The endoplasmic reticulum (ER) is an essential organelle of eukaryotic cells. Its main functions include protein synthesis, proper protein folding, protein modification, and the transportation of synthesized proteins. Any perturbations in ER function, such as increased demand for protein folding or the accumulation of unfolded or misfolded proteins in the ER lumen, lead to a stress response called the unfolded protein response (UPR). The primary aim of the UPR is to restore cellular homeostasis; however, it triggers apoptotic signaling during prolonged stress. The core mechanisms of the ER stress response, the failure to respond to cellular stress, and the final fate of the cell are not yet clear. Here, we discuss cellular fate during ER stress, cross talk between the ER and mitochondria and its significance, and conditions that can trigger ER stress response failure. We also describe how the redox environment affects the ER stress response, and vice versa, and the aftermath of the ER stress response, integrating a discussion on redox imbalance-induced ER stress response failure progressing to cell death and dynamic pathophysiological changes.Subject terms: Mechanisms of disease, Cell biology  相似文献

The macrocyclic "cholaphanes" 3a-c were synthesized from the inexpensive steroid cholic acid. Like earlier relatives they feature substantial cavities with inward-directed hydroxyl groups, suitable for binding polar molecules such as carbohydrates in nonpolar media. New features are the externally directed alkyl chains, promoting solubility in organic solvents, and (in the case of 3b/c) reduced conformational freedom resulting from truncation of the steroidal side-chain. In particular, modeling shows that the smallest macrocycle 3c possesses very little flexibility, preferring an open conformation which is also revealed in the X-ray crystal structure of its pentahydrate. NMR studies indicated that all three cholaphanes form 1:1 complexes with octyl beta-D-glucoside in CDCl(3), with K(a) = 600-1560 M(-)(1). Cholaphanes 3b/c proved able to extract methyl beta-D-glucoside from aqueous solutions into CHCl(3). The transport of methyl beta-D-glucoside across a chloroform barrier was also demonstrated for 3c.  相似文献

G-protein-coupled receptors (GPCRs) are the largest family of human membrane proteins and serve as primary targets of approximately one-third of currently marketed drugs. In particular, adenosine A1 receptor (A1AR) is an important therapeutic target for treating cardiac ischemia–reperfusion injuries, neuropathic pain, and renal diseases. As a prototypical GPCR, the A1AR is located within a phospholipid membrane bilayer and transmits cellular signals by changing between different conformational states. It is important to elucidate the lipid–protein interactions in order to understand the functional mechanism of GPCRs. Here, all-atom simulations using a robust Gaussian accelerated molecular dynamics (GaMD) method were performed on both the inactive (antagonist bound) and active (agonist and G-protein bound) A1AR, which was embedded in a 1-palmitoyl-2-oleoyl-glycero-3-phosphocholine (POPC) lipid bilayer. In the GaMD simulations, the membrane lipids played a key role in stabilizing different conformational states of the A1AR. Our simulations further identified important regions of the receptor that interacted distinctly with the lipids in highly correlated manner. Activation of the A1AR led to differential dynamics in the upper and lower leaflets of the lipid bilayer. In summary, GaMD enhanced simulations have revealed strongly coupled dynamics of the GPCR and lipids that depend on the receptor activation state. © 2019 Wiley Periodicals, Inc.  相似文献

This study deals with the synthesis of cysteine capped gold nanoparticles with an average size of 12 nm by borohydride reduction and spectroscopic identification of SAu interaction. We have studied the interaction of thiol with gold nanoparticles in aqueous medium by employing UV-vis, Raman, NMR, and FT-IR spectroscopy. The shifting of gold plasmon resonance in the UV-vis spectra shows the stabilization of gold nanoparticles by cysteine. The disappearance of S-H stretching in both the IR and Raman spectra and the shifting of the NMR signals of the protons in close proximity to the metal center supported the existence of the S-Au interaction in cysteine capped gold nanoparticles. The TEM images shows cysteine capped gold nanoparticles as distinct and spherical entities as compared to free colloidal gold nanoparticles.  相似文献

This work reports the electrolyte induced aggregation of gold nanoparticles directly conjugated to amino acid by chemical reduction in aqueous solution. The study was focused on three different classes of amino acids depending on the nature of alpha substituent, viz. l-cysteine, l-leucine, and l-asparagine. The band broadening and the red shift of surface plasmon band with increase in flocculation parameter showed the aggregation of gold nanoparticles with increase in electrolyte concentration and decrease in pH as monitored by UV-visible spectrophotometer. The (1)H NMR spectrum demonstrates that the sulfide bond of cysteine and alpha amino group of leucine and asparagine interact with nanoparticles surface. Furthermore, transmission electron microscope (TEM) and thermogravimetric analysis (TGA) were performed to characterize and to support the fate of stabilization of the gold nanoparticles by amino acid.  相似文献

Here, we present the spectroscopic ellipsometry investigation of synthetically grown wafer-scale two-dimensional (2D) MoS2 and WSe2 films to access quality and thickness uniformity. MoS2 and WSe2 samples were grown by chemical vapor deposition and atomic layer deposition, respectively. Complex dielectric function (ε=ε1+iε2) and thickness information of these 2D films were extracted from the measured data using multilayer optical calculations. Broad spectral range (1.2–6 eV) and multiple angles of incidence were used to reduce correlations among fitting parameter. Lineshape of ε of MoS2 and WSe2 monolayer films are consistent with literature but shows higher values, suggests better quality of our samples. Eight-inch wafer size MoS2 monolayer sample shows ~ 70% uniformity with an average thickness of 0.65 ± 0.2 nm, and three-layer WSe2 sample of 8 × 1 cm2 area shows ~ 80% uniformity with an average thickness of 2.5 ± 0.4 nm. Our results will be helpful to accelerate commercialization process of 2D devices.  相似文献

For the first time, the pressure and temperature dependence of a chemical reaction at the solid/solution interface is studied by scanning tunneling microscopy (STM), and thermodynamic data are derived. In particular, the STM is used to study the reversible binding of O(2) with cobalt(II) octaethylporphyrin (CoOEP) supported on highly oriented pyrolytic graphite (HOPG) at the phenyloctane/CoOEP/HOPG interface. The adsorption is shown to follow the Langmuir isotherm with P(1/2)(298K) = 3200 Torr. Over the temperature range of 10-40 °C, it was found that ΔH(P) = -68 ± 10 kJ/mol and ΔS(P) = -297 ± 30 J/(mol K). The enthalpy and entropy changes are slightly larger than expected based on solution-phase reactions, and possible origins of these differences are discussed. The big surprise here is the presence of any O(2) binding at room temperature, since CoOEP is not expected to bind O(2) in fluid solution. The stability of the bound oxygen is attributed to charge donation from the graphite substrate to the cobalt, thereby stabilizing the polarized Co-O(2) bonding. We report the surface unit cell for CoOEP on HOPG in phenyloctane at 25 °C to be A = (1.46 ± 0.1)n nm, B = (1.36 ± 0.1)m nm, and α = 54 ± 3°, where n and m are unknown nonzero non-negative integers.  相似文献
