Compact Dry TC qualifies as a rapid method kit for determining aerobic colony counts in foods. The plates are presterilized and contain culture medium and a cold-soluble gelling agent. The medium is rehydrated by inoculating 1 mL diluted sample into the center of the self-diffusible medium and allowing the solution to diffuse by capillary action. The plates can then be incubated and the colonies counted without any additional steps. The Compact Dry TC method was validated with 5 different raw meats. The performance tests were conducted at 35 degrees and 30 degrees C. In all required performance studies, no apparent differences were observed between the Compact Dry TC method and the Standard Pour Plate method (AOAC Official Method 966.23) for the detection level of aerobic microorganisms. For the accuracy claim (n = 60), a correlation factor of r2(35) = 0.9977 (35 degrees C) and r2(30) = 0.9932 (30 degrees C) could be assigned, as stated in the application for "Performance Tested Method." Quality consistency and storage robustness studies, showed no significant variations in plate count results with different production lots or plates of diverse storage age.  相似文献

Since strain localization is a precursor of failure, it is an important subject to address in the field of geomechanics. Strain localization has been analysed for geomaterials by several researchers. Many of the studies, however, treated the problems brought about by strain localization as two-dimensional problems, although the phenomena are generally three-dimensional. In the present study, undrained triaxial compression tests using rectangular specimens and their numerical simulation are conducted in order to investigate the strain localization behaviour of geomaterials under three-dimensional conditions. In the experiments, both normally consolidated and over-consolidated clay samples are tested with different strain rates. Using the distribution of shear strain obtained by an image analysis of digital photographs taken during deformation, the effects of the strain rates, the dilation, and the over-consolidation on strain localization are studied in detail. The analysis method used in the numerical simulation is a coupled fluid-structure finite element method. The method is based on the finite deformation theory, in which an elasto-viscoplastic model for water-saturated clay, which can consider structural changes, is adopted. The results of the simulation include not only the distribution of shear strain on the surfaces of the specimens, but also the distributions of strain, stress, and pore water pressure inside the specimens. Through a comparison of the experimental results and the simulation results, the mechanisms of strain localization are studied under three-dimensional conditions.  相似文献

Compact Dry CF is a ready-to-use test method for the enumeration of coliform bacteria in food. The plates are presterilized and contain culture medium and a cold water-soluble gelling agent. The medium should be rehydrated with 1 mL diluted sample inoculated into the center of the self-diffusible medium, allowing the solution to diffuse by capillary action. The plate can be incubated at 35 degrees C for 20-24 h and the colonies counted without any further working steps. The Compact Dry CF medium plates were validated with 5 different raw meats. The performance tests were conducted at 35 degrees C. In all studies performed, no apparent differences were observed between the Compact Dry CF method and the AOAC Official Method 966.24 results. For the accuracy claim (n = 75), a correlation factor of r2 = 0.91 (coliform) could be assigned, as stated in the application for Performance-Tested Method. No significant variations in coliform bacterial counts were observed with different production lots or plates of diverse storage age by the quality consistency and storage robustness studies.  相似文献

1:1 cyclic compounds 8a-c (51-55%) and 2:2 cyclic compounds 9a-c (20-49%) containing 1,4,7,10-tetraazacyclododecane (cyclen) and azobenzene units were selectively synthesized under UV irradiation (330 nm < lambda < 380 nm) and in the dark. Synthesis depended on the wavelength of irradiation light and the length of methylene chains of the linker between the cyclen and azobenzene units. A study of NMR and UV-vis spectra indicated that properties of 8a-c and 9a-c are closely related to their structural flexibility. Rate constants (k) and thermodynamic parameters (DeltaG(), DeltaH(), and DeltaS()) of 8a-c and 9a-c were studied in nonpolar media (benzene) and polar media (methanol). The cis to trans isomerization rates in the dark for these cyclic compounds increase with ring size or structural flexibility (8a < 8c < 8b < 9a < 9b < 9c). In principle, DeltaS() dominates DeltaG() in cyclic compounds.  相似文献

A novel biotinylated and enzyme‐immobilized nanobio device was prepared with heterobifunctional composite latex particles. Hemispherical poly(glycidyl methacrylate‐co‐divinylbenzene)/polystyrene [P(GMA‐DVB)/PSt] particles with epoxy and hydroxyl groups were prepared by soap‐free seeded emulsion polymerization with P(GMA‐DVB) seed particles. Hydroxyl groups were introduced to PSt chain terminals in the seeded stage by adding 2‐mercaptoethanol as a chain‐transfer agent. To obtain the desired hemispherical structure particles, we studied the effects of the preswelling process, the type and amount of solvents added in the seeded polymerization step, the weight ratio of the secondary monomer (styrene) to the seed particle (M/P), and the type of initiators. Under suitable conditions, heterobifunctional P(GMA‐DVB)/PSt was obtained, which was confirmed by observing the binding of streptavidin–colloidal gold with transmission electron microscopy (TEM). To obtain biotinylated and enzyme‐immobilized particles, 5‐(N‐succinimidyloxycarbonyl)pentyl D‐biotinamide was first reacted with the hydroxyl group on the PSt domain of the particle. Pyruvate kinase (PK) was then directly immobilized to the biotinylated particles through a reaction with the epoxy group in the PGMA domain. The monolayer of PK on the latex particle surface was considered to be formed by covalent binding. We design novel chromophoric amphiphiles 6a-c, which lead to pH-dependent membrane fusion of egg phosphatidylcholine (eggPC) liposome containing them. Lipids 6a-c comprise double alkyl chains, a single chain with a 2-nitrophenol group as a pH trigger, and dipeptide (Asp-Asp) between them. The pKa values of 2-nitrophenol groups of 6a-c in liposome are larger than that of hydrophilic compound 9 in an aqueous solution. Absorption spectra indicate that the fields around 2-nitrophenol of 6a-c situated in liposome membranes are more hydrophobic than that of 9 in an aqueous solution, whereas the environments around deprotonated 2-nitrophenolate of 6b and 6c are not so hydrophobic as that of 6a. This means that protonated 2-nitrophenol groups of 6a-c are embedded in bilayer membranes. Deprotonated 2-nitrophenol groups of 6b and 6c must be located in less hydrophobic circumstances, while that of 6a is still embedded in bilayer membranes because of its larger hydrophobicity. Absorption spectra and (1)H NMR spectra respectively suggest that protonated 2-nitrophenol groups of 6a and those of 6c might take face-to-face associations in bilayer membranes.  相似文献

History-dependent dimensional behavior of paper has been formulated within the framework of the general linear theory of viscoelasticity and the classical lamination theory. The effect of the drying history of the papermaking process was incorporated by introducing the residual stress in the reference configuration which was taken at the final drying stage in this study. This permits us to account for complex dimensional and form changes of paper in the converting and end use processes.In order to determine the prediction performance of the computer simulation model developed, in-plane and out-of-plane dimensional responses under cyclic humidity changes were predicted on the basis of hygroviscoelastic data of paper. The simulation examples demonstrated typical irreversible dimensional responses of paper both for the in-plane dimensional change and the curvature change (curl). The computer code can easily deal with the non-uniform distribution of 1) anisotropic viscoelastic properties, 2) hygrothermal properties, and 3) moisture and temperature through the thickness.  相似文献
