Development of an LC‐MS/MS method for determining the pharmacokinetics of clonidine following oral administration of Zhenju antihypertensive compound

Zhenju antihypertensive compound (ZJAHC) is a combined Chinese–Western medicine formula including clonidine (CLO), hydrochlorothiazide (HCT), rutin, Chrysanthemum indicum extract and pearl powder. Compared with CLO preparations, ZJAHC shows improved activities and decreased adverse effects. It is believed that the side effects of CLO are caused by its high peak plasma concentration. Hence, study of the influence of ZJAHC on the pharmacokinetic behaviors of clonidine seems essential. In present study, the plasma concentrations of CLO were determined with a liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method. The MS/MS transitions monitored for clonidine and internal standard were 230.2 → 213.1 and 152.2 → 110.2, respectively. The analyte was quantified in a single run within 3 min. The pharmacokinetic study showed that the area under the plasma concentration–time curve of CLO in ZJAHC (60 µg/kg CLO) was similar to that of CLO‐HCT‐high (120 µg/kg CLO) but the peak concentration was much lower than that in CLO‐HCT‐high. ZJAHC could enhance the bioavailability without greatly increasing peak concentration of clonidine. This comprehensive effect of enhancing the bioavailability and avoiding the high peak plasma concentration for CLO might mainly result from the co‐contribution of Western medicine and traditional Chinese medicine (TCM), while the effect of TCM was stronger than that of Western medicine. 熔融LiCl及其急冷非晶键取向序的分子动力学模拟研究

本文进一步讨论了我们新近提出的等近邻数键球谐函数方法,并通过分子动力学模拟研究了熔融LiCl及其急冷过程。对计算机模拟各时间步产生的瞬态构型进行平均,计算了不同温度下键序Q_l谱.将模拟Q_l谱与线性组合模型Q_l谱比较,观察到模拟Q_l谱与含90％局部正四面体结构的组合模型十分相似,表明熔融Licl及其急冷非晶中局部键取向序明显倾向于正四面体序。急冷过程中不同温度下局部键取向序可用同一线性组合模型描述,但模型方差随模拟温度明显变化。方差随温度而降低,且在发生玻璃态转变时有一显著下降。不同温度下的键角分布也作了计算,观察到键角分布在109°(正四面体局部结构键角)附近且分布峰随温度升高而展宽,与键序Q_l得到的结论一致。     

具有某种断面的半群的研究进展

本文综述了几类具有特殊断面的半群的近期研究结果。在介绍逆半群和正则半群的一般结构之后，概述了具有逆断面的正则半群的结构和同余格的研究成果。总结了作为逆断面的推广的可裂断面，纯正断面，正则^*－断面和恰当断面。提出了可以进一步研究的重要的问题。     

Amending cultures of selenium-resistant bacteria with dimethyl selenone

A possible biological intermediate in the reduction and methylation of selenium oxyanions, dimethyl selenone, was synthesized, and the first experiments involving the amendment of selenium resistant bacterial cultures with this compound are reported. The amount of volatile, reduced selenium-containing species released from these cultures into the headspace is significantly more than that produced in analogous experiments involving sodium selenate amended cultures. Dimethyl selenone is reduced in the presence of dimethyl sulfide and dimethyl disulfide in a complex growth medium, trypticase soy broth with 0.1% nitrate. This reduction occurs whether or not the reduced sulfur compounds are biologically produced.     

Optimal Investment and Dividend Strategy in the Discrete Sparre Andersen Risk Model

In the discrete-time Sparre Andersen risk model with investments and dividend payments, the company controls the dividend payments and the proportions of venture investments in order to maximize the cumulative expected discounted dividends prior to ruin. The paper gets the algorithm of the optimal dividend strategy by analyzing a Hamilton-Jacobi-Bellman equation and transforming the value function. Furthermore, the existence of the optimal solution of the transformation function is proved by using compression mapping and fixed point principle. In order to make the calculation easier, this paper also proposes an innovative random simulation method for the optimal strategy, and proves that the simulation result is the consistent estimate of the real value. Finally, the random simulation method in the Matlab is used for numerical analysis in an example, which shows the innovative simulation method is a very good and helpful method for making dividend payment and investment decisions.     

MALDI－TOF－MS法分析人血清白蛋白

用基质辅助激光解吸附飞行时间质谱（ＭＡＬＤＩ－ＴＯＦ－ＭＳ）法分析测定了人血清白蛋白。结果表明，全军血液制品研究中心应用Ｒｉｖａｎｏｌ和Ｃｏｈｎ＇ｓ两种方法分离提取的人血清白蛋白，其分子量与理论计算值相吻合，其纯度与市售标准人血清白蛋白相一致，用ＭＡＬＤＩ－ＴＯＦ－ＭＳ法分析鉴定人血清白蛋白，准确、简便和灵敏（仅需ＰＭ浓度样品），且重复性好，日内和日间差的变异系数均小于０．０３％，是ＳＤＳ－ＰＡＧＥ电泳法所无法比拟的。     

降铅冲剂对低水平铅暴露仔鼠血铅及脑铅的影响

目的:研究降铅冲剂对低水平铅暴露仔鼠血铅及脑铅的影响,为评价降铅冲剂的作用提供依据。方法:选用Wister大鼠仔鼠60只,体质量80~100 g,随机分成6组,每组10只,分别为空白组、模型对照组、EDTA对照组、降铅冲剂低剂量组、降铅冲剂中剂量组、降铅冲剂高剂量组;造模后开始给药,EDTA对照组给CaNa2EDTA注射液10(mg/kg)/d腹腔注射,降铅冲剂低、中、高剂量组分别予降铅冲剂5(g/kg)/d、10(g/kg)/d、20(g/kg)/d的剂量灌胃,空白组和模型对照组予等量蒸馏水灌胃,给药共30 d。用石墨炉原子吸收光谱仪测定仔鼠血铅、脑铅。结果:(1)染铅后,各染铅组的血铅水平与空白组比较都显著升高(P<0.001),经过30 d的治疗后,各治疗组的治疗后血铅水平均有不同程度下降,与模型对照组比较均有统计学意义。(2)染铅后,各染铅组的脑铅水平与空白组比较都显著升高(P<0.05),治疗后,各组脑铅与治疗前比较均显著升高,降铅冲剂低、中剂量组的脑铅与模型对照组比较显著下降。结论:降铅冲剂能显著降低低水平铅暴露仔鼠的血铅和脑铅含量,其综合疗效优于CaNa2EDTA。     

壳聚糖负载贵金属催化剂的研究进展

脲基苯乙炔4-′(对甲基苯基)-6-苯基-2,2′-二联吡啶铂(II)络合物的合成

本文通过异氰酸酰化反应合成了含有脲基的苯乙炔新型配体及以它作为辅助配体的4′-(对甲基苯基)-6-苯基-2,2′-二联吡啶铂(II)络合物,初步研究了铂(II)络合物的光物理性质,发现由于脲基的存在,当浓度大于3.32×10-5mol/L时铂(II)络合物能够发生分子间簇集.
