The zeta-potentials of silica, copper, platinum and gold particles have been measured as a function of pH. The isoelectric points were found to be at pH 3.0, 5.8, 3.0 and 3.5, respectively. In the pH range 3.0 to 5.8 copper and silica particles are oppositely charged and accordingly the coating of silica with copper particles could be demonstrated. In the case of gold and platinum the sign of the charge is such that direct adhesion to silica particles cannot be expected and this was also demonstrated in the case of platinum.  相似文献

Radial electric fields (E r) and their role in the establishment of edge transport barriers and improved confinement have been studied in the tokamaks TEXTOR-94 and CASTOR, where E r is externally applied to the plasma in a controlled way using a biased electrode, as well as in the tokamak T-10 where an edge transport barrier (H-mode) is obtained during electron-cyclotron resonance heating (ECRH) of the plasma.The physics of radial currents was studied and the radial conductivity in the edge of TEXTOR-94 (R = 1.75 m, a = 0.46 m) was found to be dominated by recycling (ion-neutral collisions) at the last closed flux surface (LCFS) and by parallel viscosity inside the LCFS. From a performance point of view (edge engineering), such electrode biasing was shown to induce a particle transport barrier, a reduction of particle transport, and a concomitant increase in energy confinement. An H-mode-like behaviour can be induced both with positive and negative electric fields. Positive as well as negative electric fields were shown to strongly affect the exhaust of hydrogen, helium, and impurities, not only in the H-mode-like regime.The impact of sheared radial electric fields on turbulent structures and flows at the plasma edge is investigated on the CASTOR tokamak (R = 0.4 m, a = 0.085 m). A non-intrusive biasing scheme that we call "separatrix biasing" is applied whereby the electrode is located in the scrape-off layer (SOL) with its tip just touching the LCFS. There is evidence of strongly sheared radial electric field and E×B flow, resulting in the formation of a transport barrier at the separatrix. Advanced probe diagnosis of the edge region has shown that the E×B shear rate that arises during separatrix biasing is larger than for standard edge plasma biasing. The plasma flows, especially the poloidal E×B drift velocity, are strongly modified in the sheared region, reaching Mach numbers as high as half the sound speed. The corresponding shear rates ( 5×106 s-1) derived from both the flow and electric field profiles are in excellent agreement and are at least an order of magnitude higher than the growth rate of unstable turbulent modes as estimated from fluctuation measurements.During ECRH in the tokamak T-10 (R = 1.5 m, a = 0.3 m), a regime of improved confinement is obtained with features resembling those in the H-mode in other tokamaks. Using a heavy ion beam probe, a narrow potential well is observed near the limiter together with the typical features of the L-H transition. The time evolution of the plasma profiles during L-H and H-L transitions is clearly correlated with that of the density profile and the formation of a transport barrier near the limiter. The edge electric field is initially positive after the onset of ECRH. It changes its sign during the L-H transition and grows till a steady condition is reached. Similar to the biasing experiments in TEXTOR-94 and CASTOR, the experimentally observed transport barrier is a barrier for particles.  相似文献

Atomic force microscopy (AFM) has been employed to observe in real-time and in an aqueous environment the process of ethidium bromide induced supercoiling in individual DNA plasmid molecules. Image data reveal both the onset and the progressive presence of plectonemic DNA supercoiling. In addition, significant molecular motion of the surface adsorbed DNA is observed. These data illustrate the potential of AFM in the time-resolved study of biomolecular processes, and hence, provide new insights into biomolecular structure and function.  相似文献

Poly[2-(dimethylamino)ethyl methacrylate-b-2-methacryloyloxyethyl phosphorylcholine] (DMA-MPC) is currently under investigation as a new vector candidate for gene therapy. The DMA block has been previously demonstrated to condense DNA effectively. The MPC block contains a phosphorylcholine (PC) headgroup, which can be found naturally in the outside of the cell membrane. This PC-based polymer is extremely hydrophilic and acts as a biocompatible steric stabilizer. In this study, we assess in detail the morphologies of DNA complexes obtained using the diblock copolymer series DMA(x)MPC30 (where the mean degree of polymerization of the MPC block was fixed at 30 and the DMA block length was systematically varied) using transmission electron microscopy (TEM) and liquid atomic force microscopy (AFM). Both techniques indicate more compact complex morphologies (more efficient condensation) as the length of the cationic DMA block increases. However, the detailed morphologies of the DMA(x)MPC30-DNA complexes observed by TEM in vacuo and by AFM in aqueous medium are different. This phenomena is believed to be related to the highly hydrophilic nature of the MPC block. TEM studies revealed that the morphology of the complexes changes from loosely condensed structures to highly condensed rods, toroids, and oval-shaped particles as the DMA moiety increases. In contrast, morphological changes from plectonemic loops to flower-like and rectangular block-like structures, with an increase in highly condensed central regions, are observed by in situ AFM studies. The relative population of each structure is clearly dependent on the polymer molecular composition. Enzymatic degradation assays revealed that only the DMA homopolymer provided effective DNA protection against DNase I degradation, while other highly condensed copolymer complexes, as judged from TEM and gel electrophoresis, only partially protected the DNA. However, AFM images indicated that the same highly condensed complexes have less condensed regions, which we believe to be the initiation sites for enzymatic attack. This indicates that the open structures observed by AFM of the DNA complexation by the DMA(x)MPC30 copolymer series are closer to in vivo morphology when compared to TEM.  相似文献

Background  

Microglia provide continuous immune surveillance of the CNS and upon activation rapidly change phenotype to express receptors that respond to chemoattractants during CNS damage or infection. These activated microglia undergo directed migration towards affected tissue. Importantly, the molecular species of chemoattractant encountered determines if microglia respond with pro- or anti-inflammatory behaviour, yet the signaling molecules that trigger migration remain poorly understood. The endogenous cannabinoid system regulates microglial migration via CB2 receptors and an as yet unidentified GPCR termed the 'abnormal cannabidiol' (Abn-CBD) receptor. Abn-CBD is a synthetic isomer of the phytocannabinoid cannabidiol (CBD) and is inactive at CB1 or CB2 receptors, but functions as a selective agonist at this Gi/o-coupled GPCR. N-arachidonoyl glycine (NAGly) is an endogenous metabolite of the endocannabinoid anandamide and acts as an efficacious agonist at GPR18. Here, we investigate the relationship between NAGly, Abn-CBD, the unidentified 'Abn-CBD' receptor, GPR18, and BV-2 microglial migration.  相似文献
