A post-column infusion system was developed in order to analyze suppression of electrospray ionization (ESI) tandem mass spectrometry response in the presence of endogenous plasma interferences. By enabling direct detection of these interfering components, this experimental system was used to analyze the ability of several common extraction procedures to remove endogenous plasma components that cause changes in the ESI response of model drug substances. Methyl-t-butyl ether (MTBE) liquid-liquid, Oasis and Empore solid-phase, and acetonitrile (ACN) protein precipitation sample preparation methods were tested using the post-column infusion system. In all cases, ACN protein precipitation samples showed the greatest amount of ESI response suppression while liquid-liquid extracts demonstrated the least. In addition, the three test compounds, phenacetin, caffeine, and a representative Merck compound, demonstrated that ESI response suppression is compound dependent. Suppression was greatest with caffeine, the most polar analyte, and the smallest for the Merck compound, the least polar analyte. Copyright 1999 John Wiley & Sons, Ltd.  相似文献

We report measurements of non-linear charge transport in epitaxial (La1−x Pr x )0.7Ca0.3MnO3 thin films fabricated on (100) oriented SrTiO3 single crystals by pulsed laser deposition. The end members of this series, namely Pr0.7Ca0.3MnO3 and La0.7Ca0.3MnO3 are canonical charge-ordered (CO) and ferromagnetic manganites, respectively. The onset of the CO state in Pr0.7Ca0.3MnO3 is manifested by a pronounced insulating behavior below ∼ 200 K. The CO state remains stable even when a large (∼ 2×105 V/cm) electric field is applied across the thin film samples. However, on substitution of Pr with La, a crossover from the highly resistive CO state to a state of metallic character is observed at relatively low electric fields. The current-voltage characteristics of the samples at low temperatures show hysteretic and history dependent effects. The electric field driven charge transport in the system is modelled on the basis of an inhomogeneous medium consisting of ferromagnetic metallic clusters dispersed in a CO background.  相似文献

We present laser photoacoustic spectra of ethyl hydroperoxide (EHP) for 3-6 quanta of O-H stretch. Spectra are consistent with ab initio spectral simulations that take into account the influence of torsional motion about the O-O bond on O-H stretch overtone excitation. Experimental and simulated spectra share two trends. First, spectral features due to torsional excitation, including hot bands, become more prominent with increasing O-H stretch excitation, as has been shown previously for similar molecules such as methyl hydroperoxide. Second, contributions from the two different EHP conformers become clearly distinguishable at higher O-H stretch excitation, mainly due to the role of torsional motion. Results are consistent with a higher energy separation (330 cm(-1)) between the lower energy anti and the higher energy gauche conformers than predicted by electronic structure calculations (137 cm(-1)). Calculated absorption intensities are consistently higher than experimental values by ~30% but within the experimental uncertainty. With each additional O-H stretch overtone, the dropoff in calculated integrated absorption intensities at room temperature becomes less extreme, with a 14-fold dropoff from 3ν(OH) to 4ν(OH) and an 8-fold decrease from 5ν(OH) to 6ν(OH).  相似文献

The M(CO)(4) fragment can be assigned to be isolobal with both CH(3)(+) and CH(2). In order to investigate this ambiguous isolobal assignment, we report calculations on compounds of the type M(CO)(4)L(n), where M is Fe (n = 0), Mn (n = -1), and Co (n = +1) and L is an η(2) ligand with a π bond, generally an alkene. The L's are varied in electron-withdrawing ability, and patterns in computed structural parameters are investigated. We report that the equatorial OC-M-CO angle is sensitive to the electron-withdrawing ability of the alkene just as the isolobal prediction suggests. Other structural parameters that vary monotonically with electron-withdrawing ability of the alkene are the "bending back" of the alkene, the metal-ligand bond distances, and carbon-oxygen bond distances. Changing the metal from neutral Fe to a negatively charged Mn or positively charged Co has the result of increasing and decreasing, respectively, the OC-M-CO angle. Several compounds of Ni(CO)(3)L are also investigated as a further example of the ability of the isolobal concept to yield chemically useful information.  相似文献

Two polymorphic forms of a novel pharmaceutical compound, ciprofloxacin-saccharinate (CIP-SAC), are analyzed using one dimensional (1D) and two dimensional (2D) (1)H nuclear magnetic resonance (NMR) at fast magic angle spinning (MAS). Additionally (15)N spectroscopy and (1)H-(13)C correlation experiments were performed to complement our conclusions. The 1D (1)H NMR spectra of CIP and complexes reveal valuable information about the ionic bonding between ciprofloxacin and saccharine. Additionally, these spectra allow us to perform a clear characterization of each solid form, giving the number of molecules per unit cell in one of the polymorphs. From 2D (1)H-(1)H spectra obtained through double quantum correlations we can arrive at important conclusions about the hydrogen bonding, conformation, and intra and inter-molecular interactions present in these compounds. Comparing and contrasting the (1)H-(1)H correlation data obtained for both polymorphic forms and taking into account the single crystal structure data existing for the solid form CIP-SAC (II) was possible to extract some conclusions on the polymorph CIP-SAC (I) where no single crystal information is available. (1)H MAS NMR is shown to be an important tool in the field of polymorphism and for the characterization of multicomponent pharmaceutical compounds.  相似文献

