Analysis of peroxidase activity by native polyacrylamide gel electrophoresis (PAGE) from a garlic bulb (Allium sativum L) extract showed two major activities (designated POX1 and POX2). The POX2 isoenzyme was purified to homogeneity by ammonium sulfate precipitation, gel filtration, and cation-exchange chromatography. The purified enzyme was found to be monomeric with a molecular mass of 36.5 kDa, as determined by sodium dodecyl sulfate-PAGE. The optimum temperature ranged from 25 to 40 degrees C and optimum pH was about 5.0. The apparent Km values for guaiacol and H2O2 were 9.5 and 2 mM, respectively. POX2 appeared highly stable since 50% of its activity was conserved at 50 degrees C for 5 h. Moreover POX2 was stable over a pH range of 3.5-11.0. Immobilization of POX2 was achieved by covalent binding of the enzyme to an epoxy-Sepharose matrix. The immobilized enzyme showed great stability toward heat and storage when compared with soluble enzyme. These properties permit the use of this enzyme as a biosensor to detect H2O2 in some food components such as milk or its derivatives.  相似文献

The conversion of amorphous tricalcium phosphate with different hydration ratio into apatite in water at 25 °C has been studied by microcalorimetry and several physical-chemical methods. The hydrolytic transformation was dominated by two strong exothermic events. A fast, relatively weak, wetting process and a very slow but strong heat release assigned to a slow internal rehydration and the crystallization of the amorphous phase into an apatite. The exothermic phenomenon related to the rehydration exceeded the crystalline transformation enthalpy. Rehydration occurred before the conversion of the amorphous phase into apatite and determined the advancement of the hydrolytic reaction. The apatitic phases formed evolved slightly with time after their formation. The crystallinity increased whereas the amount of HPO42− ion decreased. These data allow a better understanding of the behavior of biomaterials involving amorphous phases such as hydroxyapatite plasma-sprayed coatings.  相似文献

We propose an optical circuit design method for coherent waves as a boundary value problem. The method produces a very compact circuit in which the refractive index pattern is automatically synthesized for given input and output fields with a numerical calculation. We employ the method to design a 1.3/1.55 microm wavelength demultiplexer and also describe the features of a circuit generated by use of the method.  相似文献

Metal chelates targeted to amyloid peptides are widely explored as diagnostic tools or therapeutic agents. The attachment of a metal complex to amyloid recognition units typically leads to a decrease in peptide affinity. We show here that by separating a macrocyclic GdL chelate and a PiB targeting unit with a long hydrophobic C10 linker, it is possible to attain nanomolar affinities for both Aβ1-40 (Kd=4.4 nm ) and amylin (Kd=4.5 nm ), implicated, respectively in Alzheimer's disease and diabetes. The Scatchard analysis of surface plasmon resonance data obtained for a series of amphiphilic, PiB derivative GdL complexes indicate that their Aβ1-40 or amylin binding affinity varies with their concentration, thus micellar aggregation state. The GdL chelates also affect peptide aggregation kinetics, as probed by thioflavin-T fluorescence assays. A 2D NMR study allowed identifying that the hydrophilic region of Aβ1-40 is involved in the interaction between the monomer peptide and the Gd3+ complex. Finally, ex vivo biodistribution experiments were conducted in healthy mice by using 111In labeled analogues. Their pancreatic uptake, ∼3 %ID g−1, is promising to envisage amylin imaging in diabetic animals.  相似文献

The chemical composition of ultrafine amorphous Fe−B powders prepared by a chemical reduction depends on the mixed molar ratio of KBH4 to Fe ions. We propose the following reaction processes for the formation of ultrafine Fe−B powders: (1) 4Fe2++2BH4−+6OH→2Fe2B+6H2O+H2 and (2) 4Fe2++2BH4−+7OH→2Fe3B+Fe+BO2+5H2O+5/2H2.  相似文献

Continuous cooling transformation (CCT) diagrams were successfully constructed for Zr65Al7.5Ni10Cu17.5? x Pd x (x?=?0???17.5) glass-forming alloys, comparing phase-transformation features in the alloy system to composition. While a low-Pd alloy (x?=?5) showed a single transformation curve, corresponding to the formation of a crystalline phase on the high-temperature side of the undercooled-liquid region, for a given time-scale, a high-Pd alloy (x?=?17.5) revealed an additional curve, corresponding to quasicrystalline phase formation on the lower temperature side. The result provides a clue to the structural and property control on the alloy system. Glassy specimens of the same size but with different intrinsic structure, evaluated by structural relaxation during continuous heating, could be fabricated for the low-Pd alloy (x?=?5). Plasticity was found to increase proportionally with the relaxation enthalpy. On the other hand, the critical size for glass formation could be improved considerably from 5 to 7 mm in diameter for the high-Pd alloy (x?=?17.5).  相似文献

An optical biosensor for the determination of catechol, a widely used yet toxic and carcinogenic molecule, is proposed using a crude extract of desert truffle (Terfezia leonis Tul.) as an enzymatic source of tyrosinase. The biosensor is constructed by the immobilization of tyrosinase crude extract in a bi-layered silica gel film prepared by dip-coating of an alkoxide/colloidal silica solution containing the enzyme on glass slide. Encapsulation has a moderate effect of the enzyme optimal pH stability but largely increases its thermal stability. Immobilized enzymes have a higher substrate affinity towards catechol but smaller maximum conversion velocity. The optical biosensor provides a linear response for catechol in the concentration range of 50–400?µM and a limit of detection was 52?µM. AFM studies show that the enzymes impact on the silica gel structure, preventing further deposition of additional layers. Comparison with similar dopamine biosensors points out that the impact of encapsulation on enzymatic activity may depend on the considered substrate.  相似文献
