 50.56 L and 8.55 L/h, respectively) and men (276.1 ng h/ml, 70.9 ng/ml, 3.3 h, 0.26 1/h, 91.42 L and 24.34 L/h, respectively), receiving the same dose (5 mg), respectively. The geometric means with corresponding 90% confidence interval for Test/Reference percentage ratios were 99.73% (CI 93.69–106.16) for Cmax, 97.44% (90% CI = 91.85–103.37%) for area under curve of plasma concentration until the last concentration observed (AUClast) and 98.30% (90% CI = 92.48–104.49) for the area under curve between the first sample (pre-dosage) and infinity (AUC0–inf). Since the 90% CI for AUClast, AUC0–inf and Cmax ratios were within the 80–125% interval proposed by the US Food and Drug Administration, it was concluded that zolpidem hemitartrate formulation (5 mg orodispersible tablet) is bioequivalent to the zolpidem hemitartrate formulation (Patz SL 5 mg sublingual tablet) with regard to both the rate and the extent of absorption. A new formulation of zolpidem 2.5 mg may be useful in women for the same clinical benefits as the 5 mg formulation in men. 相似文献

In this paper we establish the characterization of the weighted BMO via two weight commutators in the settings of the Neumann Laplacian on the upper half space and the reflection Neumann Laplacian on with respect to the weights associated to and respectively. This in turn yields a weak factorization for the corresponding weighted Hardy spaces, where in particular, the weighted class associated to is strictly larger than the Muckenhoupt weighted class and contains non-doubling weights. In our study, we also make contributions to the classical Muckenhoupt–Wheeden weighted Hardy space (BMO space respectively) by showing that it can be characterized via the area function (Carleson measure respectively) involving the semigroup generated by the Laplacian on and that the duality of these weighted Hardy and BMO spaces holds for Muckenhoupt weights with while the previously known related results cover only . The application of ω‐transaminase biocatalysts for the synthesis of optically pure chiral amines presents a number of challenges, including difficulties associated with displacing the challenging reaction equilibria. Herein, we report a highly effective approach using low equivalents of the new diamine donor, cadaverine, which enables high conversions of challenging substrates to the corresponding chiral amines in excellent ee. Poly(lactic acid) (PLA) is a versatile, bioabsorbable, and biodegradable polymer with excellent biocompatibility and ability to incorporate a great variety of active agents. Silver sulfadiazine (SDZ) is an antibiotic used to control bacterial infection in external wounds. Aiming to combine the properties of PLA and SDZ, hydrotalcite ([Mg–Al]‐LDH) was used as a host matrix to obtain an antimicrobial system efficient in delivering SDZ from electrospun PLA scaffolds intended for wound skin healing. The structural reconstruction method was successfully applied to intercalate silver sulfadiazine in the [Mg–Al]‐LDH, as evidenced by X‐ray diffraction and thermogravimetric analyses. Observations by scanning electron microscopy revealed a good distribution of SDZ‐[Mg–Al]‐LDH within the PLA scaffold. Kinetics studies revealed a slow release of SDZ from the PLA scaffold due to the intercalation in the [Mg–Al]‐LDH. In vitro antimicrobial tests indicated a significant inhibitory effect of SDZ‐[Mg–Al]‐LDH against Escherichia coli and Staphylococcus aureus. This antibacterial activity was sustained in the 2.5‐wt% SDZ‐[Mg–Al]‐LDH–loaded PLA nanofibers, which also displayed excellent biocompatibility towards human cells. The multifunctionality of the PLA/SDZ‐[Mg–Al]‐LDH scaffold reported here is of great significance for various transdermal applications. 相似文献

We report a catalytic, light-driven method for the intramolecular hydroetherification of unactivated alkenols to furnish cyclic ether products. These reactions occur under visible-light irradiation in the presence of an IrIII-based photoredox catalyst, a Brønsted base catalyst, and a hydrogen-atom transfer (HAT) co-catalyst. Reactive alkoxy radicals are proposed as key intermediates, generated by direct homolytic activation of alcohol O−H bonds through a proton-coupled electron-transfer mechanism. This method exhibits a broad substrate scope and high functional-group tolerance, and it accommodates a diverse range of alkene substitution patterns. Results demonstrating the extension of this catalytic system to carboetherification reactions are also presented. 相似文献

