This work puts forth a reaction pathway for the reactivation of exogenous ligand inhibited H‐cluster, the active site of Fe‐only hydrogenases. The H‐cluster is a dimetal complex, Fe–Fe, with the metal centers bridged by di(thiomethyl)amine. Exogenous ligands, H2O, and OH?, are bound to the distal iron (Fed). Density functional theory (DFT) calculations on the native and ruthenium‐modified H‐cluster have been performed using the B3LYP functional with 6‐31+G** and 6‐311+G** basis sets. We have ascertained that there is a thermodynamically favorable pathway for the reactivation of the OH? inhibited H‐cluster, which proceeds by an initial protonation of the Fed–OH? complex. In the present study, a HPLC/DAD method was set up to allow for the determination and quantification of malondialdehyde (MDA) in the brain of rodents (rats). Chromatographic separation was achieved on Supelcosil LC-18 (3 μm) SUPELCO Column 3.3 cm × 4.6 mm and Supelco Column Saver 0.5 μm filter by using a mobile phase acetonitrile (A) and phosphate buffer (20 mM, pH = 6) (B). Isocratic elution was 14% for (A) and 86% for (B). The injection volume (loop mode) was 100 μL with an analysis time of 1.5 min. Flow rate was set at 1 mL/min. The eluted compound was detected at 532 nm by a DAD detector by keeping the column oven at room temperature. The results indicated that the method has good linearity in the range of 0.2–20 μg/g. Both intra- and inter-day precision, expressed as RSD, were ≤15% and the accuracies ranged between ±15%. The lower limit of quantification (LLOQ), stability, and robustness were evaluated and satisfied the validation criteria. The method was successfully applied in a study of chronic toxicology following different treatment regimens with haloperidol and metformin.  相似文献

The oxidation of H‐cluster in gas phase, and in aqueous enzyme phase, has been investigated by means of quantum mechanics (QM) and combined quantum mechanics–molecular mechanics (QM/MM). Several potential reaction pathways (in the above‐mentioned chemical environments) have been studied, wherein only the aqueous enzyme phase has been found to lead to an inhibited hydroxylated cluster. Specifically, the inhibitory process occurs at the distal iron (Fed) of the catalytic H‐cluster (which isalso the atom involved in H2 synthesis). The processes involved in the H‐cluster oxidative pathways are O2 binding, e? transfer, protonation, and H2O removal. We found that oxygen binding is nonspontaneous in gas phase, and spontaneous for aqueous enzyme phase where both Fe atoms have oxidation state II; however, it is spontaneous for the partially oxidized and reduced clusters in both phases. Hence, in the protein environment the hydroxylated H‐cluster is obtained by means of completely exergonic reaction pathway starting with proton transfer. A unifying endeavor has been carried out for the purpose of understanding the thermodynamic results vis‐à‐vis several other performed electronic structural methods, such as frontier molecular orbitals (FMO), natural bond orbital partial charges (NBO), and H‐cluster geometrical analysis. An interesting result of the FMO examination (for gas phase) is that an e? is transferred to LUMOα rather than to SOMOβ, which is unexpected because SOMOβ usually resides in a lower energy rather than LUMOα for open‐shell clusters. © 2008 Wiley Periodicals, Inc. The Accelerator mass spectrometry (AMS) analysing method is described with applications for the detection of tritium leaks, detection of pollution and for research in the fusion experiments. The special requirements for an experiment of AMS with tritium are also presented. This paper concludes by showing the possibilities for each AMS facility to perform applications for detecting and preventing the environment pollution with tritium.  相似文献

Three new complexes with formula [VOL2xH2O [(1) HL: N,N-dimethylbiguanide, x?=?0.5; (2) HL: 1-phenylbiguanide, x?=?0; (3) HL: 1-(o-tolyl)biguanide, x?=?0.5] were synthesised and characterised. The IR and UV?CVis spectral data indicate that these biguanide derivatives act as bidentate chelating anionic ligands and generate VO(II) complexes with a square?Cpyramidal stereochemistry. The thermal analysis (TG, DTA) elucidated the composition and also the number and nature of the water molecules.  相似文献

Increasing interest on sustainable agriculture has led to the development of new materials which can be used as seed coating agents. In this study, a new material was developed based on gelatin film reinforced with cellulose nanocrystals (CNC) which was further used as nanocomposite matrix for Trichoderma harzianum KUEN 1585 spores. The nanocomposite films were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and scanning electron microscopy (SEM), showing the formation of new hydrogen bonds between the components with a good compatibility between them. Measurements of water contact angles and tests of water vapor sorption and swelling degree revealed an improvement in the water vapor absorption properties of the films as a result of their reinforcement with CNC. Furthermore, by adding the Trichoderma harzianum KUEN 1585 spp. in the seed coating material, the germination percentage, speed of germination and roots length of the corn seeds improved. The polymeric coating did not inhibit the growth of T. harzianum KUEN 1585, with this material being a good candidate in modern agriculture.  相似文献

This paper considers the connections between the local extrema of a function f:DR and the local extrema of the restrictions of f to specific subsets of D. In particular, such subsets may be parametrized curves, integral manifolds of a Pfaff system, Pfaff inequations. The paper shows the existence of C 1 or C 2-curves containing a given sequence of points. Such curves are then exploited to establish the connections between the local extrema of f and the local extrema of f constrained by the family of C 1 or C 2-curves. Surprisingly, what is true for C 1-curves fails to be true in part for C 2-curves. Sufficient conditions are given for a point to be a global minimum point of a convex function with respect to a family of curves.  相似文献

This study aimed to develop a HPLC/DAD method in order to determine and quantify the reduced glutathione (GSH) and oxidized glutathione (GSSG) levels in rat brain. Due to the presence of the thiol group (-SH), GSH can interact with the Ellman′s reagent (DTNB), with which it forms a reaction product through which the level of GSH can be quantified, using the DAD detection system. Chromatographic separation was achieved after a derivatization process by using a mobile phase acetonitrile (A) and phosphate buffer (20 mM, pH = 2.5) (B). The compounds of interest were detected at 330 nm using a chromatographic C8 column. The method of determination met the validation criteria, specified by the regulatory bodies. The applicability of the method was demonstrated in a chronic toxicology study of central nervous system (CNS), following different treatment regimens with haloperidol.  相似文献
