Reactions of Molecules with Two Equivalent Functional Groups. 5. Anomalous Reactivity of 1,10-Cyclooctadecanedione. Crystal and Molecular Structures of cis,cis-1,10-Diphenyl-1,10-cyclooctadiene and the Stereoisomers of 1,10-Diphenyl-1,10-cyclooctadecanediol(1)

The reaction of the title diketone (3) with phenyl Grignard produces (with rate constant k(1)) the conjugate base (6-M) of 10-hydroxy-10-phenylcyclooctadecanone (6), which is subsequently converted (with rate constant k(2)) to the conjugate base of the title diol, as a mixture of the cis (7, 55%) and trans (8, 45%) isomers. The ratio k(2)/k(1), 2.2 +/- 0.4, indicates that the carbonyl group in 6-M is 4.4 times as reactive as each carbonyl in 3. Competition experiments further demonstrate that the relative rates (per carbonyl) for addition of phenylmagnesium bromide to 3, 10-methylenecyclooctadecanone (11), and cyclopentadecanone (12) are 1.0:0.60:1.92. Possible reasons for this order of reactivity are discussed. Diols 7 and 8 undergo facile double dehydration to form the title diene 13, which is predicted by molecular mechanics calculations to be the most stable of the four possible symmetrical diene isomers. The structures of 7, 8, and 13 were secured by single-crystal X-ray studies.     

Using absolute and relative reasoning in the prediction of the potential metabolism of xenobiotics

To be useful, a system which predicts the metabolic fate of a chemical should predict the more likely metabolites rather than every possibility. Reasoning can be used to prioritize biotransformations, but a real biochemical domain is complex and cannot be fully defined in terms of the likelihood of events. This paper describes the combined use of two models for reasoning under uncertainty in a working system, METEOR-one model deals with absolute reasoning and the second with relative reasoning.     

Efficient syntheses of thiochromans via cationic cycloadditions

alpha-(Benzotriazolyl)methyl thioethers 1a-e reacted with styrenes under Lewis acid catalysis to give novel polysubstituted thiochromans (3,4-dihydro-2H-1-benzothiopyrans) 3-14 and 16-20 in generally high yields. Most thiochromans were isolated as one diastereomer following recrystallization. The configuration and conformation of the products are predicted on the basis of their NMR data. A stepwise reaction, proceeding via a [4(+) + 2] cationic polar cycloaddition mechanism, is proposed.

Proton relaxation enhancement by manganese(III)TPPS4 in a model tumor system

Managanese(III)tetraphenylporphine sulfonate [Mn(III)TPPS4] has been investigated as a tumor specific paramagnetic contrast agent for magnetic resonance imaging (MRI) of L1210 solid tumors in mice. Mn(III)TPPS4 was found to clear rapidly from the blood and concentrate in the kidneys, tumor and liver. Although relatively high ratios of tumor to normal tissues could be obtained (e.g., greater than 90 for tumor/muscle), the kidneys were found to have the highest concentration of the metalloporphyrin at all doses and time periods tested. A significant decrease in the longitudinal relaxation time was measured for excised tissues (kidney, tumor, liver, muscle) from mice that were treated with Mn(III)TPPS4. A linear correlation was observed between the longitudinal relaxation rate determined for L1210 tumor and the corresponding concentration of Mn(III)TPPS4 found at various injected doses and time intervals between the injection and analysis. A small animal radiofrequency receiver coil designed for use with a 0.15-T clinical imager was employed to evaluate the ability of Mn(III)TPPS4 to selectively increase the signal intensity of the implanted L1210 tumor. The images show a conspicuous enhancement in the contrast between the tumor and adjacent tissue upon treatment with this agent. The results indicate that Mn(III)TPPS4 is a useful prototype paramagnetic metalloporphyrin MRI contrast agent with a significant affinity for the L1210 tumor.

A 3-manifold group which is not four dimensional linear

We give examples of closed orientable graph 3-manifolds having a fundamental group which is not a subgroup of GL(4,F)$\mathit{GL}(4,\mathbb{F})$ for any field F$\mathbb{F}$. This answers a question in the Kirby problem list from 1977 which is credited to the late William Thurston.     

GaAs multiple quantum well microresonator modulators grown on silicon substrates

We report new results on the modulation characteristics of GaAs/AlGaAs asymmetric Fabry-Perot modulators grown on silicon substrates. We discuss factors affecting device performance and evaluate these by growing p-i-n quantum well diodes, and multilayer reflector stacks on silicon. Using data from these test structures we have designed an asymmetric microresonator modulator and achieve, experimentally, a 40% reflection change with only 5 V and a contrast ratio of 7.4 dB, also with 5 V.Formerly at:Philips Research Laboratories, Redhill, UK
