Automated sample cleanup in HPLC using column-switching techniques

Sample pretreatment has often been the rate limiting step in analyses by HPLC. Multicolumn or column switching techniques now available have made efficient automated cleanup procedures possible, with high sample throughput and analytical precision.     

Asymmetric Synthesis of 3-Hydroxyprolines by Photocyclization of N-(2-Benzoylethyl)glycinamides

The chiral N-(2-benzoylethyl)-N-tosylglycinamides 1a-c were prepared from the C₂-symmetric pyrrolidines 5a-c . Irradiation of these ketones 1a-c gave cis-3-hydroxyprolinamides 10-12 in moderate to good yields (Scheme 3). The de of the photocyclizations depended on the size of the substituents in positions C(2) and C(5) of the chiral pyrrolidine auxiliaries. In addition, the de varied with the reaction temperature, allowing the determination of activation-parameter differences. The structure of products 10-12 were established by NMR and X-ray analyses.     

Purification of DNA-derived deoxynucleotides from leukocytes involving nuclease elution of an ion-exchange column

A method has been developed in which the DNA of leukocytes (as the buffy coat from blood) is isolated in the form of its constituent deoxynucleotides. The steps in this method are as follows: (1) lyse the leukocytes with sodium dodecyl sulfate (SDS) and enzymatically digest the proteins and RNA, (2) remove the SDS on a non-polar adsorbent (Bio-Beads SM-4) and then trap the DNA on a quaternary amine silica cartridge, (3) wash the column with 1 M NaCl-buffer, (4) digest the DNA on the column with staphylococcal nuclease and (5) elute the digested DNA with 0.5 M NaCl-buffer and digest it further with bovine spleen phosphodiesterase II to deoxynucleotide-3′-monophosphates. From a 40-μl sample of butty coat was obtained 126 ± 14 μg (two experiments, eight sample total) of deoxynucleotides. Reversed-phase high-performance liquid chromatography, which removed the added enzymes, showed only peaks for deoxynucleotides. For comparison, the amount of deoxynucleotides obtained from the leukocytes by an automated phenol extraction procedure was 101 ± 5.4 μg (one experiment in triplicate).     

Diastereoselectivity of the mercuration of acyclic allylic alcohols

The diastereoselectivity of the mercuration of acyclic alkenes $\text{4}$ can be reversed by changing the substituent in the allylic position; with alcohols the erythro isomers $\text{5}$ and with esters or hemiacetals the threo isomers $\text{6}$ and $\text{8}$ are formed predominantly (Table I).     

Sequentially photocleavable protecting groups in solid-phase synthesis

[reaction: see text] A sequential solid-phase peptide synthesis was developed using both photolabile linker and protecting groups. The chromatic sequential lability between a tert-butyl ketone-derived linker (sensitive to irradiation at 305 nm) and a nitroveratryloxycarbonyl (NVOC) group (sensitive at 360 nm) was exploited to prepare Leu-Enkephalin in a 55% overall yield. This new strategy allows the preparation of peptides in essentially neutral medium, by avoiding the use of common deprotection reagents such as trifluoroacetic acid or piperidine.     

Complete basis set extrapolated potential energy, dipole, and polarizability surfaces of alkali halide ion-neutral weakly avoided crossings with and without applied electric fields

Complete basis set extrapolations of alkali halide (LiF, LiCl, NaF, NaCl) energy, dipole, and polarizability surfaces are performed with and without applied fields along the internuclear axis using state-averaged multireference configuration interaction. Comparison between properties (equilibrium separation, dissociation energy, crossing distance, diabatic coupling constant, dipole, and polarizability) derived from the extrapolated potential energy (or dipole) surfaces are made with those obtained from direct extrapolation from the basis set trends. The two extrapolation procedures are generally found to agree well for these systems. Crossing distances from this work are compared to those of previous work and values obtained from the Rittner potential. Complete basis set extrapolated crossing distances agree well with those derived from the Rittner potential for LiF, but were significantly larger for LiCl, NaF, and NaCl. The results presented here serve as an important set of benchmark data for the development of new-generation many-body force fields that are able to model charge transfer.