We introduce a new generalization of Alan Day’s doubling construction. For ordered sets \(\mathcal {L}\) and \(\mathcal {K}\) and a subset \(E \subseteq \ \leq _{\mathcal {L}}\) we define the ordered set \(\mathcal {L} \star _{E} \mathcal {K}\) arising from inflation of \(\mathcal {L}\) along E by \(\mathcal {K}\). Under the restriction that \(\mathcal {L}\) and \(\mathcal {K}\) are finite lattices, we find those subsets \(E \subseteq \ \leq _{\mathcal {L}}\) such that the ordered set \(\mathcal {L} \star _{E} \mathcal {K}\) is a lattice. Finite lattices that can be constructed in this way are classified in terms of their congruence lattices.A finite lattice is binary cut-through codable if and only if there exists a 0?1 spanning chain \(\left \{\theta _{i}\colon 0 \leq i \leq n \right \}\) in \(Con(\mathcal {L})\) such that the cardinality of the largest block of ?? i /?? i?1 is 2 for every i with 1≤i≤n. These are exactly the lattices that can be constructed by inflation from the 1-element lattice using only the 2-element lattice. Spondias mombin L. (Anacardiaceae) has a worldwide distribution and is present in all regions of Brazil. Its leaves, flowers and bark are used as teas in folk medicine to treat diseases of the digestive system. This study aimed to evaluate the acute non-clinical toxicity, gastroprotective activity, and the related mechanisms of action of nebulized extract and tablets based on dried Spondias mombin (SmNE). SmNE screening showed the presence of flavonoids (0.65%), polyphenols (25.50%), where the major compound is gallic acid. In the acute oral toxicity assay, a dose of 2000 mg/kg of SmNE administered orally in Swiss mice did not induce any behavioral changes. SmNE (250 or 500 mg/kg p.o) significantly reduced the ulcerative lesion area when compared to the control group in ethanol and non-steroidal anti-inflammatory drug (NSAIDs) models. Results showed that treatment with SmNE (250 mg/kg) reduced acid secretion and gastric content, accompanied with an increase in pH. Previous administration of indomethacin and glibenclamide reversed the protection provided by SmNE, confirming the participation of prostaglandins (PGs) and ATP-sensitive potassium channels (KATP) in its gastroprotective effect. The SmNE tablets met the pharmacopeial quality requirements with gastroprotective activity and similar protection in comparison to the isolated extract administrated. In conclusion, SmNe has a gastroprotective activity related to cytoprotective mechanisms, such as the participation of endogenous prostaglandins and KATP channels, having an anti-secretory effect with systemic action. The formulation obtained presented gastroprotective effects similar to the administration of the extract, the tablets showed favorable compression characteristics by the direct route and met the pharmacopeial quality requirements. 相似文献

Diffusiophoresis phenomenon of aoft particles suspended in binary electrolyte solutions is explored theoretically in this study based on the spherical cell model, focusing on the chemiphoresis component in absence of diffusion potential. Both the electrostatic and hydrodynamic aspects of the boundary confinement, or steric effect, due to the presence of neighboring particles are examined extensively under various electrokinetic conditions. Significant local extrema are found in mobility profiles expressed as functions of the Debye length in general, synchronized with the strength of the motion-inducing double layer polarization. Moreover, a seemingly peculiar phenomenon is observed that the soft particles may move faster in more concentrated suspensions. The competition between the simultaneous enhancement of the motion-inducing electric driving force and the motion-retarding hydrodynamic drag force from the boundary confinement effect of the neighboring particles is found to be responsible for it. The above findings are also demonstrated experimentally in a very recent study on the diffusiophoretic motion of soft particles through porous collagen hydrogels. The results presented here are useful in various practical applications of soft particles like drug delivery. 相似文献

Among additive manufacturing, photocuring 3D printing technologies are very relevant because of its high printing speed and high precision. However, the limited performance of photosensitive thermoset polymers is the bottleneck for the application of photocuring 3D printing in some fields, particularly in the biomedical sector. Thus, the development of biodegradable and biocompatible materials is highly desirable and of utmost importance. In this work, a biodegradable and non-cytotoxic thermoset polymer for photocuring 3D printing is reported. It consists of an unsaturated polyesteramide bearing phenylalanine, 2-butene-1,4-diol and fumarate building blocks, which is photocured under UV irradiation using a low molecular weight poly(ethylene glycol) diacrylate as crosslinker. The main characteristics of the new thermoset are: (1) very high volumetric and mechanical integrity stabilities, comparable to that of photocured epoxides; (2) very high degradation temperature; (3) very low water absorption capacity; (4) relatively fast enzymatic degradation, reaching 16.5% after 3 months; and (5) non-cytotoxic response in presence of epithelial cells, even when soluble molecular fragments coming from biodegradation are considered. These properties favor the future utilization of the new polyether-polyesteramide resin in the manufacturing of more sustainable products via 3D printing methods, such as stereolithography, that uses UV sources. 相似文献
