Development of the epicardium is critical to proper heart formation. It provides all of the precursor cells that form the coronary system and supplies signals that stimulate cardiac myocyte proliferation. The epicardium forms from mesothelial cells associated with the septum transversum and is referred to as the proepicardium (PE). Two different methods by which these PE cells colonize the developing heart have been described. In avians, PE cells form a bridge to the heart over which PE cells migrate onto the heart. In fish and mammals, PE cells form vesicles of cells that detach from the mesothelium, float through the pericardial cavity, and attach to the heart. A previous study of rat PE development investigated this process at the histological level. Protein markers have been developed since this study. Thus, we investigated this important developmental process coupled with these new markers using other visualization techniques such as scanning electron microscopy (SEM) and confocal microscopy. Finally, a novel, three-dimensional (3-D) culture system was used to confirm the identity of the PE cells. In this study, we found convincing evidence that the rat PE cells directly attach to the heart in a manner similar to that observed in avians.  相似文献

A new ion trap scan function for gas chromatography/mass spectrometry (GC/MS) quantitation is described that employs alternating mass-selective storage (rf/dc isolation) of ions from an analyte and its coeluting isotopically labeled internal standard. This scan includes two separate ionization/isolation/mass analysis sequences within the same scan function, each optimized for either the analyte or the internal standard. This results in alternating between analyzing the analyte and the internal standard during their coelution. The method is conceptually similar to using two different scan functions to analyze either the analyte or the internal standard in alternating scans; however, it is much faster because it eliminates the slow procedure of continuously downloading alternating scan functions from disk. This allows more data points to be obtained over a GC peak, resulting in more reproducible GC peak profiles as well as better sensitivity and precision. Results of calibration curves spanning four orders of magnitude (0.5 pg to 5 pg injected on column) obtained by using this method give excellent linear correlations (r 2 > 0.9990) and precision (relative standard deviations of triplicate injections < 10%).  相似文献

Masking-level differences (MLDs) were measured for trains of 2000-Hz bandpass clicks as a function of the interclick interval (ICI) and the number of clicks in the train. The magnitude of the MLD grew as the number of clicks in the train was increased from 1 to 32. While the MLDs tended to be larger at longer ICIs, the effect was mediated by changes in detectability in the homophasic conditions. For click trains consisting of 4-32 clicks, the improvement in detectability in the antiphasic conditions with increases in the number of clicks appears to be the result of integration of acoustic power, as is the case for the homophasic conditions. The absence of MLDs for short trains of high-frequency transients remains quite puzzling, since large MLDs are found with single, low-frequency transients.  相似文献
