Background
The common event in transmissible spongiform encephalopathies (TSEs) or prion diseases is the conversion of host-encoded protease sensitive cellular prion protein (PrPC) into strain dependent isoforms of scrapie associated protease resistant isoform (PrPSc) of prion protein (PrP). These processes are determined by similarities as well as strain dependent variations in the PrP structure. Selective self-interaction between PrP molecules is the most probable basis for initiation of these processes, potentially influenced by chaperone molecules, however the mechanisms behind these processes are far from understood. We previously determined that polymorphisms do not affect initial PrPC to PrPSc binding but rather modulate a subsequent step in the conversion process. Determining possible sites of self-interaction could elucidate which amino acid(s) or amino acid sequences contribute to binding and further conversion into other isoforms. To this end, ovine – and bovine PrP peptide-arrays consisting of 15-mer overlapping peptides were probed with recombinant sheep PrPC fused to maltose binding protein (MBP-PrP). 相似文献2.

Atropisomeric biaryls carrying ortho‐hydroxymethyl and formyl groups were made enantioselectively by desymmetrisation of dialdehyde or diol substrates. The oxidation of the symmetrical diol substrates was achieved using a variant of galactose oxidase (GOase), and the reduction of the dialdehydes using a panel of ketoreductases. Either M or P enantiomers of the products could be formed, with absolute configurations assigned by time‐dependent DFT calculations of circular dichroism spectra. The differing selectivities observed with different biaryl structures offer an insight into the detailed structure of the active site of the GOase enzyme. 相似文献

Metal/superconductor/semiconductor (Ag/Hg-1212/CdSe) hetero-nanostructures have been fabricated using pulse-electrodeposition
technique and are characterized by X-ray diffraction (XRD), full-width at half-maximum (FWHM) and scanning electron microscopy
(SEM) studies. The junction capacitance of Ag/Hg-1212, Hg-1212/CdSe and Ag/Hg-1212/CdSe heterojunctions is measured in dark
and under laser irradiation at room temperature. The nature of the junction formed and built-in-junction potentials were determined.
The increase in carrier concentration across the junction due to photo-irradiation has been observed. 相似文献

Formation of magnetite nanocrystals by magnetotactic bacteria is controlled by specific proteins which regulate the particles’ nucleation and growth. One such protein is Mms6. This small, amphiphilic protein can self‐assemble and bind ferric ions to aid in magnetite formation. To understand the role of Mms6 during in vitro iron oxide precipitation we have performed in situ pH titrations. We find Mms6 has little effect during ferric salt precipitation, but exerts greatest influence during the incorporation of ferrous ions and conversion of this salt to mixed‐valence iron minerals, suggesting Mms6 has a hitherto unrecorded ferrous iron interacting property which promotes the formation of magnetite in ferrous‐rich solutions. We show ferrous binding to the DEEVE motif within the C‐terminal region of Mms6 by NMR spectroscopy, and model these binding events using molecular simulations. We conclude that Mms6 functions as a magnetite nucleating protein under conditions where ferrous ions predominate. 相似文献

A new series of neutral organometallic building blocks based on piano-stool ruthenium(II) complexes, RuCl2(p-cymene)Ph2PCH2Y [Y = -NHC6H4(2-CO2H) (2a), -NHC6H4(3-CO2H) (2b), -NHC6H3(3-CO2H)(6-OCH3) (2c), -NHC6H4(4-CO2H) (2d), -NHC6H3(2-CO2H)(4-OH) (2e), -NHC6H3(3-OH)(4-CO2H) (2f), -NHC6H3(2-CO2H)(5-CO2H) (2g) and -OH (2h)], were synthesised in high yields (>88%) from {RuCl2(p-cymene)}2 and the appropriate phosphines 1a-1h. The new tertiary phosphine 1b was prepared by Mannich condensation of NH2C6H4(3-CO2H) with Ph2PCH2OH in MeOH. Solution NMR (31P{1H}, 1H), FT-IR and microanalytical data are in full agreement with the proposed structures. Single crystal X-ray studies confirm that, in each case, compounds 2a, 2b and 2d-2h have piano-stool arrangements with typical Ru-P, Ru-Cl and Ru-Ccentroid bond lengths. From our crystallographic studies, factors that influence the supramolecular assemblies of these ruthenium(II) complexes include: (i) the type of functional group present, (ii) the geometric disposition of the -N(H)CH2PPh2, -CO2H and -OH groups around the central benzene scaffold, and (iii) the solvents used in the recrystallisations. Hence in isomers 2a and 2b, molecules are associated into head-to-tail dimer pairs through classical intermolecular O-H?O hydrogen bonding. This feature is also observed in isomer 2d but dimer pairs are further associated to give a 1-D chain through assisted intermolecular N-H?Cl hydrogen bonding. The additional 4-hydroxo group in 2e promotes a ladder arrangement via intermolecular O-H?O and O-H?Cl hydrogen bonding. In contrast the isomeric compound 2f does not show head-to-tail O-H?O hydrogen bonding but instead O-H?Cl and N-H?O intermolecular hydrogen bonding is observed. Depending on the choice of solvent (MeOH or DMSO), 2g forms extended networks based on chains (2g · DMSO · 1.5MeOH) or tapes (2g · 3MeOH). In 2h, a single intramolecular O-H?Cl hydrogen bond is observed for each independent molecule. The X-ray structure of one representative tertiary phosphine, 1f, has also been determined. 相似文献

Despite the burgeoning interest in the various biological functions and consequent therapeutic potential of the vast number of oligosaccharides found in nature on glycoproteins and cell surfaces, the development of combinatorial carbohydrate chemistry has not progressed as rapidly as expected. The reason for this imbalance is rooted in the difficulty of oligosaccharide assembly and analysis that renders synthesis a rather cumbersome endeavor. Parallel approaches that generate series of analogous compounds rather than real libraries have therefore typically been used. Since generally low affinity is obtained for interactions between carbohydrate receptors and modified oligosaccharides designed as mimetics of natural carbohydrate ligands, glycopeptides have been explored as alternative mimics. Glycopeptides have been proven in many cases to be superior ligands with higher affinity for a receptor than the natural carbohydrate ligand. High-affinity glycopeptide ligands have been found for several types of receptors including the E-, P-, and L-selectins, toxins, glycohydrolases, bacterial adhesins, and the mannose-6-phosphate receptor. Furthermore, the assembly of glycopeptides is considerably more facile than that of oligosaccharides and the process can be adapted to combinatorial synthesis with either glycosylated amino acid building blocks or by direct glycosylation of peptide templates. The application of the split and combine approach using ladder synthesis has allowed the generation of very large numbers of compounds which could be analyzed and screened for binding of receptors on solid phase. This powerful technique can be used generally for the identification and analysis of the complex interaction between the carbohydrates and their receptors. 相似文献
