Interferons are potent biologically active proteins synthesized and secreted by somatic cells of all mammalian species. Dye-affinity adsorption is increasingly used for protein separation. Hollow fibers have several advantages as adsorbents in comparison to conventional bead supports because they are not compressible and they eliminate internal diffusion limitations. The aim of this study was to explore in detail the performance of polyamide hollow fibers to which Cibacron Blue F3GA was attached for adsorption of recombinant interferon-α (rHuIFN-α). The hollow fiber was characterized by scanning electron microscopy. These dye-carrying hollow fibers (35.8 μmol/g) were used in the rHuIFN-α adsorption-elution studies. The effects of initial concentration of rHuIFN-α, medium pH, ionic strength and temperature on the adsorption efficiency of dye-attached hollow fibers were studied in a batch system. The non-specific adsorption of rHuIFN-α on the hollow fibers was 1.2 mg/g. Cibacron Blue F3GA attachment significantly increased the rHuIFN-α adsorption up to 99.8 mg/g. Significant amount of the adsorbed rHuIFN-α (up to 94.8%) was eluted in 1 h in the elution medium containing 1.0 M NaCl. In order to determine the effects of adsorption conditions on possible conformational changes of rHuIFN-α structure, fluorescence spectrophotometry was employed. We resulted that dye-affinity hollow fibers can be applied for rHuIFN-α adsorption without causing any significant conformational changes. Repeated adsorption/elution processes showed that these dye-attached hollow fibers are suitable for rHuIFN-α adsorption. 相似文献

Concentrations of fluoroquinolones, which are used in the treatment of many bacterial infections, should be monitored in biological fluids as they exhibit concentration-dependent bactericidal activity. In this study, a liquid chromatography method for the determination of levofloxacin, ciprofloxacin, moxifloxacin and gemifloxacin in human urine and plasma was developed for the first time. The efficiency of five different columns for the separation of these fluoroquinolones was compared. Experimental parameters that affect the separation, such as percentage of organic solvent, pH, temperature, gradient shape and detector wavelength, were optimized by a step-by-step approach. Using a pentafluorophenyl core–shell column (100 × 4.6 mm, 2.7 μm), the separation of four analytes was accomplished in <7.5 min. The developed method was validated for the determination of analytes in both urine and plasma with respect to sensitivity, specificity, linearity (r ≥ 0.9989), recovery (79.46–102.69%), accuracy, precision and stability (85.79–111.07%). The intra- and inter-day accuracies were within 89.55–111.94% with relative standard deviations of 0.35–8.05%. The feasibility of method was demonstrated by analyzing urine and plasma samples of patients orally receiving levofloxacin, ciprofloxacin or moxifloxacin. The developed method is suitable for therapeutic drug monitoring of these fluoroquinolones and can be applied to pharmacokinetic and toxicological studies. 相似文献

Metal-chelating affinity beads have attracted increasing interest in recent years for protein purification. In this study, iminodiacetic acid (IDA) was covalently attached to the poly(glycidyl methacrylate) [PGMA] beads (1.6 μm in diameter). Cu(2+) ions were chelated via IDA groups on PGMA beads for affinity binding of hemoglobin (Hb) from human blood hemolysate. The PGMA beads were characterized by scanning electron microscopy (SEM). The PGMA-Cu(2+) beads (628 μmol/g) were used in the Hb binding-elution studies. The effects of Hb concentration, pH and temperature on the binding efficiency of PGMA-Cu(2+) beads were performed in a batch system. Non-specific binding of Hb to PGMA beads in the absence of Cu(2+) ions was very low (0.39 mg/g). The maximum Hb binding was 130.3 mg/g. The equilibrium Hb binding increased with increasing temperature. The negative change in Gibbs free energy (ΔG°<0) indicated that the binding of Hb on the PGMA-Cu(2+) beads was a thermodynamically favorable process. The ΔS and ΔH values were 102.2 J/mol K and -2.02 kJ/mol, respectively. Significant amount of the bound Hb (up to 95.8%) was eluted in the elution medium containing 1.0 M NaCl in 1 h. The binding followed Langmuir isotherm model with monolayer binding capacity of 80.3-135.7 mg/g. Consecutive binding-elution experiments showed that the PGMA-Cu(2+) beads can be reused almost without any loss in the Hb binding capacity. To test the efficiency of Hb depletion from blood hemolysate, eluted portion was analyzed by fast protein liquid chromatography. The depletion efficiency for Hb was above 97.5%. This study determined that the PGMA-Cu(2+) beads had a superior binding capacity for Hb compared to the other carriers within this study. 相似文献
