In order to gain direct evidence for lipid-dependent protein conformation in membrane, effects of modification of lipid composition on mobility of spin-labeled cysteine residues were investigated in the plasma membrane of the yeast Saccharomyces cerevisiae. Conversion of the bulk of phospholipids to diglycerides by treatment of the membrane with phospholipase C substantially enhanced spectral anisotropy. However, alterations of the viscosity of the lipid-bilayer by enriching the membrane with palmitelaidic or oleic acid had no effect on mobility of spin-labeled cysteine residues. These observations indicate that while the spin-labeled residues are not in direct contact with the lipid core of the membrane, there are lipid-protein interactions to the extent that removal of the polar portion of the bulk of phospholipids induces conformational changes in proteins, which in turn restrict mobility of these residues. It is concluded that conformation of membrane proteins on lipid structure and that phospholipids have a role in preserving the native conformation of proteins. 相似文献

In this paper, a lattice Boltzmann method is employed to simulate the conjugate radiation–forced convection heat transfer in a porous medium. The absorbing, emitting, and scattering phenomena are fully included in the model. The effects of different parameters of a silicon carbide porous medium including porosity, pore size, conduction–radiation ratio, extinction coefficient and kinematic viscosity ratio on the temperature and velocity distributions are investigated. The convergence times of modified and regular LBMs for this problem are 15 s and 94 s, respectively, indicating a considerable reduction in the solution time through using the modified LBM. Further, the thermal plume formed behind the porous cylinder elongates as the porosity and pore size increase. This result reveals that the thermal penetration of the porous cylinder increases with increasing the porosity and pore size. Finally, the mean temperature at the channel output increases by about 22% as the extinction coefficient of fluid increases in the range of 0–0.03.

The present study investigates the synthesis and effectiveness of gold/gelatin nanoparticles (NPs) biopolymer as a carrier for methotrexate (MTX) drug. Two different shapes of gold particles, including spherical AuNPs (50 & 100 nm) and gold nanorods (AuNRs) with three different sizes (20, 50 and 100 nm length) were synthesized using the chemical reduction method. The effect of AuNPs size and shape on the entrapment efficiency (E.E), the release rate of the drug, and cellular uptake were investigated. The surfaces of both AuNPs and AuNRs were coated with a gelatin biopolymer, and the stability and property of the generated compounds were studied. Moreover, MTX as a chemotherapeutic agent was loaded on the gelatin-coated AuNPs/AuNRs complexes. The physicochemical properties of the gelatin-coated AuNPs/AuNRs complexes were studied using ultraviolet-visible (UV–Vis) spectroscopy, dynamic light scattering (DLS), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and Fourier transform infrared (FT-IR) spectroscopy. The E.E and MTX release behavior from the complexes at pH values of 7.4 and 5.4 and temperatures of 37 and 40 °C were investigated in vitro. The cytotoxic effects of AuNPs, AuNPs-Gelatin, AuNPs-Gelatin-MTX, AuNRs, AuNRs-Gelatin, AuNRs-Gelatin-MTX and free MTX were studied. The results indicated that the E.E of AuNPs was higher than that of AuNRs. The highest release rate of the drug was related to the AuNR1-gelatin complex (pH 5.4 and temperature of 40 °C). In addition, MTX loaded AuNR2-gelatin showed the highest cytotoxic effect on the MCF-7 breast cancer cell line so that even its cell cytotoxicity was more than that of the free drug. 相似文献

In this paper, we will first study the existence and uniqueness of the solution of a two-dimensional inverse heat conduction problem (IHCP) which is severely ill-posed, i.e., the solution does not depend continuously on the data. We propose a stable numerical approach based on the finite-difference method and the least-squares scheme to solve this problem in the presence of noisy data. We prove the convergence of the numerical solution, then to regularize the resultant ill-conditioned linear system of equations, we apply the Tikhonov regularization 0th, 1st and 2nd method to obtain the stable numerical approximation to the solution. The stability and accuracy of the scheme presented is evaluated by comparison with the Singular Value Decomposition (SVD) method. 相似文献
