The interaction of native and derivatized transferrin with lecithin, cholesterol and their mixtures was studied using monomolecular layers of phospholipids. Moreover, the thermodynamic parameters corresponding to these interactions: the mixing excess free energy, interaction energy, and interaction parameter were also calculated.The ability of these two proteins to insert into monolayers was studied working at constant area.The formation of aggregates or secondary structures in solution was determined by the fluorescence and polarization values of ANS (anilinonaphthalene sulfonate), measured at different protein/ANS relationships.Abbreviations TF Transferrin - DTF derivatized transferrin - PC lecithin - CHOL cholesterol - CER ceramides - ANS anilinonaphthalene sulfonate - TNBS trinitro benzenesulfonate method  相似文献

For determination of phosphorus in samples of vegetable origin (leaves and wheat flour) decomposition by means of molten alkali is proposed. Two procedures have been studied, one with dry reagents including an auxiliary oxidant, and the other with initially moist alkali with no extra oxidant. The fusion product is easily soluble and produces a solution suitable for elemental analysis. The phosphorus is determined by the molybdenum blue method. The suggested mineralization is rapid, accurate and precise.  相似文献

The covalent conjugation of a 20-mer peptide belonging to the VP3 capsid protein of hepatitis A virus to the surface of preformed liposomes was investigated. Three different bonds (disulfide, thioether and amide) were established between the peptide sequence and liposomes bearing at their surface appropriate reactive groups. The effect of the relative concentration of the N-[4-(p-maleimidophenyl)butyryl]dipalmitoylphosphatidylethanolamine anchor in liposomes on stability during coupling of the peptide sequence was studied. The interaction of the three liposomal preparations with phospholipids in a biomembrane model system, monolayers at the air-water interface, is also reported. The results showed that although the peptides associate with liposomes in similar yields for the three strategies studied, differences can be observed when their interaction with phospholipid monolayers composed of dipalmitoylphosphatidylcholine is analysed.  相似文献

This paper is a study of the analytical capacity of Fourier transform infrared spectroscopy (FT-IR) for any type of samples from works of art. The analytical information obtained with this technique on organic and inorganic compounds is extremely useful in the preliminary studies necessary for diagnosis and to decide on the conservation process. This paper reports the analysis and study with FT-IR on samples extracted from the microlayers in several works of art from different periods (16th to 18th century) comprising wall paintings and canvas. The most outstanding of these being the frescoes by A. Palomino from two ceilings in the Santos Juanes church. The analytical procedures for the different components of the works studied such as

1. (a) varnish applied to wood panels and canvas paintings,

2. (b) binding media and pigments used by the artists,

3. (c) inerts, mortars, stuccos and grounds have been optimized. The FT-IR technique offers a quick analysis of microsamples (less than 0.5 mg) and is able to characterise the different molecular groups which provide information on the nature of the different materials of organic and inorganic origin used by the artist and thus permits the diagnosis of pathologies requiring conservation treatment.

A method for the simultaneous spectrophotometric determination of calcium and magnesium in mineral waters with an FIA system is tested. The method is based on the reaction between the analytes and arsenazo(III) at pH 8.5. The calculations of the amounts of both analytes in the samples are carried out with the H-point standard addition method (HPSAM) for ternary mixtures, and with a partial least squares (PLS) model after a proper variable selection. The results obtained for the determination of calcium were comparable using both methods. The employment of the HPSAM brings to our attention the influence of the calcium concentration in the sample to the development of the reaction between magnesium and arsenazo(III). HPSAM also permits to estimate the concentration of magnesium in the samples.  相似文献

Summary An algorithm for quantifying interelemental effects in X-ray fluorescence techniques is developed. By applying an addition process, the ratio between the mass absorption coefficients of the analyte and the unknown sample ( i * / s * ) is calculated to correct the fluorescence intensity of the element to be determined and linearize the I-c calibration plot. This coefficient can be calculated graphically and numerically. The method is applied to the determination of tin in lead alloys with good results over wide concentration ranges.Notation used Q Constant of proportionality in Eq. The synthesis, physicochemical characterization, and interaction with membrane model systems of a peptide derived from the PA22-2 region of laminin are described. Surface activity studies indicate that this peptide is able to spread at the air-water interface being the maximal spreading pressure 20 mN/m at subphase concentrations around 10 micro M. Besides, these peptide molecules are also able to form stable monolayers. Physicochemical studies concerning the interaction of this peptide with lipids, organized in mono and bilayers, were carried out using Langmuir balance experiments and polarization fluorescence techniques. The peptide penetrates better in monolayers of DPPC than in those of PC and forms condensed mixed monolayers with DPPC. Energies of mixing are small thus indicating that deviations from ideality were almost negligible. Interactions with bilayers were studied through microviscosity changes (DPH and TMA-DPH probes), membrane permeability alterations (CF, NBD-PE/dithionite), and fusion promotion (NBD-PE/Rh-PE, resonance energy transfer). Results indicate that this sequence interacts very softly with bilayers without promoting changes in their organization. These data as well as the lack of interaction with erythrocytes suggest that coating liposomes with this peptide through chemical amide bonds can render stable inmunoliposomes for further biological applications.  相似文献

The decomposition of highly insoluble minerals (fluorspar and cryolite) by fusion with molten alkali-metal hydroxides is studied. The introduction of additives such as aluminium compounds or sodium peroxide to obtain total liberation of fluoride from calcium fluoride samples, is tested. The fusion is done in a silver crucible with a Bunsen burner. The cooled melt is easily soluble, giving solutions suitable for spectrophotometric fluoride determination by the Zr(IV)-fluoride-Erichrome Cyanine R method.  相似文献

The physicochemical interactions between an active peptide sequence derived from laminin and phospholipids have been studied. The main aim is to determine the suitability of this peptide fragment to be linked to liposome's with the purpose to develop targeting devices. Results indicate that this peptide is able to insert in lipid monolayers and also to form monomolecular layers at an air/water interface. Besides, miscibility studies carried out through compression isotherms of mixed monolayers [dipalmitoyl phosphatidylcholine (DPPC)/peptide], indicate a strong interaction at 60-80% DPPC molar composition. Studies carried out with lipid bilayers indicate that the interaction is restricted to the external face of the vesicles. Moreover, the presence of this peptide in the incubation media promotes a low level of carboxyfluorescein (CF) leakage and no fusion of vesicles. These results indicate that the association of this sequence to vesicles do not produce damage of the bilayer and can be used as potential targeting vector.  相似文献

Summary This paper discusses the requirements and scope of application of the H-Point Standard Additions Method (HPSAM) for resolving binary mixtures. Various mixtures of dyes with extensively or completely overlapping spectra were assayed with accurate, precise results. The method thus developed was also applied to the spectrophotometric resolution of binary mixtures of phenol and o-cresol, the absorption bands of which lie in similar positions and feature similar absorptivities.  相似文献