The like-sign dimuons copiously recorded in the NA 38 experiment both inp-U and O?U reactions at 200 GeV/nucleon are interpreted as resulting from decays of π andK mesons in comparable proportions. The ++/?? ratio is large (?1.7) and ascribed to theK + being more copiously produced than theK ?. Both the average transverse momentum and the ++/?? ratio are comparable forp-U and O?U reactions, and both increase only slightly with the transverse energyE T .  相似文献

Cs correctors have revolutionized transmission electron microscopy (TEM) in that they substantially improve point resolution and information limit. The object information is found sharply localized within 0.1 nm, and the intensity image can therefore be interpreted reliably on an atomic scale. However, for a conventional intensity image, the object exit wave can still not be detected completely in that the phase, and hence indispensable object information is missing. Therefore, for example, atomic electric-field distributions or magnetic domain structures cannot be accessed. Off-axis electron holography offers unique possibilities to recover completely the aberration-corrected object wave with uncorrected microscopes and hence we would not need a Cs-corrected microscope for improved lateral resolution. However, the performance of holography is affected by aberrations of the recording TEM in that the signal/noise properties ("phase detection limit") of the reconstructed wave are degraded. Therefore, we have realized off-axis electron holography with a Cs-corrected TEM. The phase detection limit improves by a factor of four. A further advantage is the possibility of fine-tuning the residual aberrations by a posteriori correction. Therefore, a combination of both methods, that is, Cs correction and off-axis electron holography, opens new perspectives for complete TEM analysis on an atomic scale.  相似文献

A set of new aluminium complexes of norfloxacin (NOR) and ciprofloxacin (CIP) that show an improvement in their pharmaceutical properties were studied using solution and solid-state nuclear magnetic resonance (NMR) and infrared (IR) spectroscopy. The complexes synthesized with two different methods were compared. One of these methods will allow formulation of the compounds at production scale. High-resolution (13)C spectra were obtained with the cross-polarization and magic angle spinning (CP-MAS) experiment. These spectra were assigned by comparing them with the solution data of the pure drug and by using a quaternary carbon edition technique. The carbon relaxation times in the rotating frame, T(1rhoC), were measured for all the complexes. A two-exponential decay evidences that the complexes are nonhomogeneous. The short T(1rhoC) values are in the range 320-1100 micros and the long values in the range 1.8-7 ms. (27)Al MAS NMR spectra revealed an octahedral coordination between the aluminium ion and oxygens of the pure drug, supporting a 3:1 ligand:metal stoichiometry in both CIP and NOR complexes. The stretching and deformation modes of carboxylic acid and carboxylate and keto groups were analyzed by IR. This technique shows that the same modes are present in the aluminium complexes obtained by the two methods and that the coordination of the fluoroquinolone to aluminium occurs through the 4-keto and 3-carboxylic groups.  相似文献

The reaction of Ru2(S2C3H6)(CO)6 (1) with 2 equiv of Et4NCN yielded (Et4N)2[Ru2(S2C3H6)(CN)2(CO)4], (Et4N)2[3], which was shown crystallographically to consist of a face-sharing bioctahedron with the cyanide ligands in the axial positions, trans to the Ru-Ru bond. Competition experiments showed that 1 underwent cyanation >100x more rapidly than the analogous Fe2(S2C3H6)(CO)6. Furthermore, Ru2(S2C3H6)(CO)6 underwent dicyanation faster than [Ru2(S2C3H6)(CN)(CO)5]-, implicating a highly electrophilic intermediate [Ru2(S2C3H6)(mu-CO)(CN)(CO)5]-. Ru2(S2C3H6)(CO)6 (1) is noticeably more basic than the diiron compound, as demonstrated by the generation of [Ru2(S2C3H6)(mu-H)(CO)6]+, [1H]+. In contrast to 1, the complex [1H]+ is unstable in MeCN solution and converts to [Ru2(S2C3H6)(mu-H)(CO)5(MeCN)]+. (Et4N)2[3] was shown to protonate with HOAc (pKa = 22.3, MeCN) and, slowly, with MeOH and H2O. Dicyanide [3]2- is stable toward excess acid, unlike the diiron complex; it slowly forms the coordination polymer [Ru2(S2C3H6)(mu-H)(CN)(CNH)(CO)4]n, which can be deprotonated with Et3N to regenerate [H3]-. Electrochemical experiments demonstrate that [3H]- catalyzes proton reduction at -1.8 V vs Ag/AgCl. In contrast to [3]2-, the CO ligands in [3H]- undergo displacement. For example, PMe3 and [3H]- react to produce [Ru2(S2C3H6)(mu-H)(CN)2(CO)3(PMe3)]-. Oxidation of (Et4N)2[3] with 1 equiv of Cp2Fe+ gave a mixture of [Ru2(S2C3H6)(mu-CO)(CN)3(CO)3]- and [Ru2(S2C3H6)(CN)(CO)5]-, via a proposed [Ru2]2(mu-CN) intermediate. Overall, the ruthenium analogues of the diiron dithiolates exhibit reactivity highly reminiscent of the diiron species, but the products are more robust and the catalytic properties appear to be less promising.  相似文献
